Composition of native vegetation on a polychlorinated biphenyls (PCB)-contaminated soil dumpsite at Lhenice, South Bohemia (Czech Republic), was determined and species variability in the accumulation of PCBs in plant biomass was investigated. Soil stripping contaminated by PCBs originated at a factory producing electrical transformers that mostly used the commercial PCB mixture Delor 103 and 106. The PCB content of soil in the most contaminated part of the dumpsite reached 153 mg kg(-1) dry soil. Low diversity of plant species was found on the dumpsite. Results showed three grass species, Festuca arundinacea Schreb., Phalaroides arundinacea (L.) Rauschert., and Calamagrostis epigeios (L.) Roth., to be the major components of the vegetation and confirmed their high tolerance toward PCB contamination. The highest content of PCB in plant biomass--813.2 microg kg(-1) dry biomass--was determined in Festuca aboveground biomass. For phytoextraction purposes especially, Festuca can be recommended due to its high biomass yield, but its bioconcentration factor was very low (0.006). Tripleurospermum maritimum (L.) Sch. Bip. and Cirsium arvense (L.) Scop. grew mainly at the margins of the most contaminated part of the dumpsite. The PCB content determined in their aboveground biomass-278.7 and 289.5 microg kg(-1) dry biomass, respectively--was nonsignificantly lower compared to grass species Phalaroides and Calamagrostis. Polyelectrolyte multilayer sensor capsules, 5 microm in diameter, which contained fluorescein-labeled poly(acrylic acid) (PAAAF) as pH-sensitive reporter molecules, were fabricated and employed to explore their endocytotic uptake into HEK 293T cells by flow cytometry. The percentage of capsules residing in the endolysosomal compartment was estimated from the fluorescence intensity decrease caused by acidification. Capsules attached to the extracellular surface of the plasma membrane were identified by trypan blue quenching. The number of capsules in the cytoplasm was rather small, being below the detection limit of the method. The advantages of polyelectrolyte multilayer capsules are that the fluorophore is protected from interaction with cellular compartments and that the multilayer can be equipped with additional functions. 相似文献

The purpose of the present study was to elucidate signaling pathways by which insulin like-growth factor 1 (IGF1) promotes FSH-stimulated synthesis and retention of hyaluronic acid (HA) in pig oocyte-cumulus complexes (OCCs) cultured in serum-free medium. We found that IGF1 had no effects on FSH-stimulated production of cAMP and activation of protein kinase A in the OCCs. Immunoblotting with phospho-specific antibodies showed that FSH moderately phosphorylated v-akt murine thymoma viral oncogene homolog (AKT) and mitogen-activated kinase 3 and 1 (MAPK3/1) in cumulus cells. The exposure of OCCs to both FSH and IGF1 resulted in a significant (P < 0.05) increase in AKT and MAPK3/1 phosphorylation. An inhibitor of phosphoinositide-3-kinase (PIK3), LY 294002, significantly (P < 0.05) reduced the IGF1-enhanced phosphorylation of AKT, and inhibitors of AKT (SH6) and MAPK3/1 (U0126) significantly (P < 0.05) decreased the synthesis and retention of HA stimulated by concomitant exposure of OCCs to both FSH and IGF1. The IGF1-promoted synthesis of HA was not accompanied by an increase in the relative abundance of hyaluronan synthase 2 (HAS2) mRNA in the cumulus cells. We conclude that IGF1 promotes the FSH-stimulated synthesis and retention of HA in pig OCCs by PIK3/AKT- and MAPK3/1-dependent mechanisms. 相似文献

The large‐scale spatial distribution of fish was investigated within a morphometrically simple canyon‐shaped reservoir with a single major tributary and a longitudinal trophic gradient (Římov Reservoir, Czech Republic). Samples of fish were taken by Nordic survey gill nets (several mesh sizes from 8 to 70 mm knot to knot) installed as surface nets at several offshore areas located along the longitudinal axis of the reservoir. Surveys were carried out in late summer during 1999–2003. An obvious distribution gradient of fish was revealed along the longitudinal axis of the Římov Reservoir. The total relative fish abundance and biomass (catch per unit effort) decreased considerably from the upstream end of the reservoir toward the dam. Roach (Rutilus rutilus), bleak (Alburnus alburnus) and bream (Abramis brama) comprised the bulk of catches at all areas. Enhanced dominance of bream was observed in the fish assemblage at the uppermost, more eutrophic area of the reservoir. The highest number of fish species and the highest abundance of young‐of‐the‐year fish were also observed in the tributary area. In the downstream part of the reservoir, gill net surveys along the vertical depth profiles indicated that offshore fish occupied mostly the epilimnion. The effects of aphidicolin, a specific inhibitor of DNA polymerase α, on cell growth, DNA synthesis and myogenic differentiation in the human alveolar rhabdomyosarcoma cell line KFR were studied. The treatment with aphidicolin at 5 × 10−6 M concentration, which completely inhibited DNA synthesis and cell growth, induced morphological differentiation of small mononuclear cells to elongated, multinucleated (myotube-like) structures. The morphological differentiation was accompanied by the expression of skeletal muscle myosin; about 30% myosin-positive cells were observed after 14 days of treatment, compared to 2.3% in untreated cultures. The results showed that aphidicolin induces differentiation of human rhabdomyosarcoma cells and that multinucleated myotube-like elements may develop simply by cell fusion without cell division and DNA synthesis. 相似文献

The genetic analysis of 5 tomato hybrids (Danubius F1, Luna F1, Lido F1, Balkan F1 and Mi-10 F1) was made. We produced their F1, F2, BC1 and BC2 generations and analysed their yield (on the first three flower branches) as well as some of the yield components of tomato
fruits (mean fruit weight, mean fruit weight on the first flower branch, fruit length, fruit width, and number of locules).
In order to estimate the gene effects, we applied the additive-dominance mode with three and six parameters. Epistatic gene
effects were estimated by applying the six-parameter mode (Mather and Jinks 1982). As for yield and yield components, there
were significant differences between the mean values of parents and their progeny. On the basis of the investigated genetic
parameters, the obtained results suggested that the additive and dominance gene effects prevailed in the yield and yield components
(Danubius F1, Luna F1, Lido F1, Mi-10 F1), whereas epistatic gene effects were excluded. As for the hybrid Balkan F1, we recorded significant gene effects, both the additive and the dominance ones in the yield inheritance: additive x additive
and dominance x dominance (with the negative sign). The estimated values of the epistatic gene effects were the most prominent
in inheriting the feature average fruit weight on the first flower branch — additive x dominance gene effects. They represented
the most frequent type of the interallele interaction recorded in the investigated hybrids. 相似文献
