Abstract. A phenological map has been prepared for the Podyjí/Thayatal National Park, located on the border between the Czech Republic and Austria. The area is characterized by V-shaped river valleys deeply incised into a gently undulating landscape. Five stages of phenological development were defined, based upon phenological spectra of plant communities in late April; 96 sites, more or less regularly spaced over the area, were assigned to one of these stages using field observations of these spectra. Potential direct solar irradiation was calculated from a digital elevation model for a 25 m x 25 m grid extending over the study area. A method was developed for interpolation of phenological observations by weighted regression of phenology on the irradiation model. This interpolation enables the prediction of the local stage of phenological development across the study area without levelling out phenological patterns dependent upon small-scale topographic variation. Spring phenological events appear to be more advanced in the valleys (except for steep north-facing slopes) than in the adjacent landscape. Possible climatic processes underlying this pattern are discussed with emphasis on temperature inversions in the river valleys. Cold-air-drainage inversions reported from the valleys are probably too infrequent and of too short duration to delay plant phenology. The combined effects of advanced phenology and the increased risk of spring frost injury due to these inversions in the valleys may be an explanation for local distributional patterns of flora and vegetation.  相似文献

We have previously suggested that ozone(O3)-induced pain-relatedsymptoms and inhibition of maximal inspiration are due to stimulationof airway C fibers (M. J. Hazucha, D. V. Bates, and P. A. Bromberg.J. Appl.Physiol. 67: 1535-1541, 1989). If this were so,pain suppression or inhibition by opioid-receptor agonists shouldpartially or fully reverseO3-induced symptomatic and lung functional responses. The objectives of this study were to determine whether O3-induced pain limitsmaximal inspiration and whether endogenous opioids contribute tomodulation of the effects of inhaledO3 on lung function. Theparticipants in this double-blind crossover study were healthyvolunteers (18-59 yr) known to be "weak" (WR;n = 20) and "strong"O3 responders (SR;n = 42). They underwent either two 2-hexposures to air or two 2-h exposures to 0.42 parts/millionO3 with moderate intermittentexercise. Immediately afterpost-O3 spirometry, the WR wererandomly given either naloxone (0.15 mg/kg iv) or saline, whereas SRrandomly received either sufentanil (0.2 µg/kg iv) or saline.O3 exposure significantly(P < 0.001) impaired lung function.In SR, sufentanil rapidly, although not completely, reversed both thechest pain and spirometric effects (forced expiratory volume in 1 s;P < 0.0001) compared with saline.Immediate postexposure administration of saline or naloxone had nosignificant effect on WR. Plasma -endorphin levels were not relatedto an individual's O3responsiveness. Cutaneous pain variables showed a nonsignificantweak association with O3responsiveness. These observations demonstrate that nociceptive mechanisms play a key role in modulatingO3-induced inhibition ofinspiration but not in causing lack of spirometric response toO3 exposure in WR.

The first objective of this computational study was to assess the strain magnitude and distribution within the three-dimensional (3D) trabecular bone structure around an osseointegrated dental implant loaded axially. The second objective was to investigate the relative micromotions between the implant and the surrounding bone. The work hypothesis adopted was that these virtual measurements would be a useful indicator of bone adaptation (resorption, homeostasis, formation).In order to reach these objectives, a μCT-based finite element model of an oral implant implanted into a Berkshire pig mandible was developed along with a robust software methodology. The finite element mesh of the 3D trabecular bone architecture was generated from the segmentation of μCT scans. The implant was meshed independently from its CAD file obtained from the manufacturer. The meshes of the implant and the bone sample were registered together in an integrated software environment. A series of non-linear contact finite element (FE) analyses considering an axial load applied to the top of the implant in combination with three sets of mechanical properties for the trabecular bone tissue was devised. Complex strain distribution patterns are reported and discussed. It was found that considering the Young’s modulus of the trabecular bone tissue to be 5, 10 and 15 GPa resulted in maximum peri-implant bone microstrains of about 3000, 2100 and 1400. These results indicate that, for the three sets of mechanical properties considered, the magnitude of maximum strain lies within an homeostatic range known to be sufficient to maintain/form bone. The corresponding micro-motions of the implant with respect to the bone microstructure were shown to be sufficiently low to prevent fibrous tissue formation and to favour long-term osseointegration.  相似文献

Stress induces the release of the peptide corticotropin-releasing factor (CRF) into the ventral tegmental area (VTA), and also increases dopamine (DA) levels in brain regions receiving dense VTA input. Since the role of stress in drug addiction is well established, the present study examined the possible involvement of CRF1 receptor in the interaction between morphine withdrawal and catecholaminergic pathways in the reward system. The effects of naloxone-precipitated morphine withdrawal on signs of withdrawal, hypothalamo-pituitary-adrenocortical (HPA) axis activity, dopamine (DA) and noradrenaline (NA) turnover in the nucleus accumbens (NAc) and activation of VTA dopaminergic neurons, were investigated in rats pretreated with vehicle or CP-154,526 (selective CRF1R antagonist). CP-154,526 attenuated the increases in body weight loss and suppressed some of withdrawal signs. Pretreatment with CRF1 receptor antagonist resulted in no significant modification of the increased NA turnover at NAc or plasma corticosterone levels that were seen during morphine withdrawal. However, blockade of CRF1 receptor significantly reduced morphine withdrawal-induced increases in plasma adrenocorticotropin (ACTH) levels, DA turnover and TH phosphorylation at Ser40 in the NAc. In addition, CP-154,526 reduced the number of TH containing neurons expressing c-Fos in the VTA after naloxone-precipitated morphine withdrawal. Altogether, these results support the idea that VTA dopaminergic neurons are activated in response to naloxone-precipitated morphine withdrawal and suggest that CRF1 receptors are involved in the activation of dopaminergic pathways which project to NAc.  相似文献

Metallothioneins (MTs) are a class of ubiquitously occurring low molecular weight cysteine- and metal-rich proteins containing sulfur-based metal clusters. The conservation of these clusters in an increasing number of three-dimensional structures of invertebrate, vertebrate and bacterial MTs signifies the importance of this structural motif. In the postgenomic era, it is becoming increasingly clear that MTs fulfil different functions. Increasing body of evidence show that diverse functions of the mammalian MT-1/MT-2 isoforms including involvement in zinc homeostasis, protection against heavy metal toxicity and oxidative damage are related to their clusters. In contrast, the biological properties of the brain-specific MT-3 isoform imply that the clusters in this protein play a structural role. The recent highlights of MT research are the subject of this review.  相似文献

In a typical cell culture system, growth factors immobilized on the cell culture surfaces can serve as a reservoir of bio-signaling molecules, without the need to supplement them additionally into the culture medium. In this paper, we report on the fabrication of albumin/heparin (Alb/Hep) assemblies for controlled binding of basic fibroblast growth factor (FGF-2). The surfaces were constructed by layer-by-layer adsorption of polyelectrolytes albumin and heparin and were subsequently stabilized by covalent crosslinking with glutaraldehyde. An analysis of the surface morphology by atomic force microscopy showed that two Alb/Hep bilayers are required to cover the surface of substrate. The formation of the Alb/Hep assemblies was monitored by the surface plasmon resonance (SPR), the infrared multiinternal reflection spectroscopy (FTIR MIRS) and UV/VIS spectroscopy. The adsorption of FGF-2 on the cross-linked Alb/Hep was followed by SPR. The results revealed that FGF-2 binds to the Alb/Hep assembly in a dose and time-dependent manner up to the surface concentration of 120 ng/cm2. The bioactivity of the adsorbed FGF-2 was assessed in experiments in vitro, using calf pulmonary arterial endothelial cells (CPAE). CPAE cells could attach and proliferate on Alb/Hep surfaces. The adsorbed FGF-2 was bioactive and stimulated both the proliferation and the differentiation of CPAE cells. The improvement was more pronounced at a lower FGF-2 surface concentration (30 ng/cm2) than on surfaces with a higher concentration of FGF-2 (120 ng/cm2).  相似文献

Shiga toxin (Stx) is the main virulence factor of enterohemorrhagic Escherichia coli, which are non-invasive strains that can lead to hemolytic uremic syndrome (HUS), associated with renal failure and death. Although bacteremia does not occur, bacterial virulence factors gain access to the circulation and are thereafter presumed to cause target organ damage. Stx was previously shown to circulate bound to blood cells but the mechanism by which it would potentially transfer to target organ cells has not been elucidated. Here we show that blood cell-derived microvesicles, shed during HUS, contain Stx and are found within patient renal cortical cells. The finding was reproduced in mice infected with Stx-producing Escherichia coli exhibiting Stx-containing blood cell-derived microvesicles in the circulation that reached the kidney where they were transferred into glomerular and peritubular capillary endothelial cells and further through their basement membranes followed by podocytes and tubular epithelial cells, respectively. In vitro studies demonstrated that blood cell-derived microvesicles containing Stx undergo endocytosis in glomerular endothelial cells leading to cell death secondary to inhibited protein synthesis. This study demonstrates a novel virulence mechanism whereby bacterial toxin is transferred within host blood cell-derived microvesicles in which it may evade the host immune system.  相似文献

Adenosine A3 receptor knockout (A3AR KO) mice and their wild-type (WT) counterparts were compared from the point of view of their abilities to survive exposures to lethal doses of γ-radiation belonging to the range of radiation doses inducing the bone marrow acute radiation syndrome. Parameters of cumulative 30-day survival (experiment using a midlethal radiation dose) or cumulative 11-day survival (experiment using an absolutely lethal radiation dose), and of mean survival time were evaluated. The values of A3AR KO mice always reflected their higher survival in comparison with WT ones, the P values being above the limit for statistical significance after the midlethal radiation dose and standing for statistical significance after the absolutely lethal radiation dose. This finding was considered surprising, taking into account the previously obtained findings on defects in numbers and functional properties of peripheral blood cells in A3AR KO mice. Therefore, previous hematological analyses of A3AR KO mice were supplemented in the present studies with determination of serum levels of the granulocyte colony-stimulating factor, erythropoietin, and thrombopoietin. Though distinct differences in these parameters were observed between A3AR KO and WT mice, none of them could explain the relatively high postirradiation survival of A3AR KO mice. Further studies on these mice comprising also those on other than hemopoietic tissues and organs can help to clarify their relative radioresistance.  相似文献

Measuring Biomass and Carbon Stock in Resprouting Woody Plants   总被引:1,自引:0,他引:1  
Resprouting multi-stemmed woody plants form an important component of the woody vegetation in many ecosystems, but a clear methodology for reliable measurement of their size and quick, non-destructive estimation of their woody biomass and carbon stock is lacking. Our goal was to find a minimum number of sprouts, i.e., the most easily obtainable, and sprout parameters that should be measured for accurate sprout biomass and carbon stock estimates. Using data for 5 common temperate woody species, we modelled carbon stock and sprout biomass as a function of an increasing number of sprouts in an interaction with different sprout parameters. The mean basal diameter of only two to five of the thickest sprouts and the basal diameter and DBH of the thickest sprouts per stump proved to be accurate estimators for the total sprout biomass of the individual resprouters and the populations of resprouters, respectively. Carbon stock estimates were strongly correlated with biomass estimates, but relative carbon content varied among species. Our study demonstrated that the size of the resprouters can be easily measured, and their biomass and carbon stock estimated; therefore, resprouters can be simply incorporated into studies of woody vegetation.  相似文献
