Ischemia results in increased phosphorylation of NMDA receptors. To investigate the possible role of lipid rafts in this increase, lipid rafts and post-synaptic densities (PSDs) were isolated by the extraction of rat brain synaptosomes with Triton X-100 followed by sucrose density gradient centrifugation. Lipid rafts accounted for the majority of PSD-95, whereas SAP102 was predominantly located in PSDs. Between 50 and 60% of NMDA receptors were associated with lipid rafts. Greater than 85-90% of Src and Fyn were present in lipid rafts, whereas Pyk2 was mainly associated with PSDs. Lipid rafts and PSDs were isolated from animals subjected to 15 min of global ischemia followed by 6 h of recovery. Ischemia did not affect the yield, density, flotillin-1 or cholesterol content of lipid rafts. Following ischemia, the phosphorylation of NR1 by protein kinase C and tyrosine phosphorylation of NR2A and NR2B was increased in both lipid rafts and PSDs, with a greater increase in tyrosine phosphorylation occurring in the raft fraction. Following ischemia, NR1, NR2A and NR2B levels were elevated in PSDs and reduced in lipid rafts. The findings are consistent with a model involving close interaction between lipid rafts and PSDs and a role for lipid rafts in ischemia-induced signaling pathways.  相似文献

The introduction of novel molecular tools in research and clinical medicine has created a need for more refined information management systems. This article describes the design and implementation of such a new information platform: the Molecular Imaging Portal (MIPortal). The platform was created to organize, archive, and rapidly retrieve large datasets using Web-based browsers as access points. The system has been implemented in a heterogeneous, academic research environment serving Macintosh, Unix, and Microsoft Windows clients and has been shown to be extraordinarily robust and versatile. In addition, it has served as a useful tool for clinical trials and collaborative multi-institutional small-animal imaging research.  相似文献

Sex allocation theory predicts that: (1) resources allocated to androecium should decrease with an increase in selfing, (2) a decrease in androecium biomass should be accompanied by an increase in the biomass of pistils, and (3) a decrease in androecium biomass should be coupled with a decrease in flower size, specifically corolla biomass. Another predicted change in reproductive traits associated with variation in selfing concerns seed to ovule ratios, but does not directly stem from sex allocation theory. It has been postulated that seed to ovule ratios should be positively correlated with the amount of selfing. These predictions were tested for six accessions of pigeonpea,Cajanus cajan L., that differed in selfing rates. The results were remarkably in accordance with the predictions. We conclude that sex allocation theory provides a powerful tool to understand the evolution of many reproductive traits in plants.  相似文献

The Role of Inhibition in an Associative Memory Model of the Olfactory Bulb   总被引:1,自引:0,他引:1  
The external plexiform layer is where theinteractions between the mitral (excitatory) and granule (inhibitory)cells of the olfactory bulb (OB) take place. Two outstanding features ofthese interactions are that they aredendrodendritic and that there seem to be nonebetween excitatory cells. The latter are usually credited with the role of forming Hebbian cell assemblies.Hence, it would seem that this structure lacks the necessaryingredients for an associative memory system.In this article we show that in spite of these two properties thissystem can serve as an associative memory. Our model incorporates theessential anatomical characteristics of the OB. The memories in oursystem, defined by Hebbian mitral assemblies, are activated viathe interactions with the inhibitory granule cells. The nonlinearityis introduced in our model via a sigmoid function that describesneurotransmitter release in reciprocal dendrodendritic synapses. Thecapacity (maximal number of odors that can be memorized) depends onthe sparseness of coding that is being used. For very low memoryactivities, the capacity grows as a fractional power of the number ofneurons. We validate the theoretical results by numericalsimulations. An interesting result of our model is that its capacityincreases as a function of the ratio of inhibitory to excitatorypopulations. This may provide an explanation for the dominance ofinhibitory cells in the olfactory bulb.  相似文献

While many species of animals can solve food-baited problems, most studies are conducted in captivity, which may not reflect the natural behavioural and cognitive abilities of wild animals. As few studies have explored problem solving of Australian animals generally, we investigated the problem solving abilities of native Australian species in natural rainforest in the Wet Tropics of Queensland. We baited multiple types of puzzles (matchbox task, cylinder task, and tile and lever tasks on a Trixie Dog Activity Board) with different food types (seeds, fruit, sardines) and placed the puzzles in front of trail cameras. We noted the species captured on camera, whether or not individuals interacted with the puzzles, the number of interactions with puzzles, and whether or not different animals solved them. We found that seven species from multiple taxa (mammals, birds, reptiles) could solve food-baited problems in the wild, providing the first evidence of problem solving in these native species. As problem solving may help animals cope with anthropogenic threats, these results provide some insights into which Wet Tropics species may potentially be more vulnerable and which ones might be better at coping with changing conditions.  相似文献

The selective transport of molecules across membrane pores is an essential biological process that occurs in all living organisms. Examples include the chaperone of proteins and mRNA in and out the cell nucleus mediated by the nuclear pore complex and the transport of specific molecules across a biological membrane mediated by active and passive transporters. Here, I describe an artificial DNA transporter that is formed by incorporating a DNA carrier element into a biological nanopore imbedded in a lipid bilayer. This device is able to transport specific DNA strands across a biological membrane mediated by a simple reaction mechanism based on DNA strand displacement. Similar to biological secondary active transporters, this system uses an electrochemical potential difference to pump a specific substrate across a biological membrane at a constant transmembrane potential. Our DNA actuator might be used to separate or concentrate nucleic acids, or to vehicle genetic information across the biological membranes.  相似文献
