The messenger RNAs of many proto-oncogenes, cytokines and lymphokines are targeted for rapid degradation through AU-rich elements (AREs) located in their 3'' untranslated regions (UTRs). HuR, a ubiquitously expressed member of the Elav family of RNA binding proteins, exhibits specific affinities for ARE-containing RNA sequences in vitro which correlate with their in vivo decay rates, thereby implicating HuR in the ARE-mediated degradation pathway. We have transiently transfected HuR into mouse L929 cells and observed that overexpression of HuR enhances the stability of beta-globin reporter mRNAs containing either class I or class II AREs. The increase in mRNA stability parallels the level of HuR overexpression, establishing an in vivo role for HuR in mRNA decay. Furthermore, overexpression of HuR deletion mutants lacking RNA recognition motif 3 (RRM 3) does not exert a stabilizing effect, indicating that RRM 3 is important for HuR function. We have also developed polyclonal anti-HuR antibodies. Immunofluorescent staining of HeLa and L929 cells using affinity-purified anti-HuR antibody shows that both endogenous and overexpressed HuR proteins are localized in the nucleus. By forming HeLa-L929 cell heterokaryons, we demonstrate that HuR shuttles between the nucleus and cytoplasm. Thus, HuR may initially bind to ARE-containing mRNAs in the nucleus and provide protection during and after their export to the cytoplasmic compartment.  相似文献

Guinea pig brain membranes treated with cyanogen bromide (CNBr) demonstrate a loss in the number of mu opioid receptors and a lower binding affinity of delta opioid receptors. These receptor changes are irreversible. Results from ligand protection experiments support the hypothesis that the location of the methionine groups, the sites at which CNBr cleaves peptides, differs between these two types of opioid receptors. Kappa receptors are significantly less sensitive to the action of CNBr than mu or delta receptors.  相似文献

We used data on loud duetted and solo songs collected from one habituated polygynous group of black‐crested gibbons (Nomascus concolor jingdongensis) on Mt. Wuliang, Yunnan, to test several hypotheses about the functions of these songs. The major functions proposed for loud gibbon songs include resource defense, mate defense, pairbonding, group cohesion and mate attraction. Duet bouts are generally initiated by adult males, who select the highest trees near to ridges or on steep slopes as singing trees. Such trees facilitate voice transmission and inter‐group communication. Singing trees tended to be located near important food patches and sleeping sites, which supports the resource defense hypothesis. The adult male and two adult females always sang interactively, alternating male phrases with the females' stereotyped great calls, to produce the duets, and females rarely produced great calls if they were more than 30 m from the male. The two females usually produced great calls synchronously during the duet, especially when they were close together. These features support both the mate defense and pairbonding hypotheses. The number of great calls and their degree of synchrony transmit information about spatial relationships and possibly pairbond strength to members to neighboring groups and floating animals. During or after the duet bouts, the adult females and juvenile moved toward to the adult male; and group members maintained a close spatial relationship, which supports the group cohesion hypothesis. Other incidents observed suggest a mate competition role for duets. The adult male always sang when the females started duetting with the subadult male. The subadult male sang solo bouts, but they were not more frequent or longer than bouts initiated by the adult male. Although mate attraction is the likely function of subadult solos, it was not convincingly demonstrated. In conclusion, all hypotheses concerning the function of singing are supported by at least some of the data, and none can be excluded. Am. J. Bacterial infections represent serious diseases in aquaculture, rapidly leading to fish death by septicemia. We investigated whether the electrotransfer of green fluorescent protein gene fusion epinecidin-1 (CMV-gfp-epi) DNA into zebrafish muscle could regulate the fish immune response and inhibit bacterial growth. Electroporation parameters such as the number of pulses, voltage, and amount of plasmid DNA were analyzed, and results demonstrated the greatest mRNA expression level of gfp-epi relative to β-actin was obtained with a pulse number of 4, a voltage strength of 100 V/cm, a concentration of DNA of 90 μg/fish, and electroporation for 96 h. In addition, the cytomegalovirus (CMV) promoter exhibited higher activity compared to the mylz promoter in muscle for electrotransfer in zebrafish. GFP fluorescence and gfp-epi mRNA expression in tissues after electroporation were also studied by a polymerase chain reaction, immunohistochemistry, and fluorescence microscopy. gfp-epi expression was significantly correlated with decreased bacterial numbers and immune-related gene expression. These data demonstrate that electroporation of epinecidin-1 might have provoked an inflammatory response that accounts for the improvement in bacterial clearance.  相似文献

Two kinds of chitosans, namely N-acetylated and N-deacetylated chitosan were prepared by the modified processes. They can dissolve in both acid and alkali solution. 13C NMR was used to study the basic solution of chitosan, and XRD, FT-IR and SEM were used to study the structure of N-acetylated and N-deacetylated chitosan. The result from X-ray diffraction showed that a transformation of crystal structure occurred during the N-acetylation or N-deacetylation process with the decrease of crystallinity and expansion of crystal lattices. FT-IR spectra revealed that the intermolecular and intramolecular hydrogen bonds were destroyed by both treatments and a looser structure was observed by the SEM. The lower crystallinity, the decreased intermolecular interactions, the more disordered and looser structure were easy for the permeation of LiOH/urea aqueous solution and coordinated with the breakage of intermolecular and intramolecular hydrogen bond by LiOH at low temperature, the prepared chitosans dissolved in LiOH/urea/H2O mixture.  相似文献

Supercapacitors, also known as electrochemical capacitors, can provide much faster charge–discharge, greater power density, and cyclability than batteries, but they are still limited by lower energy densities (or the amount of energy stored per unit volume). Here, a novel strategy for the synthesis of functional pillared graphene frameworks, in which graphene fragments in‐between graphene sheets, through simple thermal‐treatment of ozone (O3)‐treated graphene oxide at very low temperature of 200 °C is reported. Due to its high packing density, high content of stable oxygen species, and continues ion transport network in‐between graphene sheets, the functional pillared‐graphene framework delivers not only high gravimetric capacitance (353 F g?1 based on the mass of the active material) and ultrahigh volumetric capacitance (400 F cm?3 based on total mass of electrode material) in aqueous electrolyte but also excellent cyclic stability with 104% of its initial capacitance retention after 10 000 cycles. Moreover, the assembled symmetric supercapacitor achieves as high as 27 Wh L?1 of volumetric energy density at a power density of 272 W L?1. This novel strategy holds great promise for future design of high volumetric capacitance supercapacitors.  相似文献

In vitro digestion products of proteins were compared among beef, pork, chicken, and fish. Gastric and jejunal contents from the rats fed these meat proteins were also compared. Cooked pork, beef, chicken, and fish were homogenized and incubated with pepsin alone or followed by trypsin. The digestion products with molecular weights of less than 3000 Da were identified with MALDI‐TOF‐MS and nano‐LC‐MS/MS. Gastric and jejunal contents obtained from the rats fed the four meat proteins for 7 days were also analyzed. After pepsin digestion, pork, and beef samples had a greater number of fragments in similarity than chicken and fish samples, but the in vitro digestibility was the greatest (p < 0.05) for pork and the smallest for beef samples. After trypsin digestion, the species differences were less pronounced (p > 0.05). A total of 822 and 659 peptides were identified from the in vitro and in vivo digestion products, respectively. Our results could interpret for the differences in physiological functions after the ingestion of different species of meat.  相似文献
