The structural elucidation of metabolites of penehyclidine in rats, a novel anti-cholinergic drug, by the method of liquid chromatography-electrospray ionization mass spectrometry, gas chromatography-mass spectrometry with electron impact ion source and stable isotope ion cluster was described. Identification and elucidation of the phase I and phase II metabolites were performed by comparing the daughter ion pairs of stable isotope cluster, changes of the protonated molecular masses, full scan MS(n) spectra and retention times with those of the parent drug, penehyclidine and penehyclidine deuterium-labeled. Penehyclidine was easily biotransformed by the pathways of oxidative, hydroxylated, methoxylated and phase II conjugated reactions to form several metabolites that retained the some features of the parent molecules. Twelve metabolites (penehyclidine monoxide, hydroxypenehyclidine, penehyclidine dioxide, hydroxypenehyclidine monoxide, dihydroxypenehyclidine, dihydroxypenehyclidine (position isomer), dihydroxypenehyclidine monoxide, trihydroxypenehyclidine, methoxypenehyclidine dioxide, dimethoxypenehyclidine, trihydroxymethoxypenehyclidine and glucuronide conjugated hydroxypenehyclidine) were identified. The results from electrospray ion and electron impact ion data with the stable isotope cluster showed the qualitative differences in the mass spectral patterns, suggesting that these technologies should be used in parallel to ensure comprehensive metabolites detection and characterization. Background
Gram-positive bacteria of the genus Anoxybacillus have been found in diverse thermophilic habitats, such as geothermal hot springs and manure, and in processed foods such as gelatin and milk powder. Anoxybacillus flavithermus is a facultatively anaerobic bacterium found in super-saturated silica solutions and in opaline silica sinter. The ability of A. flavithermus to grow in super-saturated silica solutions makes it an ideal subject to study the processes of sinter formation, which might be similar to the biomineralization processes that occurred at the dawn of life.Results
We report here the complete genome sequence of A. flavithermus strain WK1, isolated from the waste water drain at the Wairakei geothermal power station in New Zealand. It consists of a single chromosome of 2,846,746 base pairs and is predicted to encode 2,863 proteins. In silico genome analysis identified several enzymes that could be involved in silica adaptation and biofilm formation, and their predicted functions were experimentally validated in vitro. Proteomic analysis confirmed the regulation of biofilm-related proteins and crucial enzymes for the synthesis of long-chain polyamines as constituents of silica nanospheres.Conclusions
Microbial fossils preserved in silica and silica sinters are excellent objects for studying ancient life, a new paleobiological frontier. An integrated analysis of the A. flavithermus genome and proteome provides the first glimpse of metabolic adaptation during silicification and sinter formation. The effects of sheep urine deposition volume (0, 1, 2 or 4 L/m2) and deposition stage of plant growth (vegetative or reproductive) on the number and size of tillers/branches and the biomass of Stipa bungeana, Artemisia capillaries and Lespedeza davurica in a Chinese steppe grassland were determined. The results indicate that the response of the three plant species to sheep urine deposition differs, and is influenced by both urine deposition volume and deposition stage of plant growth. Urine deposition had a short-term scorch effect on grassland plants, which mainly occurred in the inner zone of urine patches. Urine application had a long-term positive effect on S. bungeana and a long-term negative effect on A. capillaries and L. davurica, which lasted at least two years and decreased with decrease in urine deposition volume. All species growing in the inner zone of urine patches were scorched by sheep urine deposition, some species in the marginal zone of patches were also scorched, while no species were scorched in the outer zones. The reproductive and vegetative stages of A. capillaries and the reproductive stages of S. bungeana and L. davurica were sensitive to sheep urine deposition. 相似文献

Chemical synapses are asymmetric intercellular junctions through which neurons send nerve impulses to communicate with other neurons or excitable cells. The appropriate formation of synapses, both spatially and temporally, is essential for brain function and depends on the intercellular protein-protein interactions of cell adhesion molecules (CAMs) at synaptic clefts. The CAM proteins link pre- and post-synaptic sites, and play essential roles in promoting synapse formation and maturation, maintaining synapse number and type, accumulating neurotransmitter receptors and ion channels, controlling neuronal differentiation, and even regulating synaptic plasticity directly. Alteration of the interactions of CAMs leads to structural and functional impairments, which results in many neurological disorders, such as autism, Alzheimer’s disease and schizophrenia. Therefore, it is crucial to understand the functions of CAMs during development and in the mature neural system, as well as in the pathogenesis of some neurological disorders. Pathogenicity ofHeliothis nuclear polyhedrosis virus (HSNPV) to the corn earworm,Heliothis armigera, was studied using 3 different inoculative methods. The LD50 values of 4th-instar larvae inoculated with corn-fed, diet-fed and inoculum-imbiding method were 1.85×106, 2.55×105 and 1,22×103 PIBs/larva, respectively. The inoculum-imbiding is more sensitive and convenient for inoculatingH. armigera with HSNPV. The HSNPV product, Elcar®, was highly pathogenic toH. armigera, the LD50 values of 2nd-, 3rd- and 4th-instar larvae being 27, 83 and 1,221 PIBs/larva, respectively, as measured by the inoculum-imbiding method. The mortality of 4th-instar larvae caused by HSNPV was increased, but the incubation period was shortened with higher incubation temperatures. However, the high temperature at 35°C caused a lower mortality, and a prolongation of the median lethal time (LT50). Stability and persistence of HSNPV preparations were better in January–February and April–May than in June–July and October–November periods when sprayed on corn silks under field conditions. The HSNPV was inactivated by weak alkaline dew (pH 8.1) collected from soybean leaves, but it remained active on those from corn, tomato and asparagus with pH 7.2–7.3. The artificial heavy rainfall of 242 mm/h for 30 min did not wash off HSNPV preparations sprayed on the corn silks. 相似文献
