DNA methylation-based age estimators (DNAm ageing clocks) are currently one of the most promising biomarkers for predicting biological age. However, the relationships between cardiorespiratory fitness (CRF), measured directly by expiratory gas analysis, and DNAm ageing clocks are largely unknown. We investigated the relationships between CRF and the age-adjusted value from the residuals of the regression of DNAm ageing clock to chronological age (DNAmAgeAcceleration: DNAmAgeAccel) and attempted to determine the relative contribution of CRF to DNAmAgeAccel in the presence of other lifestyle factors. DNA samples from 144 Japanese men aged 65–72 years were used to appraise first- (i.e., DNAmHorvath and DNAmHannum) and second- (i.e., DNAmPhenoAge, DNAmGrimAge, and DNAmFitAge) generation DNAm ageing clocks. Various surveys and measurements were conducted, including physical fitness, body composition, blood biochemical parameters, nutrient intake, smoking, alcohol consumption, disease status, sleep status, and chronotype. Both oxygen uptake at ventilatory threshold (VO2/kg at VT) and peak oxygen uptake (VO2/kg at Peak) showed a significant negative correlation with GrimAgeAccel, even after adjustments for chronological age and smoking and drinking status. Notably, VO2/kg at VT and VO2/kg at Peak above the reference value were also associated with delayed GrimAgeAccel. Multiple regression analysis showed that calf circumference, serum triglyceride, carbohydrate intake, and smoking status, rather than CRF, contributed more to GrimAgeAccel and FitAgeAccel. In conclusion, although the contribution of CRF to GrimAgeAccel and FitAgeAccel is relatively low compared to lifestyle-related factors such as smoking, the results suggest that the maintenance of CRF is associated with delayed biological ageing in older men.  相似文献

Benzyladenine (BA) stimulated 5-aminolevulinic acid (ALA) accumulation in the presence of levulinic acid during illumination with 43 μmol m−2 s−1 light in excised etiolated cotyledons of cucumber ( Cucumis sativus L. cv. Aonagajibai). A short dark-pretreatment (6 h) with BA eliminated the lag phase of ALA accumulation. The rate of ALA accumulation during the steady-state phase in cotyledons pretreated with BA for a long period (14 h) was considerably accelerated compared to that in cotyledons pretreated with BA for 6 h. The rate of ALA accumulation during the lag phase was saturated at a very low light fluence (<1.4 μmol m−2 s−1) in both BA-pretreated and water-control cotyledons. The steady-state rate of ALA accumulation increased with increasing light fluence up to 43 μmol m−2 s−1 (parallel to that of Chl formation) in water-control cotyledons. In contrast, in cotyledons pretreated with BA for either 6 or 14 h, the steady-state rate reached a plateau at a very low light fluence. Based on the above results together with our finding that there are two components of Chl formation (M. Dei, 1984. Physiol. Plant. 62: 521–526) possible intermediate steps of Chl biosynthesis pathway affected by BA and light intensity are discussed.  相似文献

Although the sex-determining gene DMY has been identified on the Y chromosome in the medaka (Oryzias latipes), this gene is absent in most Oryzias species, suggesting that closely related species have different sex-determining genes. Here, we investigated the sex-determination mechanism in O. dancena, which does not possess the DMY gene. Since heteromorphic sex chromosomes have not been reported in this species, a progeny test of sex-reversed individuals produced by hormone treatment was performed. Sex-reversed males yielded all-female progeny, indicating that O. dancena has an XX/XY sex-determination system. To uncover the cryptic sex chromosomes, sex-linked DNA markers were screened using expressed sequence tags (ESTs) established in O. latipes. Linkage analysis of isolated sex-linked ESTs showed a conserved synteny between the sex chromosomes in O. dancena and an autosome in O. latipes. Fluorescence in situ hybridization (FISH) analysis of these markers confirmed that sex chromosomes of these species are not homologous. These findings strongly suggest an independent origin of sex chromosomes in O. dancena and O. latipes. Further analysis of the sex-determining region in O. dancena should provide crucial insights into the evolution of sex-determination mechanisms in vertebrates.  相似文献

Transport of cytoplasmically synthesized proteins into chloroplasts uses an import machinery present in the envelope membranes. To identify the components of this machinery and to begin to examine how these components interact during transport, chemical cross-linking was performed on intact chloroplasts containing precursor proteins trapped at a particular stage of transport by ATP limitation. Large crosslinked complexes were observed using three different reversible homobifunctional cross-linkers. Three outer envelope membrane proteins (OEP86, OEP75, and OEP34) and one inner envelope membrane protein (IEP110), previously reported to be involved in protein import, were identified as components of these complexes. In addition to these membrane proteins, a stromal member of the hsp100 family, ClpC, was also present in the complexes. We propose that ClpC functions as a molecular chaperone, cooperating with other components to accomplish the transport of precursor proteins into chloroplasts. We also propose that each envelope membrane contains distinct translocation complexes and that a portion of these interact to form contact sites even in the absence of precursor proteins.  相似文献

Bacteria, including cyanobacteria, as well as some fungi, are known to deposit calcium carbonate (CaCO3) extracellularly in calcium-containing artificial medium. Despite extensive investigation, the mechanisms involved in extracellular formation of CaCO3 by bacteria have remained unclear. The ability of synthetic amines to remove carbon dioxide (CO2) from natural gas led us to examine the role of biogenic polyamines in CaCO3 deposition by bacteria. Here, we demonstrated that biogenic polyamines such as putrescine, spermidine, and spermine were able to react with atmospheric CO2 and the resultant carbamate anion was characterized by using nuclear magnetic resonance (NMR) analysis. Biogenic polyamines accelerated the formation of CaCO3, and we artificially synthesized the dumbbell-shaped calcites, which had the same form as observed with bacterial CaCO3 precipitates, under nonbacterial conditions by using polyamines. The reaction rate of calcification increased with temperature with an optimum of around 40 °C. Our observation suggests a novel scheme for CO2 dissipation that could be a potential tool in reducing atmospheric CO2 levels and, therefore, global warming.  相似文献

Aims To describe the biodiversity patterns of plants along an altitudinal gradient on the Qinghai-Tibetan Plateau and to clarify the bias in plant specimen records at high altitude.Methods We conducted a large-scale investigation of vegetation at a wide range of altitudes, focusing on a high-altitudinal range (3?200–5?200 m) at different locations on the Qinghai-Tibetan Plateau. We then compared the altitudinal distribution of plant species obtained from our field investigation with that in plant specimen records from published sources and an online database.Important findings Our data provide evidence that altitude plays a large role in regulating species composition on the Qinghai-Tibetan Plateau. We could not, however, detect a clear relationship between altitude and species richness, although a weak monotonically increasing trend of richness was detected with increasing altitude. According to specimen records, most species have been sampled at a wide range of altitudes, and the average range of 145 species is>2?000 m. Despite this wide range, more than half of the species we observed were at higher altitudes than the specimen records indicate. High-altitude areas have probably been so poorly sampled that only a small fraction of the resident species has been recorded. This study clearly shows the regional bias of specimen records in the Qinghai-Tibetan Plateau.  相似文献

The molecular-weight dependence of the rms radius of gyration of poly(γ-benzyl L -glutamate) (PBLG) in helicogenic solvents shows negative and positive deviations from expectations for an intact and rigid α-helix in the higher and lower molecular-weight ranges, respectively. In order to study the reason for both deviations, we compare the extant experimental data of with those computed for wormlike chain, freely jointed rod, and a rigid rod having random-coil portions at both ends. The computation for the freely jointed rod and the rigid rod having frayed ends is carried out by a simulation method of Muroga. From the Zimm and Bragg theory and the above comparisons, it is concluded that both deviations can be self-consistently explained if PBLG in helicogenic solvents has an essentially intact α-helical structure with some flexibility arising from random fluctuations in hydrogen bond length. This flexibility explains the negative deviations in the high molecular weight region. We determined partial sequences of the gyrA and parC genes of Citrobacter freundii type strain, and then examined 38 C. freundii clinical strains isolated from patients with urinary tract infections for the association of alterations in GyrA and ParC with susceptibility to fluoroquinolones. Our results suggest that in C. freundii DNA gyrase may be a primary target of quinolones, that an amino acid change at Thr-83 or Asp-87 in GyrA is sufficient to decrease susceptibility to fluoroquinolones, and that accumulation of changes in GyrA with the simultaneous presence of an alteration at Ser-80 or Glu-84 in ParC may be associated with the development of high-level fluoroquinolone resistance in C. freundii clinical isolates.  相似文献

植物叶片对环境变化十分敏感,能反映植物适应环境所形成的生存策略。为揭示高寒植物叶片性状对海拔高度变化的响应,对位于青藏高原东北部的冷龙岭3400—4200 m之间5个不同海拔高度的矮火绒草(Leontopodium nanum)叶片进行取样,采用常规石蜡制片技术和显微观察方法测定叶片外部形态、表皮气孔特征和解剖结构,探讨其叶片性状随海拔的变化,结果表明:(1)随海拔高度升高,叶面积呈减小的趋势,而比叶重和叶干物质含量增加;(2)叶片下表皮气孔密度随海拔升高呈先增加后下降的趋势,且气孔密度、气孔器面积、长度、宽度和潜在气孔导度指数等气孔特征之间存在显著相关性;(3)叶厚、栅栏组织和海绵组织厚度随海拔升高呈显著增厚的趋势;(4)叶片解剖结构可塑性和相关性分析显示,上、下角质层厚度的可塑性指数最大,而部分解剖结构指标间存在极显著的相关性。研究表明,矮火绒草为适应沿海拔上升温度降低的环境,主要采取叶片变小、变厚的对策,使植物趋于保温、保水和抗机械损伤的方向发展,并将资源最大化地投入到自身生长发育中。  相似文献
