T细胞抗原受体(T ceIl receptor,TCR)是T细胞表面关键的受体分子。TCR特异性地识别各种多肽抗原并通过胞内区ITAM磷酸化传递抗原刺激信号,进而引发T细胞的免疫效应。TCR的活性异常将会导致自身免疫病和免疫缺陷病的发生。对于TCR结构和功能的深入研究有助于我们更好地理解免疫反应的分子机理,从而为相关免疫疾病的预防和治疗提供重要的理论依据。该文对TCR的分类、基因重排机制、受体组装方式及其结构基础、TCR对抗原的识别以及活化机制等方面的研究成果进行了总结,综述了近几年来的最新研究进展。 相似文献

The high‐capacity cathode material V2O5·n H2O has attracted considerable attention for metal ion batteries due to the multielectron redox reaction during electrochemical processes. It has an expanded layer structure, which can host large ions or multivalent ions. However, structural instability and poor electronic and ionic conductivities greatly handicap its application. Here, in cell tests, self‐assembly V2O5·n H2O nanoflakes shows excellent electrochemical performance with either monovalent or multivalent cation intercalation. They are directly grown on a 3D conductive stainless steel mesh substrate via a simple and green hydrothermal method. Well‐layered nanoflakes are obtained after heat treatment at 300 °C (V2O5·0.3H2O). Nanoflakes with ultrathin flower petals deliver a stable capacity of 250 mA h g?1 in a Li‐ion cell, 110 mA h g?1 in a Na‐ion cell, and 80 mA h g?1 in an Al‐ion cell in their respective potential ranges (2.0–4.0 V for Li and Na‐ion batteries and 0.1–2.5 V for Al‐ion battery) after 100 cycles. 相似文献

Three new sesquiterpenoids, 4α‐hydroxyeudesm‐11(13)‐en‐12‐yl 3‐methylbutanoate ( 1 ), diaspanolide E ( 2 ), and (13α)‐germacra‐1(10),4‐dien‐12,8α‐olid‐15‐oic acid ( 3 ), along with eight known sesquiterpenoids ( 4 – 11 ), were isolated from the aerial parts of Ainsliaea henryi. The chemical structures of compounds 1 – 3 were elucidated by spectroscopic analysis (1D‐, 2D‐NMR, MS and HR/MS). All isolates were evaluated for their inhibitory activities against nitric oxide (NO) production in lipopolysaccharide‐induced RAW264.7 macrophage cells. Compound 10 exhibited significantly inhibition against NO release with an IC50 value of 6.54 ± 0.16 μm . Also, all isolated compounds were tested for cytotoxicity against three human tumor cell lines A549, MGC803, and HCT116, among which compound 5 significantly inhibited the proliferation of MGC803 cell lines with an IC50 value of 2.2 ± 0.2 μm . 相似文献

This study details the introduction of a gfp marker into an endophytic bacterial strain (Achromobacter marplatensis strain 17, isolated from sugar beet) to monitor its colonization of sugar beet (Beta. vulgaris L.). Stability of the plasmid encoding the gfp was confirmed in vitro for at least 72 h of bacterial growth and after the colonization of tissues, under nonselective conditions. The colonization was observed using fluorescence microscopy and enumeration of culturable endophytes in inoculated sugar beet plants that grew for 10 or 20 days. gfp-Expressing strains were re-isolated from the inner tissues of surface-sterilized roots and stems of inoculated plants, and the survival of the Achromobacter marplatensis 17:gfp strain in plants 20 days after inoculation, even in the absence of selective pressure, suggests that it is good colonizer. These results also suggest that this strain could be a useful tool for the delivery of enzymes or other proteins into plants. In addition, the study highlights that sugar beet plants can be used effectively for detailed in vitro studies on the interactions between A. marplatensis strain 17 and its host, particularly if a gfp-tagged strain of the pathogen is used. 相似文献

The enzyme chondroitin polymerizing factor (ChPF) is primarily involved in extension of the chondroitin sulfate backbone required for the synthesis of sulfated glycosaminoglycan (sGAG). Transforming growth factor beta (TGF‐β) upregulates sGAG synthesis in nucleus pulposus cells; however, the mechanisms mediating this induction are incompletely understood. Our study demonstrated that ChPF expression was negatively correlated with the grade of degenerative intervertebral disc disease. Treatment of nucleus pulposus cells with TGF‐β induced ChPF expression and enhanced Smad2/3, RhoA/ROCK activation, and the JNK, p38, and ERK1/2 MAPK signaling pathways. Selective inhibitors of Smad2/3, RhoA or ROCK1/2, and knockdown of Smad3 and ROCK1 attenuated ChPF expression and sGAG synthesis induced by TGF‐β. In addition, we showed that RhoA/ROCK1 signaling upregulated ChPF via activation of the JNK pathway but not the p38 and ERK1/2 signaling pathways. Moreover, inhibitors of JNK, p38 and ERK1/2 activity also blocked ChPF expression and sGAG synthesis induced by TGF‐β in a Smad3‐independent manner. 肝星状细胞(hepatic stellate cell,HSC)是肝纤维化发展过程中过量细胞外基质的主要来源。该研究首先利用MTT法检测IL-13实验剂量和时间条件下肝星状细胞增殖情况;然后运用RT-PCR技术检测IL-13对人肝星状细胞LX-2细胞系IL-13Rα1、IL-4Rα、TGF-β和Ⅰ型胶原蛋白转录水平的影响;最后通过羟脯氨酸法定量分析各组细胞培养上清液中的胶原蛋白含量。结果发现:IL-13能促进肝星状细胞的增殖;在不改变IL-13Rα1和IL-4Rα转录水平的同时,对TGF-β和Ⅰ型胶原蛋白mRNA的表达以及细胞总胶原蛋白含量的上调作用均呈现出较为明显的剂量和时间依赖性。 相似文献

Four gram-negative, aerobic, motile, non-spore, forming rods with a wide pH and temperature range for growth (pH 7.0–11.0,
optimum pH 8.0; 20–45°C, optimum 28°C) strains were isolated from root nodules of Sphaerophysa salsula and characterized by means of a polyphasic approach. Phylogenetic analysis based on 16S rRNA gene sequences revealed that
the four strains formed a new lineage related to the genus Rhizobium and the sequence similarities between the isolate and the most related type strain Rhizobium giardinii was 96.5%. These strains also formed a distinctive group from the reference strains for defined Rhizobium species based on housekeeping gene sequences (atpD and recA), BOX-PCR fingerprinting, phenotypic features and symbiotic properties. The representative strain CCNWGS0238T has DNA-DNA relatedness of less than 33.4% with the most closely related species R. giardinii. It is therefore proposed as a new species, Rhizobium sphaerophysae sp. nov., with isolate CCNWGS0238T (=ACCC17498T = HAMBI3074T) as the type strain. 相似文献
