Engineering the Substrate Specificity of a Thermophilic Penicillin Acylase from Thermus thermophilus

A homologue of the Escherichia coli penicillin acylase is encoded in the genomes of several thermophiles, including in different Thermus thermophilus strains. Although the natural substrate of this enzyme is not known, this acylase shows a marked preference for penicillin K over penicillin G. Three-dimensional models were created in which the catalytic residues and the substrate binding pocket were identified. Through rational redesign, residues were replaced to mimic the aromatic binding site of the E. coli penicillin G acylase. A set of enzyme variants containing between one and four amino acid replacements was generated, with altered catalytic properties in the hydrolyses of penicillins K and G. The introduction of a single phenylalanine residue in position α188, α189, or β24 improved the Km for penicillin G between 9- and 12-fold, and the catalytic efficiency of these variants for penicillin G was improved up to 6.6-fold. Structural models, as well as docking analyses, can predict the positioning of penicillins G and K for catalysis and can demonstrate how binding in a productive pose is compromised when more than one bulky phenylalanine residue is introduced into the active site. 相似文献

Antimicrobial resistance is a persistent problem in the public health sphere. However, recent attempts to find effective substitutes to combat infections have been directed at identifying natural antimicrobial peptides in order to circumvent resistance to commercial antibiotics. This study describes the development of synthetic peptides with antimicrobial activity, created in silico by site-directed mutation modeling using wild-type peptides as scaffolds for these mutations. Fragments of antimicrobial peptides were used for modeling with molecular modeling computational tools. To analyze these peptides, a decision tree model, which indicated the action range of peptides on the types of microorganisms on which they can exercise biological activity, was created. The decision tree model was processed using physicochemistry properties from known antimicrobial peptides available at the Antimicrobial Peptide Database (APD). The two most promising peptides were synthesized, and antimicrobial assays showed inhibitory activity against Gram-positive and Gram-negative bacteria. Colossomin C and colossomin D were the most inhibitory peptides at 5 μg/ml against Staphylococcus aureus and Escherichia coli. The methods described in this work and the results obtained are useful for the identification and development of new compounds with antimicrobial activity through the use of computational tools. 相似文献

Bcharreglaris amunobi n. gen., n. sp., Setoglaris reemae n.gen., n. sp., Libanoglaris chehabi n.sp., and Libanoglaris randatae n. sp. Ce catalogue des Vesperidae et Cerambycidae de la faune de France est fondé sur une vaste étude bibliographique et sur les observations des auteurs ainsi que celles, vérifiées, de nombreux amateurs. Les nombreuses modifications montrent la nécessité de la révision du dernier ouvrage de référence (Villiers 1978). Le bilan faunistique des recherches permet de dresser une liste de 251 taxa représentant 242 espèces actuellement présentes en France continentale et en Corse. Cette liste est sujette à de perpétuelles modifications liées aux fluctuations de populations, aux introductions et aux activités de recherche de nombreux amateurs. Ce travail de synthèse systématique, taxinomique et faunistique est une première étape pour la réalisation d’un atlas des Vesperidae et Cerambycidae de France et de Corse. 相似文献

The first forensic entomological study performed in Portugal is presented. Two piglet (Sus scrofa L.) carcasses were used to determine adult Calliphoridae activity on carrion over a period of 121 days, all along the end of spring and the summer, both in a shaded and a sunny site. Five decomposition stages were observed and a total of 10723 adult Calliphoridae, belonging to 11 species, were collected. Calliphora vicina, Calliphora vomitoria, Chrysomya albiceps and Lucilia caesar were the dominant species in this study. Decomposition was faster on the carcass exposed to the sun and the number of Calliphoridae specimens was higher there than in the shaded site. It was found a significant effect of the decomposition stage in the number of specimens attracted to the carcass, as well as a significant effect of the interaction between the decomposition stage and insolation regime. Calliphora and Lucilia species did not show preference for sunny or shaded areas. Important differences in the Calliphoridae community structure were found compared to other regions of the Iberian Peninsula, reinforcing the need of further studies in different environments and regions of this geographical area in order to collect information about the local necrophagous fauna used in forensic practice. 相似文献

The diet of Perla madritensis, endemic species of the northern half of the Iberian Peninsula, is described for the first time by means of a study carried out in north-western Spain. As other species of the genus Perla, this taxon behaves mainly as predator, with Chironomidae, followed by Baetidae, being the most abundant prey in its gut. Shifts in diet composition were detected in relation to size in this species, with smaller prey being replaced by larger ones such as Simuliidae and Leptophlebiidae when the nymphs become larger. 相似文献

Wasmannia auropunctata is known as one of the worst invasive ants in the World. A cytogenetic study was conducted on two native populations from southeastern Bahia, Brazil. The analysis of the chromosomes observed in mitotic metaphases was made by a combination of methods: Giemsa conventional staining, chromomycin A3 (CMA3) and 4-6-diamidino-2-phenylindole (DAPI) fluorochrome staining, and acridine orange banding. The workers have all the karyotype 2n=32, with ten pairs of metacentric and six pairs of acrocentric chromosomes. One chromosome arm of the pair ten was positive for CMA3 and acridine orange, suggesting the occurrence of a nucleolar organizing region. This region is an interesting marker because is very conservative and seems to constitute an interesting specific taxonomic character. The pericentromeric region of many chromosomes was stained with DAPI, evidencing the occurrence of AT bases rich heterochromatin. 相似文献

New Ceratopogonidae from the Early Cretaceous amber of Hammana (Central Lebanon) are studied. Five new species (Lebanoculicoides daheri sp. nov., Protoculicoides krzeminskii sp. nov., Archiaustroconops annae sp. nov., Archiaustroconops hammanensis sp. nov., and Archiaustroconops dominiakae sp. nov.) are characterized, described, illustrated, and compared with other Cretaceous taxa. The crystal structure of three head-to-head dimers (having two cholic acid or deoxycholic acid units) linked at carbon atoms C3 by aromatic or alkyl bridges is studied. An internal coordinates system is necessary for describing the relative orientation in the space of the two bile acid residues. Five angles (three torsion and two common ones) are necessary for defining the relative position of both steroid residues in space. Carbon atoms C3 (which always carries a α-hydroxy group in natural bile acids), and C10 and C13 (which always carry β-methyl groups) of each steroid residue are suitable for this purpose. Furthermore, the distance between each C3 carbon atoms of both steroid residues will allow one to locate the steroids in space. The three dimers selected provide a large range of values for these angles. The packing, hydrogen bond network, and location of guest in the three crystals are discussed. 相似文献
