The wild banana Musa ornata is an inhabitant of the tropical regions of Mexico characterized by patches of tropical rainforest. The overexploitation of its habitat has caused the extinction of several populations affecting diversity and population genetic structure of remaining ones. We used microsatellite markers to determine the genetic diversity and the population’s genetic structure of all extant populations. The thirty-two microsatellite loci previously characterized for M. acuminata and M. balbisiana were tested in M. ornata. Only twelve amplified. From these seven were polymorphic and were used for genetic analyses. The Nei’s diversity estimator shows low levels of genetic diversity (H e = 0.263) with a mean of 4.40 alleles per locus. Excess homozygosity was evident in all populations indicating high levels of inbreeding. F ST pairwise analyses and AMOVA indicated low genetic differentiation. However, 28 % of private alleles were registered, suggesting limited gene flow. Genetic distances, Jaccard’s coefficient and principal component analysis showed a good correspondence to geographical locations. The Mantel test performed was not significant. The results support the hypothesis of recent fragmentation events; therefore, not enough time has passed to detect differences between populations. However, it is also likely that results are caused by factors such as bottleneck, decline in pollinator populations, self-pollination and/or a tendency towards clonal reproduction. It is proposed that the preservation strategy focuses on maintaining all the remaining populations and ensuring their connectivity, so as to maintain gene flow and increase the genetic diversity of this species.  相似文献

Serine-arginine-rich (SR) proteins are essential for splicing in metazoans but are absent in yeast. By contrast, many fungi have SR protein homologs with variable arginine-rich regions analogous to the arginine-serine-rich (RS) domain in metazoans. The density of RS repeats in these regions correlates with the conservation of the branch site signal, providing evidence for an ancestral origin of SR proteins and indicating that the SR proteins and the branch site co-evolved.  相似文献

Background and Aims

Excessive uptake of commensal bacterial antigens through a permeable intestinal barrier may influence host responses to specific antigen in a genetically predisposed host. The aim of this study was to investigate whether intestinal barrier dysfunction induced by indomethacin treatment affects the host response to intestinal microbiota in gluten-sensitized HLA-DQ8/HCD4 mice.

Methodology/Principal Findings

HLA-DQ8/HCD4 mice were sensitized with gluten, and gavaged with indomethacin plus gluten. Intestinal permeability was assessed by Ussing chamber; epithelial cell (EC) ultra-structure by electron microscopy; RNA expression of genes coding for junctional proteins by Q-real-time PCR; immune response by in-vitro antigen-specific T-cell proliferation and cytokine analysis by cytometric bead array; intestinal microbiota by fluorescence in situ hybridization and analysis of systemic antibodies against intestinal microbiota by surface staining of live bacteria with serum followed by FACS analysis. Indomethacin led to a more pronounced increase in intestinal permeability in gluten-sensitized mice. These changes were accompanied by severe EC damage, decreased E-cadherin RNA level, elevated IFN-γ in splenocyte culture supernatant, and production of significant IgM antibody against intestinal microbiota.

Conclusion

Indomethacin potentiates barrier dysfunction and EC injury induced by gluten, affects systemic IFN-γ production and the host response to intestinal microbiota antigens in HLA-DQ8/HCD4 mice. The results suggest that environmental factors that alter the intestinal barrier may predispose individuals to an increased susceptibility to gluten through a bystander immune activation to intestinal microbiota.  相似文献

Summary At pH 4.0, >10–7 m nigericin was found capable of conducting net charge transfer across bimolecular lecithin membranes, with a stoichiometry of three uncharged ionophore moieties per cation. At neutral or alkaline pH, nigericin catalyzed the transfer of net charge through dimer forms. In agreement with these results, quantitative analysis of nigericin-potassium complexes formed at pH 4.0 showed a 31 ratio, and a 21 ratio at neutral or alkaline pH. A 11 stoichiometry was observed when the ionophore complex was not transferred from methanol-water to chloroform. Moreover,1H-NMR spectra of nigericin-cation complexes formed at pH 4.0, displayed clear-cut chemical shift variations different to those observed at neutral or alkaline pH. Thus, it is apparent that acid pH causes a transition from dimeric to trimeric forms of nigericin-cation complexes. The membrane conductance increased up to ten times when negatively charged phosphatidyl glycerol was used, while the conductance decreased in positively charged cetylpyridinium containing membranes at pH 4.0. These results suggest that the nigericin-K+ oligomeric complex is positively charged. In this respect, pK a values around 8.0 were obtained for the nigericin carboxylate group in media of different dielectric constant, indicating that this chemical group is undissociated under these conditions. Moreover, the values for the complex formation constants as well as the G values calculated for the dimers and trimers indicated that such ionophore cation oligomeric complexes are thermodynamically stable.  相似文献

Cyperaceae, the third largest family of monocotyledons, show unusual asexual reproductive strategies, such as pseudovivipary involving the formation of new individuals from somatic tissues of floral structures. In Eleocharis pseudovivipary is an important reproductive process in series Tenuissimae, which results in a structure called a proliferous spikelet. With the aim of describing the ontogenetic process of the pseudovivipary in E. viridans, proliferous spikelets at different developmental stages were analysed by light microscopy and scanning electron microscopy. The proliferous spikelet develops from a meristem located in the axil of the basal glume, homologous to bracts of other groups of Cyperaceae, in the culm of the parental plant. Each proliferous spikelet is formed by sympodial units, which consist of an addorsed prophyll, an outer and inner sheath and a culm, which develops a primordium of floriferous spikelet at the terminal region, which can be aborted, and may develop a proliferous spikelet lateral in the axil of the basal glume. The second internode of each sympodial unit contains a root primordium and an intercalary meristem at the culm base. Our results indicate that pseudovivipary can coexist with sexual reproduction as an alternative reproductive strategy, allowing the rapid spread of populations. © 2014 The Linnean Society of London, Botanical Journal of the Linnean Society, 2014, 176 , 524–539.  相似文献

As part of a project aiming to characterize the role of maize plastidial transglutaminase (chlTGZ) in the plant chloroplast, this paper presents results on stress induced by continuous chlTGZ over-expression in transplastomic tobacco leaves. Thylakoid remodelling induced by chlTGZ over-expression in young leaves of tobacco chloroplasts has already been reported (Ioannidis et al. in Biochem Biophys Acta 1787:1215–1222, 2009). In the present work, we determined the induced alterations in the photosynthetic apparatus, in the chloroplast ultrastructure, and, particularly, the activation of oxidative and antioxidative metabolism pathways, regarding ageing and functionality of the tobacco transformed plants. The results revealed that photochemistry impairment and oxidative stress increased with transplastomic leaf age. The decrease in pigment levels in the transformed leaves was accompanied by an increase in H2O2 and lipid peroxidation. The rise in H2O2 correlated with a decrease in catalase activity, whereas there was an increase in peroxidase activity. In addition, chlTGZ over-expression lead to a drop in reduced glutathione, while Fe-superoxide dismutase activity was higher in transformed than in wild-type leaves. Together with the induced oxidative stress, the over-expressed chlTGZ protein accumulated progressively in chloroplast inclusion bodies. These traits were accompanied by thylakoid scattering, membrane degradation and reduction of thylakoid interconnections. Consequently, the electron transport between photosystems decrease in the old leaves. In spite of these alterations, transplastomic plants can be maintained and reproduced in vitro. These results are discussed in line with chlTGZ involvement in chloroplast functionality.  相似文献

Reliable soil organic carbon (SOC) stock measurements of all major ecosystems are essential for predicting the influence of global warming on global soil carbon pools, but hardly any detailed soil survey data are available for tropical montane cloud forests (TMCF) and adjacent high elevation grasslands above (puna). TMCF are among the most threatened of ecosystems under current predicted global warming scenarios. We conducted an intensive soil sampling campaign extending 40 km along the tree line in the Peruvian Andes between 2994 and 3860 m asl to quantify SOC stocks of TMCF, puna grassland, and shrubland sites in the transition zone between the two habitats. SOC stocks from the soil surface down to the bedrock averaged (±standard error SE) 11.8 (±1.5, N = 24) kg C/m2 in TMCF, 14.7 (±1.4, N = 9) kg C/m2 in the shrublands and 11.9 (±0.8, N = 35) kg C/m2 in the grasslands and were not significantly different (P > 0.05 for all comparisons). However, soil profile analysis revealed distinct differences, with TMCF profiles showing a uniform SOC distribution with depth, shrublands a linear decrease, and puna sites an exponential decrease in SOC densities with soil depth. Organic soil layer thickness reached a maximum (~70 cm) at the upper limit of the TMCF and declined with increasing altitude toward puna sites. Within TMCF, no significant increase in SOC stocks with increasing altitude was observed, probably because of the large variations among SOC stocks at different sites, which in turn were correlated with spatial variation in soil depth.  相似文献

The formation of insoluble protein deposits in human tissues is linked to the onset of more than 40 different disorders, ranging from dementia to diabetes. In these diseases, the proteins usually self-assemble into ordered β-sheet enriched aggregates known as amyloid fibrils. Here we study the structure of the inclusions formed by maize transglutaminase (TGZ) in the chloroplasts of tobacco transplastomic plants and demonstrate that they have an amyloid-like nature. Together with the evidence of amyloid structures in bacteria and fungi our data argue that amyloid formation is likely a ubiquitous process occurring across the different kingdoms of life. The discovery of amyloid conformations inside inclusions of genetically modified plants might have implications regarding their use for human applications.  相似文献
