The 2,3,4- (9) and 2,4,6-tribenzyl (19) ethers of 1-thio-β-d-galactopyranose were prepared from the corresponding O-benzylated normal (1-hydroxyl) sugars 4 and 15 via the sequence: normal sugar → diacetate → O-acetylglycosyl bromide → O-acetyl-glycosyl ethylxanthate → 1-thio sugar. 2,3,4-Tri-O-benzyl-α-d-galactopyranose (4) is most advantageously made from allyl 6-O-allyl-α-d-galactopyranoside (2) by a published synthesis. An improved synthesis of 2,4,6-tri-O-benzyl-d-galactopyranose (15) was devised; it involves the selective 3-O-benzoylation of allyl 2,6-di-O-benzyl-α-d-galactopyranoside (10).  相似文献

Summary The productivity of actinomycin D withStreptomyces parvullus could be enhanced with calcium alginate immobilized cells and a synthetic starvation medium in a continuous air-lift bioreactor with moderately low dilution rate. The intracellular localization of 3-methylcrotonyl-CoA carboxylase (MCase), an enzyme of the leucine oxidative pathway, was studied in bovine kidney. Differential centrifugation of kidney homogenates demonstrated that the majority of the enzyme was associated with the mitochondrial and cytosolic fractions. Isopycnic centrifugation of the mitochondrial fraction demonstrated cofractionation of MCase with mitochondrial markers, but not with lysosomal markers, consistent with a mitochondrial location for the enzyme. Using different homogenization techniques and comparing the fractional extraction of MCase and mitochondrial and cytosolic marker enzymes, the appearance of MCase in the “cytosolic” fraction was shown to be due to mitochondrial damage. The intramitochondrial distribution of MCase was determined using a digitonin procedure, and indicated that the enzyme is associated with the inner mitochondrial membrane. Although a fraction of MCase (30–40%) was “solubilized” by homogenization of whole mitochondria, the remaining MCase (60–70%) was tightly associated with the mitochondrial membrane fraction. Release and “solubilization” of this latter fraction was achieved by polyethylene glycol treatment. The “solubilized” MCase was stabilized in a glycerol-containing buffer.  相似文献

We established a method of KC transplantation by intraperitoneal (i.p.) injection using EGFP-expressing cells (EGFP-KCs) and normal KCs. The novel method is easier and less invasive than conventional methods so that it is not only technically advantageous but also ethically preferable for experiments using animals. We demonstrated that KCs migrated to the liver following i.p. Injection. Engraftment in the liver was not observed for peritoneal macrophages (pMPs). This suggests that KCs migrate to the liver via a sorting mechanism. KC injection decreased the KC number at 24 h and then recovered the KCs at 10 days to a normal level. Additionally, recovery to the normal level by KC injection was observed in mice with KC depletion induced by GdCl3. These results suggest that a regulatory mechanism exists for controlling the number of KCs.  相似文献

In the mammary gland of non-ruminant animals, glucose is utilized in a characteristic and unique way during lactation [11]. By measuring the incorporation of glucose carbon from [U-14C]glucose into intermediary metabolites and metabolic products in mammary epithelial cells from virgin, pregnant, and lactating mice, we demonstrate that glucose metabolite patterns can be used to recognize stages of differentiated function. For these cells, the rates of synthesis of glycogen and lactose, the ratio of lactate to alanine, and the ratio of citrate to malate are important parameters in identifying the degree of expression of differentiation. We further show that these patterns can be used as markers to determine the differentiated state of cultured mammary epithelial cells. Cells maintained on plastic substrates lose their distinctive glucose metabolite patterns while those on floating collagen gels do not. Cells isolated from pregnant mice and cultured on collagen gels have a pattern similar to that of their freshly isolated counterparts. When isolated from lactating mice, the metabolite patterns of cells cultured on collagen gels are different from that of the cells of origin, and resembles that of freshly isolated cells from pregnant mice. Our findings suggest that the floating collagen gels under the culture conditions used in these experiments provide an environment for the functional expression of the pregnant state, while additional factors are needed for the expression of the lactating state.  相似文献

Summary We have identified a major mouse milk protein as transferrin (Tf) using immunoprecipitation, 2-dimensional electrophoresis, Ouchterlony diffusion and V-8 protease digests. We show that Tf is synthesized by mammary epithelial cells themselves and that its synthesis and secretion is regulated distinctly from that of other milk proteins. In culture, the kinetics of Tf synthesis and secretion are distinct from that of β-casein; furthermore, Tf is relatively insensitive to lactogenic hormones whereas β-casein is hormone-dependent.In vivo, however, Tf is regulated by pregnancy. While the virgin gland produces small amounts of Tf, its production is greatly increased during pregnancy and lactation. Thus, Tf synthesis in the mammary gland is modulated by as yet unknown factorsin vivo. These observations are discussed in terms of Tf’s possible role in mammary gland growth, differentiation and function. This research was supported by the OHER office of U. S. An attempt was made to investigate which factor temperature or fungal infection, has a greater effect in accelerating physiological processes such as respiration and ethylene evolution in fruit. For this study the effect of infection with Trichothecium roseum, Alternaria alternata and Diplodia natalensis on banana fruit, at 14°C and 20°C was examined. The results show that the development of rot caused by different fungi and the physiological changes in the infected fruit may depend not only on the effect of temperature on fungi (as in vitro) but also on fruit ripening, or on processes connected with fruit ripening, which may accelerate fungal development.  相似文献

The potential for making functional interpretations from a single postcranial element for marsupials was investigated through morphometric analysis of the calcanea of 61 extant species from Australia and New Guinea. Extant species were grouped into locomotor categories and a canonical variates analysis was carried out on measurements of their calcanea. A relationship between measurements of the calcanea and the locomotor behavior of species was found, allowing for prediction of locomotor behavior from calcaneum morphometrics. This was applied to fossil marsupial taxa, from early–late Miocene/?Pliocene deposits at Riversleigh, in an attempt to determine their locomotor behavior. Hopping (saltatorial) taxa are distinguished from quadruped terrestrial taxa and taxa capable of climbing (arboreal and scansorial) by their relatively longer tuber calcis and wider calcaneal head, by their dorso-ventrally thicker calcaneal head, and by their calcaneocuboid facet being less steeply angled antero-posteriorly. Taxa capable of climbing are distinguished from quadruped terrestrial taxa by their shorter tuber calcis relative to the calcaneal head and by their smaller calcaneo-astragalar facet. The locomotor categories distinguished in this study (arboreal/scansorial, quadruped terrestrial, and saltatorial) highlight differences between species in their use of available substrates and thus are informative with regards to the structural components of their habitat. The results of this analysis can be used, in combination with other data, to make inferences about the habitats of paleocommunities at Riversleigh through the Miocene. The calcaneum is a dense and very robust element and, therefore, has a good chance of being preserved. This method provides a quick and easy way of inferring locomotion and has a wide potential for application to many fossil deposits because it requires only a single element.  相似文献

A simple method to analyze media consumption and by-product build-up in high-cell-density perfusion cultures has been developed. This technique makes use of media replenishment strategy which can be used to decouple the two phenomena.
