Tufts of long delicate bristles were detected on mature colonies of Pediastrum boryanum (Turp.) Meneghini. The bristles are not uniform in length and can exceed 100 μm. Bristle number per tuft can exceed eighty. Ultrastructure studies revealed that tufts are an aggregate of hexagonal tubules arranged in staggered rows. Each hexagonal tubule has an inner diameter of 6 nm and is made up of six interconnected subunits 4 nm in diameter that are shared by adjoining tubules. The bristles can be easily removed from mature colonies with a vortex stirrer. Removal of bristles from mature colonies in culture results in the loss of buoyancy and subsequent settling to the bottom of the flask.  相似文献

A di-(carboxamidomethyl) derivative of molybdopterin, the organic component of the molybdenum cofactor, has been prepared under conditions favoring retention of all of the structural features of the molecule. The specific radioactivity of [1-14C]iodoacetamide incorporated relative to the amount of phosphate indicated two alkylation sites per pterin. Energy-dispersive x-ray analysis of the derivative showed the presence of 2 sulfurs in the derivative. An exact mass corresponding to the molecular formula C14H18N7O5S2 was obtained for the MH+ ion of the alkylated, dephosphorylated compound by fast atom bombardment mass spectroscopy. 1H NMR spectra of the phosphorylated and dephosphorylated forms of alkylated molybdopterin, in conjunction with the other data, have provided strong corroboration of the validity of the proposed structure of molybdopterin (Johnson, J. L., and Rajagopalan, K. V. (1982) Proc. Natl. Acad. Sci. U. S. A. 79, 6856-6860) as a 6-alkylpterin with a 4-carbon side chain containing an enedithiol on C-1' and C-2', a secondary alcohol on C-3', and a phosphorylated primary alcohol on C-4'. As isolated, the di-(carboxamido-methyl)molybdopterin was found to be a 5,6,7,8-tetrahydropterin.  相似文献

Tendrils on long shoots of Parthenocissus inserta occur in a regular pattern opposite the alternate distichous leaves at two successive nodes of each three nodes. Ontogenetic study shows that the tendril is initiated at the flank of the shoot apex during the second plastochron in an essentially axillary position. It is carried upward with growth of the internode above the axillant leaf and ultimately is situated opposite the next younger leaf. In a rhythmic pattern a different group of appendages is produced by the shoot apex at each node in the sequence of three. In acropetal order these are: at the tendrilless node the leaf subtends an axillary bud complex which in turn subtends a tendril; the leaf at the lower tendril-bearing node directly subtends a tendril, and the leaf at the upper tendril-bearing node subtends an axillary bud. Tendril primordia were not induced to develop as foliaceous shoots when cultured in vitro or in decapitation experiments, indicating that the meristem which becomes a tendril is determined early in its inception. Although built on a shoot pattern, the tendril is regarded as an organ sui generis with a possible relationship to the inflorescence. The morphological nature of the tendril is discussed in the light of theories postulated in the literature.  相似文献

This report describes the application of high-performance liquid chromatography combined with continuous-flow fast atom bombardment mass spectrometry to analytical problems in the biomedical laboratory. Applications include the compound-specific detection of diagnostic acylcarnitines in human urine, the separation and analysis of acyl-coenzyme A thioesters, and qualitative studies on complex mixtures of modified peptides (dansyl and dinitrophenyl derivatives). For each of these applications standard analytical columns (3.9 mm I.D.) and 1 ml/min flow-rates were employed with post-column stream splitting (1:100) before mass spectrometry. Various mobile phase compositions and solvent gradients were employed. The addition of 1–5% glycerol to the mobile phase was shown to have little effect on the chromatography. For all compounds studied (acylcarnitines, acyl-coenzyme A thioesters, and derivatized peptides) molecular weight information was obtained and sufficient sensitivity was achieved to allow unambiguous identification of trace components in complex mixtures.  相似文献

Summary The scanning electron microscope has been used to investigate the surfaces of epidermis, hair follicles, fat cells, glands and blood vessels in sections of human skin. The appearances of structures associated with these surfaces have been described. The results demonstrate that this new instrument can become an important tool in the investigation of surface changes that might accompany the application of loads (extensive, compressive or torsional), or physiological or pathological abnormalities.We wish to thank Prof. R. Chemiluminescence (CL) with maximum emission in the range 550–650 nm is observed when proteins and certain amino acids are heated in air, and CL intensity is significantly reduced in nitrogen. Of the 20 common amino acids, lysine (Lys) has the highest thermal CL intensity by a factor of ~30 over arginine, threonine and asparagine. This finding differs from previous studies on amino acids and proteins oxidised using free radical initiators or singlet oxygen, where tryptophan was the dominant factor for CL emission. CL from heating solid Lys in air is accompanied by browning and the generation of fluorescent products which are characteristic of advanced glycosylation end products (AGEs) in thermally treated milk proteins. During thermal oxidation, Lys may react with its own carbonyl oxidation products to form fluorescent compounds similar to AGEs via the formation of Schiff bases. The mechanism of thermal oxidation of proteins may be similar to polyamide polymers, where reaction of free primary amino groups with carbonyls to form Schiff bases plays a key role.  相似文献

The cyanobacteria Phormidium valderianum, P. tenue and Microcoleus chthonoplastes and the green algae Rhizoclonium fontinale, Ulva intestinalis, Chara zeylanica and Pithophora oedogoniana were exposed to hydrogen tetrachloroaurate solution and were screened for their suitability for producing nano‐gold. All three cyanobacteria genera and two of the green algae (Rhizoclonium fontinale and Ulva intestinalis) produced gold nanoparticles intracellularly, confirmed by purple colouration of the thallus within 72?h of treatment at 20°C. Extracted nanoparticle solutions were examined by UV‐vis spectroscopy, transmission electron microscopy (TEM) and X‐ray diffractometry (XRD). XRD confirmed the reduction of Au (III) to Au (0). UV‐vis spectroscopy and TEM studies indicated the production of nanoparticles having different shapes and sizes. Phormidium valderianum synthesized mostly spherical nanoparticles, along with hexagonal and triangular nanoparticles, at basic and neutral pHs (pH 9 and pH 7, respectively). Medicinally important gold nanorods were synthesized (together with gold nanospheres) only by P. valderianum at acidic pH (pH 5); this was initially determined by two surface plasmon bands in UV‐vis spectroscopy and later confirmed by TEM. Spherical to somewhat irregular particles were produced by P. tenue and Ulva intestinalis (TEM studies). The UV‐vis spectroscopy of the supernatant of other algal extracts indicated the formation of mostly spherical particles. Production of gold nanoparticles by algae is more ecofriendly than purely chemical synthesis. However, the choice of algae is important: Chara zeylanica and Pithophora oedogoniana were found to be unable to produce nanoparticles.  相似文献
