The axial interactions of Cu(2+) in type 1 copper proteins control the physical characteristics of the proteins. We tuned the geometries of a de novo designed blue copper protein with a four-helical bundle structure. The designed protein axially bound various ligands, such as chloride, phosphate, sulfate, acetate, azide, and imidazole, to Cu(2+), exhibiting a blue or green color. The UV-vis spectral bands were observed at approximately 600?nm and approximately 450?nm, with the A (~450)/A (~600) ratios between 0.14 and 1.58. The stronger axial interaction shifted the geometry of the type 1 copper site from trigonal planar geometry (blue copper) toward a tetrahedral-like geometry (green copper). Resonance Raman spectral analyses showed that the phosphate-bound type had the highest-strength Cu-S bond, similar to that of plastocyanin. The chloride-bound type exhibited features similar to those of stellacyanin and nitrite reductase, and the imidazole-bound type exhibited features similar to those of azurin M121E mutant. 相似文献

Competitive effects of vines on their tree hosts are well documented, but the mechanisms involved in these interactions are poorly understood. The objectives of this study were to measure the effects of below- and/or aboveground competition from the vines Lonicera japonica and Parthenocissus quinquefolia on availability of light, water, and nitrogen to the host tree Liquidambar styraciflua, and to examine the relationship between resource availability and tree growth. Light penetration through tree canopies, pre-dawn leaf water potential, and leaf nitrogen concentration were used as predictors of light, water, and nitrogen availability to the tree, respectively. Vine presence significantly reduced light penetration through the tree canopies, but this reduction was not clearly related to the growth responses of trees. Vines did not reduce the pre-dawn leaf water potential of competing trees, which was consistently above -0.5 MPa for the duration of the study. Leaf nitrogen concentration of trees, on the other hand, was significantly reduced by belowground competition with L. japonica. The positive correlation between the annual average leaf nitrogen concentration and tree diameter growth suggested that competition for nitrogen mediated the effects of belowground competition of vines on tree growth. 相似文献

The photosynthetic apparatus of some plant species appears to be well-protected from direct damage from UV-B radiation. Leaf optical properties of these species apparently minimizes exposure of sensitive targets to UV-B radiation. However, damage by UV-B radiation to Photosystem II and Rubisco has also been reported. Secondary effects of this damage may include reductions in photosynthetic capacity, RuBP regeneration and quantum yield. Furthermore, UV-B radiation may decrease the penetration of PAR, reduce photosynthetic and accessory pigments, impair stomatal function and alter canopy morphology, and thus indirectly retard photosynthetic carbon assimilation. Subsequently, UV-B radiation may limit productivity in many plant species. In addition to variability in sensitivity to UV-B radiation, the effects of UV-B radiation are further confounded by other environmental factors such as CO2, temperature, light and water or nutrient availability. Therefore, we need a better understanding of the mechanisms of tolerance to UV-B radiation and of the interaction between UV-B and other environmental factors in order to adequately assess the probable consequences of a change in solar radiation.Abbreviations Amax

A chimeric protein consisting of a cell-adhesive peptide derived from a neural cell adhesion molecule and a collagen-binding domain was synthesized using recombinant DNA technology. Here, we demonstrate that the chimeric protein binds to type I collagen and promotes the adhesion and neurite extension of hippocampus neurons. These results suggest that the chimeric protein has potential to provide microenvironments for neurons to adhere and survive in collagen-based matrices for use in cell-based therapies for central nervous disorders. 相似文献

Quick and facile transient RNA interference (RNAi) is one of the most valuable plant biotechnologies for analysing plant gene functions. To establish a novel double‐strand RNA (dsRNA) delivery system for plants, we developed an ionic complex of synthetic dsRNA with a carrier peptide in which a cell‐penetrating peptide is fused with a polycation sequence as a gene carrier. The dsRNA–peptide complex is 100–300 nm in diameter and positively charged. Infiltration of the complex into intact leaf cells of Arabidopsis thaliana successfully induced rapid and efficient down‐regulation of exogenous and endogenous genes such as yellow fluorescent protein and chalcone synthase. ObjectiveTo explore the baseline predictors of clinical effectiveness after tocilizumab or infliximab treatment in biologic-naïve rheumatoid arthritis patients.MethodsConsecutive biologic-naïve patients with rheumatoid arthritis initiating infliximab (n = 57) or tocilizumab (n = 70) treatment were included in our prospective cohort study. Our cohort started in February 2010, and the patients observed for at least 1 year as of April 2013 were analysed. We assessed baseline variables including patients'' characteristics (age, sex, disease duration, prednisolone dose, methotrexate dose, other disease-modifying antirheumatic drug use, Clinical Disease Activity Index [CDAI]) and serum biomarker levels (C-reactive protein, immunoglobulin M-rheumatoid factor, anti-cyclic citrullinated protein/peptide antibodies, interferon-γ, interleukin (IL)-1β, IL-2, IL-6, IL-8, IL-10, IL-17, tumor necrosis factor-α, soluble intercellular adhesion molecule-1, bone alkaline phosphatase, osteonectin, osteopontin) to extract factors associated with clinical remission (CDAI≤2.8) at 1 year using univariate analyses, and the extracted factors were entered into a multivariate logistic regression model. Similar analyses were also performed for Simplified Disease Activity Index (SDAI) remission (≤3.3) and Disease Activity Score with 28 joint counts, erythrocyte sedimentation rate (DAS28-ESR) remission (<2.6).ResultsThere were no significant differences in the baseline characteristics except for methotrexate use between the groups. In the multivariate analyses, the low baseline osteopontin levels (OR 0.9145, 95% CI 0.8399–0.9857) were identified as predictors of CDAI remission in the tocilizumab group, whereas no predictors of CDAI remission were found in the infliximab group. Similar results were obtained when using SDAI and DAS28-ESR remission criteria.ConclusionBaseline low serum osteopontin levels predict clinical remission 1 year after tocilizumab treatment and not infliximab treatment in biologic-naïve patients with rheumatoid arthritis. Our prediction model provided insights into how to optimize the choice of biologics and warrants external validation in other cohorts. 相似文献

Exercise is known to induce the oxidation of blood low-molecular-weight (LMW) thiols such as reduced glutathione (GSH). We previously reported that full-marathon running induced a decrease in human plasma levels of protein-bound sulfhydryl groups (p-SHs). Moderate exercise, a 30-min running at the intensity of the individual ventilatory threshold, performed by untrained healthy females caused a significant decrease in erythrocyte levels of p-SHs (mostly hemoglobin cysteine residues) and LMW thiols, but their levels returned to each baseline by 2 h. No significant change in plasma LMW thiols was observed. However, plasma levels of p-SHs significantly decreased after running and remained unchanged after 24 h. These results suggest that moderate exercise causes the oxidation of blood thiols, especially protein-bound thiols. 相似文献
