Epidemiological studies have demonstrated that hormonereplacement therapy with estrogen (E2) or E2plus progesterone in postmenopausal women decreases the age-associatedrisk of cardiovascular disease by 30-50%. Treatment of vascularsmooth muscle (VSM) cells with physiological concentrations ofE2 has been shown to inhibit growth factor-stimulated cellproliferation. In this study, we tested the hypothesis thatE2 inhibits the age-associated increase in VSM cellproliferation by inhibiting nuclear factor (NF)-B pathway. Weinvestigated the effects of E2 treatment andadenovirus-mediated estrogen receptor (ER)- gene transfer on cellproliferation and NF-B activation using VSM cells cultured from3-mo-old and 24-mo-old Fischer 344 female rats. Our results demonstratethat VSM cell proliferation was significantly increased(P < 0.05) in aged compared with young adult femalerats. Treatment of VSM cells with physiological concentrations ofE2 inhibited VSM cell proliferation, and this inhibitionwas significantly greater (P < 0.05) in cells from aged female rats compared with young adults. The inhibitory effects ofE2 on cell proliferation in aged female rats weresignificantly potentiated by overexpression of the human ER- geneinto VSM cells. Constitutive and interleukin (IL)-1-stimulatedNF-B activation was significantly greater (P < 0.05) in VSM cells from aged compared with young female rats.E2 treatment of VSM cells from aged female rats inhibitedboth constitutive and IL-1-stimulated NF-B activation. ER-gene transfer into VSM cells from aged female rats further augmentedthe inhibitory effects of E2. In conclusion, our data demonstrate that constitutive and IL-1-stimulated NF-B activation is increased in VSM cells from aged female rats due to loss of E2 and this can be restored back to normal levels by ER-gene transfer and E2 treatment. In addition, increasedNF-B signaling may be responsible for increased incidence ofcardiovascular disease in postmenopausal females.

The inter-simple-sequence repeat (ISSR) primers that anneal to a simple repeat of various length and at non-repetitive motifs at 3 and 5 end were attempted for PCR amplification of Leptospira genome. Of the six ISSR primers tested, namely, (AG)8T, (AG)8C, (AG)8G, (CA)8A, (TG)8C and (TG)8G, only primer (AG)8T produced amplification of 1000 bp in the two non-pathogenic Leptospira species tested, viz; Leptospira biflexa serovar patoc and L. meyeri serovar ranarum, with no amplification in any of the 16 standard pathogenic serovars tested. The remaining five ISSR primers did not exhibit any amplification of the Leptospira genome in either pathogenic or non-pathogenic species. From among 35 Leptospira isolates recovered from hospitalized patients with pyrexia of unknown origin and/or febrile jaundice (12 in number) and from different environmental water sources (23 in number), (AG)8T ISSR-PCR correctly identified all the 22 isolates from water sources that were confirmed to be non-pathogenic by conventional tests. The results therefore, confirmed the ability of a primer, based on simple-sequence repeat motif, to produce a fragment that is useful as a group genetic marker in Leptospira species. The single nucleotide anchor, T, at the 3 end of the primer appeared to play an important role in differentiation of pathogenic and non-pathogenic species of Leptospira. Multiplex PCR, using ISSR primer, (AG)8T and the reported 16S rRNA gene primers, specific for pathogenic Leptospira species, or the 23S rRNA Leptospira genus specific primers, provided clear identification of serovars and isolates into pathogenic or non-pathogenic groups.  相似文献

Generation of H2O2 by rat brain mitochondria using succinate and glycerol-1-phosphate as substrates has been demonstrated. Earlier workers were unable to detect this activity in sucrose-Tris buffer. We found that this was due to a lag in the expression of activity in sucrose medium. Using phosphate buffer (50 mM), good rates are now obtained. Generation of H2O2 by rat brain mitochondria required the presence of antimycin A and was dependent on the substrates succinate and glycerol-1-phosphate. Low rates were obtained with NAD+-linked substrates and none with choline, glutamate, and NADH. The Km and Vmax values for H2O2 generation were considerably lower than the corresponding values for the respective dehydrogenase activity, measured by dye reduction. Oxygen-radical scavengers inhibited H2O2 generation, suggesting oxygen radical involvement. Depletion of ubiquinone from mitochondria resulted in loss of H2O2 generation. Reconstitution of such depleted particles with ubiquinone restored the capacity to generate H2O2 in a concentration-dependent manner. Levels of H2O2 production were found to be maximal in cerebellum. Brain mitochondria from rabbit, hamster, mouse, and guinea pig also have the capacity to generate H2O2 on oxidation of glycerol-1-phosphate.  相似文献

Two near shore small cetaceans occur commonly along the Maharashtra coast, the Indian Ocean humpback dolphin and Indo Pacific finless porpoise. These cetaceans frequently interact with fisheries in this region due to overlap in space and resource use. Besides stranding records, little ecological information is available about these species from Maharashtra. We conducted 143 semistructured interviews to document local ecological knowledge and community perceptions of small cetaceans in 30 coastal fishing villages in Sindhudurg. Perceptions of finless porpoises were largely neutral, whereas humpback dolphins were negative. A classification regression tree (CART) analysis (root node error: 60%) showed that the annual cost of gear damage was an important predictor variable of humpback dolphin perceptions, followed by occupation (gear type) and age. Entanglements were reported for both species in large and small gill nets, and shore seines. Perceived net damage and catch loss due to humpback dolphins was six times greater than that of finless porpoises. However, finless porpoises were reportedly more frequently entangled in gear than humpback dolphins. We provide an insight into the perceptions of cetaceans in the local community and the fisheries‐cetacean interactions that shape them.  相似文献

Pseudomonas maltophilia CSV89, a bacterium isolated from soil in our laboratory, grows on 1-naphthoic acid as the sole source of carbon and energy. To elucidate the pathway for degradation of 1-naphthoic acid, the metabolites were isolated from spent medium, purified by TLC, and characterized by gas chromatography-mass spectrometry. The involvement of various metabolites as intermediates in the pathway was established by demonstrating relevant enzyme activities in cell-free extracts, oxygen uptake and transformation of metabolites by the whole cells. The results obtained from such studies suggest that the degradation of 1-naphthoic acid is initiated by double hydroxylation of the aromatic ring adjacent to the one bearing the carboxyl group, resulting in the formation of 1,2-dihydroxy-8-carboxynaphthalene. The resultant diol was oxidized via 3-formyl salicylate, 2-hydroxyisophthalate, salicylate and catechol to TCA cycle intermediates.  相似文献

Fungal infection represents a severe problem that decreases the yield and market value of fruit crops. The use of fungicides is a conventional method to control infections but it is associated with disadvantages, such as hazardous impact on public health, environmental contamination, resistance development among pathogens and high cost of agrochemicals. Biological control is an alternative approach for the treatment of fungal infections. The species of Bacillus, Pseudomonas, Enterobacter, Pantoea, Burkholderia, Lysobacter and Serratia have been successfully used in the control of fungal infections. The mechanisms involved in biocontrol are hyperparasitism or predation, production of antibiotics, lytic enzymes and induction of host resistance. Lactic acid bacteria have been used as biopreservative organisms in food and feed systems. They are a cluster of Gram-positive bacteria and include species of the genera Enterococcus, Lactobacillus, Leuconostoc, Lactococcus and Pediococcus. The ability to produce several antibacterial and antifungal substances confers biopreservation potential to lactic acid bacteria. Many have ‘generally regarded as safe’ status and are considered as safe from both human and environmental points of view. Their isolation is reported from vegetables, aerial plant surfaces, pickled cabbage, grass silage, malted cereals and also from soil. They produce antifungal substances, such as cyclic dipeptides, proteinaceous compounds, organic acids, fatty acids and reuterin. The biocontrol potential of lactic acid bacteria is demonstrated in the prevention of fungal infections of fruits, such as apples and grapes. Thus, living cells or product formulations of antifungal lactic acid bacteria may be prepared and used as an alternative biocontrol technology.  相似文献

Crop raiding by wild herbivores close to an area of protected wildlife is a serious problem that can potentially undermine conservation efforts. Since there is orders of magnitude difference between farmers’ perception of damage and the compensation given by the government, an objective and realistic estimate of damage was found essential. We employed four different approaches to estimate the extent of and patterns in crop damage by wild herbivores along the western boundary of Tadoba-Andhari Tiger Reserve in the state of Maharashtra, central India. These approaches highlight different aspects of the problem but converge on an estimated damage of over 50% for the fields adjacent to the forest, gradually reducing in intensity with distance. We found that the visual damage assessment method currently employed by the government for paying compensation to farmers was uncorrelated to and grossly underestimated actual damage. The findings necessitate a radical rethinking of policies to assess, mitigate as well as compensate for crop damage caused by protected wildlife species.  相似文献
