Eight swainsonine (SW)-degrading bacteria were isolated from the soil where locoweed was buried for 6 months and one of the
strains (YLZZ-1) was selected for further study. Based on morphology, physiologic tests, 16S rRNA gene sequence, and phylogenetic
characteristics, the strain showed the greatest similarity to members of the order Acinetobacters and within the order to members of the Acinetobacter calcoaceticus group. The ability of the strain for degrading SW, as sole carbon source, was investigated under different culture conditions.
The preferential temperature and initial pH for the strain were 25–35°C and 6–9, respectively. The optimal temperature for
the strain was 30°C and the optimal pH was 7.0. There was a positive correlation between degradation rate and inoculation
amount. The concentration of SW affected the degradation ability. When the concentration of SW was lower than 100 mg/l, SW
decreased immediately after incubation, and when the concentration of SW was 200 mg/l, there was an inhibiting effect for
bacteria growth and SW degradation. The strain could degrade SW completely within 14 h when the concentration of SW was 50 mg/l.
These results highlight the potential of this bacterium to be used in detoxifying of SW in livestock consuming locoweed. 相似文献

本文以缺刻缘绿藻(Parietochloris incisa)为实验材料,研究了其在四种不同氮、磷浓度下的生长、总脂(TL)及花生四烯酸(AA)含量的变化,分析测定了低氮和低磷浓度下脂肪酸的组成。结果表明:在BG-11培养基基础上,氮浓度的改变对缺刻缘绿藻的生物量影响不明显,不同氮浓度下的最终生物量均在2.1 g.L-1左右。在低氮浓度下TL和AA达到最大,分别为33.37%(%干重)和36.63%(%总脂肪酸,TFA),其中AA占藻体干重的11.56%。不同磷浓度对缺刻缘绿藻的生长有显著影响,最终生物量在0.90~2.07 g.L-1之间。TL在8~20 mg.L-1的磷浓度范围内没有明显变化,为28%左右。TL与氮、磷浓度呈显著负相关关系,且低氮有利于AA的积累。 相似文献

Respiratory virus infection results in considerable pulmonary immunopathology, a component of which results from the host immune responses. We have developed a murine model to specifically examine the lung injury due to CD8(+) T cell recognition of an influenza hemagglutinin (HA) transgene on lung epithelium in the absence of replicating virus, after adoptive transfer. Lung injury is largely mediated by chemokines expressed by the epithelial cells upon T cell recognition mediated by TNF-alpha. To determine the critical source of TNF-alpha, HA-specific TNF(-/-) CD8(+) T cells were transferred into HA transgenic animals, and lung injury was not observed, though these T cells exhibited no defect in antiviral activity in vivo. This indicates that the initiating event in the injury process is Ag-specific expression of TNF-alpha by antiviral CD8(+) T cells upon recognition of alveolar epithelial Ag, and that the effector activities responsible for viral clearance may be dissociable from those resulting in immunopathology. 相似文献

Human thrombopoietin (hTPO) is a heavily glycosylated protein with 6 and 24 potential N- and O-glycosylation sites, respectively. To determine the effect of sodium butyrate (NaBu) on the production and quality of hTPO in recombinant Chinese hamster ovary (rCHO) cells, NaBu (0-10 mM) was added to the cultures of exponentially growing cells. NaBu addition significantly increased both the specific and volumetric hTPO production, although it decreased the cell viability by apoptosis in a dose-dependent manner. The highest hTPO concentration of 82.2 +/- 5.6 microgml-1 was obtained in the culture with 3 mM NaBu addition. Compared with the culture without NaBu addition, the culture with 3 mM NaBu resulted in a 6.4-fold increase in qTPO and a 3.3-fold increase in the final hTPO concentration on day 7. However, NaBu deteriorated the quality of hTPO, resulting from increased heterogeneity, reduced acidic hTPO isoforms, reduced alpha(2 --> 3) sialylation, and decreased in vivo biological activity. We also found that the biological activity of hTPO in the culture with 3 mM NaBu addition collected on day 7 was 72% of that in the culture without NaBu addition. Taken together, the use of NaBu or its optimal concentration for high-level expression of a heavily glycosylated protein like hTPO should be determined by considering its detrimental effect on the quality of glycoprotein. 相似文献

Gutless adenoviruses (GAds), namely, all gene-deleted adenoviruses, were developed to minimize their immune responses and toxic effects for a successful gene delivery tool in gene therapy. The Cre/loxP system has been widely used for GAd production. To produce GAd with a low amount of helper adenovirus (HAd) as byproduct, it is indispensable to use 293Cre cells expressing a high level of Cre for GAd production. In this study, we constructed the HAd containing enhanced green fluorescent protein gene flanked by two parallel loxP sites (HAd/GFP). The use of HAd/GFP with flow cytometry allows one to select 293Cre cells expressing a high level of Cre without using conventional Western blot analysis. Unlike conventional HAd titration methods such as plaque assay and end-point dilution assay, it also allows one to monitor rapidly the HAd as byproduct in earlier stages of GAd amplification. Paragonimus westermani is a common fluke in Korea. The present study aimed to determine serum total IgE and specific IgG levels in experimental paragonimiasis of rats. Each Wistar rat was inoculated orally with 20-25 metacercariae of P. westermani from Cambaroides similis. Before and after infection (1, 2, 3, 4, 6, 8 weeks) of P. westermani, the blood was collected from the retro-orbital venous plexus of rats and kept serum at -70 degrees C. Serum total IgE and specific IgG levels were determined by the capture and conventional enzyme-linked immunosorbent assay, respectively. The results were as follows; 1. Serum IgE values were increased to 0.18 +/- 0.042 at 2 weeks, 0.28 +/- 0.151 at 4 weeks and 0.43 +/- 0.055 at 8 weeks after infection. The absorbances of non-infected rats ranged 0.07 +/- 0.021-0.12 +/- 0.025. 2. Specific IgG values were slightly increased at 3 weeks (0.20 +/- 0.032) and gradually increased up to 8 weeks (0.31 +/- 0.067) after infection. The absorbances of non-infected rats ranged 0.11 +/- 0.035-0.18 +/- 0.019. The present results suggested that P. westermani could elevate serum IgE and specific IgG antibodies in Wistar rats which were not a good definitive host. 相似文献
