By preparative polyacrylamide gel electrophoresis at pH 8.5, and in the absence of nickel ions, two types of subunit dimers of the NAD-linked hydrogenase from Nocardia opaca 1b were separated and isolated, and their properties were compared with each other as well as with the properties of the native enzyme. The intact hydrogenase contained 14.3 +/- 0.4 labile sulphur, 13.6 +/- 1.1 iron and 3.8 +/- 0.1 nickel atoms and approximately 1 FMN molecule per enzyme molecule. The oxidized hydrogenase showed an absorption spectrum with maxima (shoulders) at 380 nm and 420 nm and an electron spin resonance (ESR) spectrum with a signal at g = 2.01. The midpoint redox potential of the Fe-S cluster giving rise to this signal was +25 mV. In the reduced state, hydrogenase gave characteristic low-temperature (10-20 K) and high-temperature (greater than 40 K) ESR spectra which were interpreted as due to [4Fe-4S] and [2Fe-2S] clusters, respectively. The midpoint redox potentials of these clusters were determined to be -420 mV and -285 mV, respectively. The large hydrogenase dimer, consisting of subunits with relative molecular masses Mr, of 64000 and 31000, contained 9.9 +/- 0.4 S2- and 9.3 +/- 0.5 iron atoms per protein molecule. This dimer contained the FMN molecule, but no nickel. The absorption and ESR spectra of the large dimer were qualitatively similar to the spectra of the whole enzyme. This dimer did not show any hydrogenase activity, but reduced several electron acceptors with NADH as electron donor (diaphorase activity). The small hydrogenase dimer, consisting of subunits with Mr of 56000 and 27000, was demonstrated to have substantially different properties. For iron and labile sulphur average values of 3.9 and 4.3 atoms/dimer molecule have been determined, respectively. The dimer contained, in addition, about 2 atoms of nickel and was free of flavins. In the oxidized state this dimer showed an absorption spectrum with a broad band in the 400-nm region and a characteristic ESR signal at g = 2.01. The reduced form of the dimer was ESR-silent. The small dimer alone was diaphorase-inactive and did not reduce NAD with H2, but it displayed high H2-uptake activities with viologen dyes, methylene blue and FMN, and H2-evolving activity with reduced methyl viologen. Hydrogen-dependent NAD reduction was fully restored by recombining both subunit dimers, although the reconstituted enzyme differed from the original in its activity towards artificial acceptors and the ESR spectrum in the oxidized state.  相似文献

A continuous line derived from a human skin squamous cell carcinoma has been grown in media of high, normal and low Ca2+ concentrations. The growth rate was unaffected by the Ca2+ levels even though morphological changes were observed. Desmosomes were absent at low Ca2+ and areas of cell piling were observed at high Ca2+. Cell protein staining patterns on polyacrylamide gels were identical for cells grown at the three Ca2+ levels. The variations were minor for the glycoproteins reacted with 125I-conA. Lactoper-oxidase iodination revealed changes in cell surface proteins, most markedly in the emergence of new proteins at high Ca2+.  相似文献

The glycoprotein from Friend murine leukemia virus was digested with protease from Staphylococcus aureus V8. A glycopeptide comprising the N-terminal glycosylation site (Asn-12) was isolated from the mixture of fragments and analyzed by amino acid sequencing and methylation-capillary gas chromatography-mass spectrometry before and after treatment with sialidase from Vibrio cholerae. Asn-12 was thus found to be substituted by a family of partially sialylated, fucosylated, and intersected glycoprotein N-glycans of the hybrid type.  相似文献

Renal cell cultures were initiated using fresh autopsy material from two individuals with cystinosis, ages 5 and 8 yr. Cells obtained from collagenase treated autopsy material were grown in a selective kidney medium containing Coon's modified F12, 2.5% fetal bovine serum, transferrin, insulin, selenium, hydrocortisone, PGE1, and fibronectin. These cells had an epithelial appearance, formed domes, and were periodic acid-Schiff positive. Both tight junctions and microvilli were seen by electron microscopy. Fibroblasts had a cloning efficiency of zero in the selective medium and grew poorly compared to their growth in Coon's F12 with 10% fetal bovine serum. The lysosomal cystine content of the renal cells was greatly elevated and comparable to that of fibroblasts from cystinotic patients. Renal cell lysosomal cystine levels were only partially reduced by exposure to either pantethine or the aminothiol, cysteamine. However, exposure to either compound effectively depleted cystinotic cultured fibroblasts of their lysosomal cystine. Study of cultured renal material may have practical significance in pharmacologic considerations.  相似文献

Behaviour of the egg-laying Monarch in captivity suggests that the concentration and quality of cardiac glycosides in the food plant are not important oviposition cues. The presence of eggs (as previously noted by Urquhart, 1960) and larvae feeding on the food plant, act as mild deterrents.
The butterfly's emetic potency (see Table XIII(a)) can sometimes surpass that of the leaves of the host plant itself. Unidentified factors, providing the internal plant environment, are more important as cardiac glycoside storage stimulants than either the quantity or quality of the cardenolides present. In the laboratory D. plexippus oviposited preferentially on a plant with relatively low cardiac glycoside content, but which produced the most powerfully protected (emetic) adult.
Metabolic changes during the pharate pupal stage, but also, in the case of Euploea core , in the larval fifth instar, rather than larval sequestration, may account for the major increase or decrease in butterfly toxicity compared with that of the food plant.
Temperature does not affect the storage of cardenolides except indirectly by altering metabolic rate. There is no evidence to support the concept that current "physiological cost" of cardenolide storage is high. Like the toad, this butterfly can be assumed to have evolved an enzvmatic system well adjusted to the presence of cardenolides in its bodv tissues.  相似文献

When the thymidine analog BrdU was incorporated into the DNA of a fibroblast cell line derived from the cactus mouse Peromyscus eremicus, a chromosome region with an increased frequency of gaps and breaks was observed. Nearly a third of the chromatid aberrations found at this site were associated with a sister-chromatid exchange (SCE) although this chromosome region showed no increase in sister-chromatid exchange in the absence of a gap or break. SCEs were significantly decreased in the remainder of the chromosome arm when it contained an aberration at the unstable site. This BrdU-sensitive region, unlike others reported, was found not to be late-replicating. — In this chromosome complement, the frequency of sisterchromatid exchange in C-band positive regions was significantly lower than that in C-band negative regions.  相似文献
