Fimbrin, previously recognized as a major structural protein of the microfilament core bundles of intestinal epithelial cell microvilli, has been purified to homogeneity and characterized. It is a nearly globular monomeric protein of apparent molecular weight 68,000 and has a single calcium binding site (Kd = 9 microM), for which magnesium ions compete. Fimbrin binds to F-actin and this interaction is characterized in detail. Under our optimal binding of conditions, fimbrin induces tightly packed F-actin bundles, similar to the bundles induced by villin, another microvillus structural protein. The formation of mixed fimbrin-villin-actin bundles provides a further step toward the full in vitro reconstitution of microvillus core filaments from its purified individual components. The reconstituted fimbrin-villin-actin bundles do not display the side arms characteristic of isolated microvillus cores. These results are discussed in terms of our current understanding of the organization of the microvillus core filaments and indicate that this structure contains two bundling proteins, villin and fimbrin. The results complement previous studies and now provide a minimal biochemical characterization of all four major actin-associated structural proteins so far identified in core filaments. Three of these (villin, fimbrin, and calmodulin) are calcium-binding proteins, emphasizing the concept of calcium control over submembranous microfilament organization.  相似文献

The relationship between dissipation of the flash-induced membranepotential across the thylakoid membrane and the high energystate was studied in Zea mays leaves. The dark decay of theflash-induced 515-nm absorbance change was accelerated by shortpreillumination of the leaf. No acceleration of the decay bypreillumination was observed when leaves were incubated in argonor CO2 gas or treated with DCMU. These effects of preilluminationand incubation were reversible. The delayed fluorescence from chlorophyll a was reversibly decreasedby incubating leaves in argon or CO2 gas, though the modes ofdepression were somewhat different from each other. In leavesincubated in argon or CO2 gas, the phase of slow decrease ofthe intensity of prompt fluorescence during illumination reversiblydisappeared. The results suggested that the dissipation of membrane potentialgenerated by a flash was accelerated after the energizationof chloroplasts in leaves, probably by increased H permeabilityof the thylakoid membrane. O2 was important in maintaining (indarkness) and forming (under illumination) the high energy statein chloroplasts in intact leaves. (Received October 1, 1980; Accepted December 15, 1980)  相似文献

Pure phloem sap from rice plant was collected by a method similarto the aphid technique using leafhoppers and planthoppers instead.A Yttrium-Aluminium Garnet laser was used to sever the insectstylets. Sap exuded from the plant through the stylet for upto 3 hr at a rate of 0.2 µl/hr. The electric birefringence and circular dichroism spectra of poly(l-ornithine hydrobromide) have been measured in ethanol/water, 2-propanol/water and tertiary butyl alcohol/water mixtures of various compositions. This charged polypeptide underwent a transition from the coil conformation to the helical conformation at high alcohol content in every case tested. Anomalous birefringence signals, indicative of a field-induced helix-to-coil transition. were observed at high electric fields only in the case of ethanol/water mixtures. The reversing-pulse electric birefringence of this polypeptide has been studied in ethanol/water mixtures and in neutral aqueous solution. Upon rapid reversal of the pulse field, no transient could be observed. This confirms that the electric-field orientation of poly(l-ornithine hydrobromide) results predominantly from the contribution of the counterion-induced dipole moment, regardless of its molecular conformations. It is very probable that the backbone permanent dipole moment of the helical conformation is largely suppressed by the counterion-induced dipole moment in the ionized form.  相似文献

The arginine and lysine residues of calf thymus histone H1 were modified with large molar excesses of 2,3-butanedione and O-methylisourea, respectively. Kinetic study of the modification reaction of the arginine residue revealed that the reaction is divided into the two pseudo-first-order processes. About a third (1 Arg) of the total arginine residues of the H1 molecule was rapidly modified without causing any detectable structural change of the molecule, and the slow modification of the remaining arginine residues (2 Arg) led to a loss of the folded structure of H1. In the case of lysine residue modification, 93% (56 Lys) of the total lysine residues of the H1 was modified with the same rate constant, while 7% (4 Lys) of lysine residue remained unmodified. When the reaction was performed in the presence of 6M guanidine-HCl, all of lysine residues were modified. It is concluded that the 2 arginine and 4 lysine residues resistant to modification are buried in interior regions of the H1 molecule and play an important role in the formation of the H1 globular structure, while the other 1 arginine and 56 lysine residues are exposed to solvent.  相似文献

Nonphosphorylating electron transport particles (ETP) prepared from beef heart mitochondrion were immobilized in agar gel. The immobilized ETP showed an oxidase activity to both NADH and succinate. The immobilized ETP was reusable. An electrochemical device for the determination of either NADH or succinate was assembled consisting of the membrane-bound ETP and an oxygen probe. The response to succinate was specifically inhibited by the addition of malonate.  相似文献

Induction and repair of DNA breaks following irradiation with NIRS cyclotron neutrons were studied in cultured mammalian cells (L5178Y) in comparison to those following gamma-rays. The yield of the total single-strand breaks, 3'OH terminals and sites susceptible to S1 endonuclease following fast neutrons was found to be approximately 50 per cent of that following gamma-irradiation. On the other hand, the yield of double-strand breaks was slightly higher after fast neutrons than after gamma-rays. The percentage of the total single-strand breaks remaining unrejoined at 3 hours after post-irradiation incubation was found to be distinctly higher after the fast neutrons than after gamma-rays. The neutron-induced damage appears to carry a higher proportion of alkali-labile lesions compared to gamma-rays. It was concluded that the increase in the yield of double-strand breaks and of unrejoinable breaks is responsible for a high r.b.e. of the cyclotron neutrons.  相似文献

The course of resin canals in stem cortex and the continuity between resin canals in leaves and those in stem cortex were investigated. The present paper is the first of three parts of the investigation. In this paper, fundamental features of resin canals and actual resin canal patterns in the Taxaceae, Cephalotaxaceae and Podocarpaceae are reported. From the observation of serial transections of shoots, composite diagrams and three-dimensional models of resin canal patterns are drawn. Central canals, if present, run vertically in stem cortex and sometimes divide, end blindly or unite each other. The distance between two adjacent central canals fluctuates rhythmically in connection with the vascular supply from the stem to leaves. The resin canal patterns of the families are classified into four types. Those ofTaxus, Nothotaxus and three species ofDacrydium belong to the Taxus type, those ofTorreya andCephalotaxus to the Torreya type, those ofDacrydium elatum, Podocarpus alpinus, P. elatus, P. elongatus andP. neriifolius to the Dacrydium type, those ofP. macrophyllus, P. nagi andP. koordersii to the Podocarpus type.  相似文献

Summary The degradation of poly(l-glutamic acid) and poly(l-lysine) by X-rays and ultraviolet radiation has been studied at variouspH values, including the helix-coil transition region. It was found that X-rays cause maximum degradation in the transition region for both polypeptides, indicating that the efficiency of degradation is affected not only by the secondary structure but also by some other factors related to the transition. On the contrary, the degradation by monochromatic ultraviolet 2537 radiation showed a strong dependence on helix content of PLGA, suggesting that degradation occurs more effectively in the helical form than in the coil form. Theoretical calculations were made to approximate thepH dependence of degradation, assuming different cross sections for the peptide bonds in helix, coil, and junctions between helical and coil regions.  相似文献

A hemoglobin polymorphism involving variant -chains was demonstrated in the Celebes black ape, Macaca nigra. Fingerprinting and amino acid analysis of the tryptic peptides from the two chain types have shown that they differ by a single amino acid substitution, between lysine and aspartic acid, which requires a two-nucleotide change in the corresponding codon. Another substitution in the same codon is found as a species difference between the black ape -chain and that of other macaques.  相似文献
