The interaction and coupling between photosynthetic processeswere studied in ethanol-treated chloroplasts (in the absenceof ethanol) or in the presence of ethanol. Light-induced H+uptake and photophosphorylation were suppressed and electrontransfer was enhanced by ethanol treatment or in the presenceof ethanol (10–25%). Dark recoveries of the H+ uptakeand the 515-nm absorbance change were accelerated by ethanol.The half-effective concentrations of ethanol for these processeswere higher in the ethanol-treated chloroplasts than when ethanolwas present in the reaction mixture. The maximum rate of electrontransfer in the ethanol-treated chloroplasts, which was at thesame level as that of the control with an uncoupler, was notaffected by uncouplers. The marked acceleration of recoveryof the 515-nm absorbance change by SCN- or valinomycin plusK+ in the untreated chloroplasts was much smaller in the ethanol-treatedchloroplasts or in the presence of ethanol. The ethanol-treatedpreparation had the same characteristics as those of the control,in chlorophyll fluorescence, light-intensity dependence of electrontransfer (compared with the control with an uncoupler), andsensitivity to sucrose osmolarity except for a slight increaseof the packed volume. Under the present conditions, the ethanoltreatment mainly induced an increase of permeability of thylakoidmembranes to ions. In chloroplasts treated with ethanol at various concentrations,the relationships of the uncoupler-stimulated part of electrontransfer with the dark half-recovery time of H+ uptake and withphotophosphorylation were linear. Logarithms of the photophosphorylationand the extent of H+ uptake had a linear relationship with aslope of about 3. This slope may indicate the stoichiometryof H+/ATP. Membrane surface potential on the periplasmic side of the photosynthetic membrane was estimated in cells, spheroplasts and chromatophores of Rhodopseudomonas sphaeroides. When the membrane potential (potential difference between bulk aqueous phases) was kept constant in the presence of carbonylcyanide m-chlorophenylhydrazone, addition of salt to a suspension of cells or spheroplasts induced a red shift in the carotenoid absorption spectrum which indicated a change in the intramembrane electrical field. The spectral shift is explained by a rise in electrical potential at the outside surface of the photosynthetic membrane due to a decrease in extent of the negative surface potential.The spectral shift occurred in the direction opposite to that in chromatophores, indicating that the sidedness of the membrane of cells or spheroplasts is opposite to that of chromatophores. The dependences of the extent of the potential change on concentration and valence of cations of salts agreed with the Gouy-Chapman relationship on the electrical diffuse double layer. The charge density on the periplasmic surface of the photosynthetic membrane was estimated to be ?2.9 · 10?3 elementary charge per Å2, while that on the cytoplasmic side surface was calculated as ?1.9 · 10?3 elementary charge per Å2 (Matsuura, K., Masamoto, K., Itoh, S. and Nishimura, M. (1979) Biochim. Biophys. Acta 547, 91–102). Surface potential on the periplasmic side of the photosynthetic membrane was estimated to be about ?50 mV at pH 7.8 in the presence of 0.1 M monovalent salt.  相似文献

The effect of salt addition on the rate of reduction of P-700 oxidized by flash illumination was analyzed. In broken chloroplasts, the rate of P-700 reduction was accelerated by salts of mono-, di- and trivalent cations, with the increasing effectiveness in this order, in the presence of various artificial electron donors or acceptors. The rate was not dependent on the concentration and the valence of anions. On the other hand, in Photosystem I-enriched subchloroplast particles, added KCl did not induce the acceleration of direct reduction of P-700 by reduced DCIP.At low KCl concentrations (below 10 mM), the rate of P-700 reduction was also accelerated by added KCl in sonicated chloroplasts to which purified plastocyanin was added. The curves of dependence of the reduction rate on plastocyanin concentration were not of the Michaelis-Menten type, but sigmoidal. The maximal of P-700 reduction was higher at higher salt concentrations and the half-maximal plastocyanin concentration for P-700 reduction became lower with increasing NaCl concentrations.In broken chloroplasts treated with 50 mM glutaraldehyde, the rate of P-700 reduction was not accelerated by added KCl.The Debye-Hückel theory and the Gouy-Chapman theory were applied to our data to analyze the electrostatic interaction between electron tranfer components on thylakoid membranes. It is suggested that the major factor determining the rate of P-700 reduction is the donation of electrons from plastocyanin to P-700. Most of the observed effect is probably due to the increase in the local concentration or accessibility of plastocyanin to the site of P-700 reduction which is expected when the negative surface potential rises when salt is added.  相似文献

The changes in carotenoid absorbance induced by illumination or by a diffusion potential were larger in chromatophores from cells cultured under low light intensity than those in chromatophores from high-light culture in a photosynthetic bacterium, Rhodopseudomonas sphaeroides. The carotenoid molecules which are associated with the pigment-protein complex (with the infrared bacteriochlorophyll peaks at 800 and 850 nm) (complex II) probably respond to the electrical field changes in the chromatophore membrane.  相似文献

Pittendrigh first found that the circadian rhythm of locomotor activity in nocturnal rodents split into two components. Hoffman then reported that the splitting phenomenon was even more reproducible in the small diurnal primate Tupaia. These “splitting” experiments and many other experiments suggest that two coupled oscillators may constitute the circadian pacemaker system. Pittendrigh proposed a phenomenological two-oscillator model. Daan and Berde developed a quantitative model assuming that the interaction between the two constituent oscillators is by instantaneous resets. Their model system can simulate several qualitative features in the experimental data. As the assumption of instantaneous resets seems to be unnatural, we study two limit cycle oscillators, which are coupled continuously to each other, as a model of the circadian pacemaker. We assume the following points, (i) One oscillator in a resting state does not affect another oscillator, (ii) Two oscillators are identical, (iii) The coupling is symmetrical. By the theory of Hopf bifurcation it is found that the general two-oscillator system has two stable periodic solutions. One is the in-phase solution where the two constituent oscillators oscillate in phase synchrony. Another is the anti-phase solution where the two oscillators oscillate 180 ° out of phase. The former corresponds to a single pattern of locomotor activity and the latter corresponds to a splitting pattern. Furthermore, we study specific two-neural oscillators, which are linearly coupled to each other. By the method of secondary bifurcation we find that the model shows simultaneous stability of the two alternative phase relationships and the hysteresis phenomena found in Tupaia. A natural period of the uncoupled constituent oscillator is longer than that of the in-phase solution but it is shorter than that of the anti-phase solution. This is in agreement with the data of Tupaia.  相似文献

The aim of this study was to assess whether a particular value of noninvasive salivary ultra‐weak chemiluminescence (UCL) could be used as a biomarker of psychological stress. Our study covered two groups. Group 1 comprised six healthy volunteers who stayed in a hospital for one night and group 2 comprised 15 patients with lung cancer and 24 patients with respiratory diseases other than lung cancer who were in hospital for an extended stay. First, we evaluated the UCL of saliva from six healthy volunteers before and after one night in hospital. Immunoglobulin A (IgA) concentrations were also measured. The integrated intensity value of UCL was correlated with the IgA concentration (correlation coefficient 0.90). Second, in the case of a long hospital stay, we found that the maximum salivary UCL intensities were higher in patients with lung cancer than in those with respiratory diseases other than lung cancer or in 28 healthy controls. Many hematological and immunological parameters remain unclear in the study of the guinea pig. In this study, we established the mean values of blood counts, the percentage of leukocyte fractions and lymphocyte subsets in blood and various lymphoid tissues of the guinea pig with a flow cytometric procedure using MIL4/SSC. The mean counts of WBC and RBC in the blood were lower, and MCV and MCH were higher than those of other rodents, resembling those of humans. Furthermore, the mean percentages of blood lymphocytes were smaller and that of granulocyte was larger than those of other rodents, resembling those of humans. We further established a flow cytometric procedure for lymphocyte subsets and clarified the mean percentages of T- and B-cells, CD4(+)-, CD8(+)- and MHC Class II(+)- T-cells, and CD4(-)CD8 (-) T-cells. The latter were morphologically larger in cell size and cytoplasm than CD4(+)- plus CD8(+) T-cells, and this subset had a significantly higher percentage in newborn animals. Furthermore, the appearance of the MHC Class II(+) T-cell subset was suggested to be a marker of hyper-activation of T-cells in BCG-immunized animals. Thus, both the novel flow cytometric procedure for leukocyte fractions and lymphocyte subsets, and the established normal values will be useful tools in studying guinea pigs as models of various diseases and biological phenomena.  相似文献

Inostamycin, which was recently isolated from Streptomyces sp. MH816-AF15 as an inhibitor of cytidine 5'-diphosphate 1,2-diacyl-sn-glycerol (CDP-DG): inositol transferase, caused a G1-phase accumulation in the cell cycle of small cell lung carcinomas. To investigate whether the cytostatic effect of inostamycin is restricted to lung carcinoma cell lines or applicable to other type of cells, we tested five oral squamous cell carcinoma (SCC) cell lines. Cell growth was suppressed in 62.5--125 ng/ml inostamycin in the culture medium in all oral cancer cell lines tested, with non-viable cells being <1%, indicating inostamycin is cytostatic on SCC cell lines. Decrease in cyclin D1 mRNA and protein expression due to the inostamycin treatment was accompanied by suppression of phosphorylated retinoblastoma susceptibility gene product (pRB-P) levels. Moreover, flow cytometric analysis showed that inostamycin induced an increase in G1/G0 cells (1.2--3.2 fold) over 24 h. These results suggest that inostamycin is a useful agent for tumour dormant cytostatic therapy for oral SCC.  相似文献
