The synthesis, crystallographic analysis and magnetic studies of six new copper(II) complexes of formulae [Cu(μ-ala)(im)(H2O)]n(ClO4)n (1), [Cu(μ-ala)(pz)(μ-ClO4)] (2), [Cu(μ-phe)(im)(H2O)]n(ClO4)n (3), [Cu(μ-gly)(H2O)(ClO4)]n (4), [Cu(μ-gly)(pz)(ClO4)]n(5) and [Cu(μ-pro)(pz)(ClO4)]n (6) have been carried out (ala = alanine; phe = phenylalanine; gly = glycine; pro = proline; im = imidazole; pz = pyrazole). In all cases, the deprotonated aminoacid ligand acts as chelate through the N(amine) and one O(carboxylato), whereas the second O atom of the same carboxylato acts as a bridge to the neighbouring copper(II) ion. The coordination of copper(II) ions is square-pyramidal in all complexes but 2 (elongated Oh). All complexes (1–6) are uniform chains with syn–anti (equatorial–equatorial) coordination mode of the carboxylato bridging ligand, exhibiting intrachain ferromagnetic interactions. 相似文献

Cocaine dependence is a neuropsychiatric disorder in which both environmental and genetic factors are involved. Several processes, that include reward and neuroadaptations, mediate the transition from use to dependence. In this regard, dopamine and serotonin neurotransmission systems are clearly involved in reward and other cocaine‐related effects, whereas neurotrophic factors may be responsible for neuroadaptations associated with cocaine dependence. We examined the contribution to cocaine dependence of 37 genes related to the dopaminergic and serotoninergic systems, neurotrophic factors and their receptors through a case–control association study with 319 single nucleotide polymorphisms selected according to genetic coverage criteria in 432 cocaine‐dependent patients and 482 sex‐matched unrelated controls. Single marker analyses provided evidence for association of the serotonin receptor HTR2A with cocaine dependence [rs6561333; nominal P‐value adjusted for age = 1.9e?04, odds ratio = 1.72 (1.29–2.30)]. When patients were subdivided according to the presence or absence of psychotic symptoms, we confirmed the association between cocaine dependence and HTR2A in both subgroups of patients. Our data show additional evidence for the involvement of the serotoninergic system in the genetic susceptibility to cocaine dependence. 相似文献

The cps5-138 fission yeast mutant shows an abnormal lemon-like morphology at 28 degrees C in minimal medium and a lethal thermosensitive phenotype at 37 degrees C. Cell growth is completely inhibited at 28 degrees C in a Ca2+-free medium, in which the wild type is capable of growing normally. Under these conditions, actin patches become randomly distributed throughout the cell, and defects in septum formation and subsequent cytokinesis appear. The mutant cell is hypersensitive to the cell wall-digesting enzymatic complex Novozym234 even under permissive conditions. The gene SPBC31E1.02c, which complements all the mutant phenotypes described above, was cloned and codes for the Ca2+-ATPase homologue Pmr1p. The gene is not essential under optimal growth conditions but is required under conditions of low Ca2+ (<0.1 mM) or high temperature (>35 degrees C). The green fluorescent protein-tagged Cps5 proteins, which are expressed under physiological conditions (an integrated single copy with its own promoter in the cps5Delta strain), display a localization pattern typical of endoplasmic reticulum proteins. Biochemical analyses show that 1,3-beta-D-glucan synthase activity in the mutant is decreased to nearly half that of the wild type and that the mutant cell wall contains no detectable galactomannan when the cells are exposed to a Ca2+-free medium. The mutant acid phosphatase has an increased electrophoretic mobility, suggesting that incomplete protein glycosylation takes place in the mutant cells. These results indicate that S. pombe Pmr1p is essential for the maintenance of cell wall integrity and cytokinesis, possibly by allowing protein glycosylation and the polarized actin distribution to take place normally. Disruption and complementation analyses suggest that Pmr1p shares its function with a vacuolar Ca2+-ATPase homologue, Pmc1p (SPAPB2B4.04c), to prevent lethal activation of calcineurin for cell growth. 相似文献

In this work the porin Omp1 of Serratia marcescens was expressed in a porin deficient mutant (Escherichia coli UH302) and its functionality studied following the accumulation of ciprofloxacin in bacteria. The protein was extracted, purified and reconstituted in proteoliposomes of different composition (lipopolysaccharide (LPS), 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC) and, 1,2-dimyristoyl-sn-glycero-3-phosphocholine (DMPC)). Maximum extraction of the detergent was achieved applying different steps of dialysis and centrifugation. Proteolipid sheets with different composition were spread onto mica and observed by atomic force microscopy. Two-dimensional crystal of Omp1 was not observed in any case due to low resolution achieved. The number of existing protein sequences spans a very small fraction of sequence space. Natural proteins have overcome a strong negative selective pressure to avoid the formation of insoluble aggregates. Stably folded globular proteins and intrinsically disordered proteins (IDPs) use alternative solutions to the aggregation problem. While in globular proteins folding minimizes the access to aggregation prone regions, IDPs on average display large exposed contact areas. Here, we introduce the concept of average meta-structure correlation maps to analyze sequence space. Using this novel conceptual view we show that representative ensembles of folded and ID proteins show distinct characteristics and respond differently to sequence randomization. By studying the way evolutionary constraints act on IDPs to disable a negative function (aggregation) we might gain insight into the mechanisms by which function-enabling information is encoded in IDPs. 相似文献

Propionic (PA) and methylmalonic (MMA) acidurias are inherited disorders caused by deficiency of propionyl-CoA carboxylase
and methylmalonyl-CoA mutase, respectively. Affected patients present acute metabolic crises in the neonatal period and long-term
neurological deficits. Treatments of these diseases include a protein restricted diet and l-carnitine supplementation. l-Carnitine is widely used in the therapy of these diseases to prevent secondary l-carnitine deficiency and promote detoxification, and several recent in vitro and in vivo studies have reported antioxidant
and antiperoxidative effects of this compound. In this study, we evaluated the oxidative stress parameters, isoprostane and
di-tyrosine levels, and the antioxidant capacity, in urine from patients with PA and MMA at the diagnosis, and during treatment
with l-carnitine and protein-restricted diet. We verified a significant increase of isoprostanes and di-tyrosine, as well as a significant
reduction of the antioxidant capacity in urine from these patients at diagnosis, as compared to controls. Furthermore, treated
patients presented a marked reduction of isoprostanes and di-tyrosine levels in relation to untreated patients. In addition,
patients with higher levels of protein and lipid oxidative damage, determined by di-tyrosine and isoprostanes levels, also
presented lower urinary concentrations of total and free l-carnitine. In conclusion, the present results indicate that treatment with low protein diet and l-carnitine significantly reduces urinary biomarkers of protein and lipid oxidative damage in patients with disorders of propionate

Surplus biological sludge from wastewater treatment operations was converted into activated carbon and then added to the aerated vessel of an activated sludge process treating phenol and glucose. The addition of activated carbon, either sludge-based or commercial, enhanced phenol removal from 58 to 98.7% and from 87 to 93% for COD with feed concentrations of 100 mg phenol l–1 and 2500 mg COD l–1. No differences were found between the activated sludge-activated carbon bench scale continuous reactors operating with either commercial or sludge-based activated carbon in spite of the higher adsorption capacity of the former. 相似文献
