In germinating grains of barley, Hordeum vulgare L. cv. Himalaya, free proline accumulated in the starchy endosperm during the period of rapid mobilization of reserve proteins. When starchy endosperms were separated from germinating grains and homogenized in a dilute buffer of pH 5 (the pH of the starchy endosperm), the liberation of proline continued in these suspensions. The process was completely inhibited by diisopropylfluorophosphate, indicating that it was totally dependent on serine carboxy-peptidases. The carboxypeptidases present in the starchy endosperms of germinating grains were fractionated by chromatography on DEAE-cellulose. Four peaks were obtained, all with different activity spectra on the seven carbobenzoxydipeptides (Z-dipeptides) tested. Two of the peaks corresponded to previously known barley carboxypeptidases; these as well as a third peak hydrolyzed substrates of the types Z-X-Y and Z-X-Pro (X and Y denote any amino acid residue except proline). The fourth peak corresponded to a proline carboxypeptidase specific for substrates of the Z-Pro-X type. Apparently, in the hydrolysis of longer proline-containing peptides there must be sequential cooperation between the two carboxypeptidase types. The carboxypeptidases in extracts of starchy endosperms also liberated proline from the peptides Ala-Ala-Ala-Pro and Ala-Ala-Pro while Ala-Pro and Pro-Ala were not attacked. The dipeptides, however, were rapidly hydrolyzed around pH 7 by extracts prepared from the scutella of germinating grains. It is concluded that one part of the proline residues of the reserve proteins is liberated in situ in the starchy endosperm through the combined action of acid proteinases and carboxypeptidases, while another part is taken up in the form of small peptides by the scutellum, where proline is liberated by amino- and/or dipeptidases in some neutral compartment.Abbreviations DFP

Legume–grass interactions have a great influence on grassland primary production and it was recently shown how defoliation of a legume can increase the transfer of fixed N to a neighbouring grass. It has also been shown that defoliation of a plant can increase soil microbial activity and lead to better soil N availability in the rhizosphere of the defoliated plant. We combined these two perspectives and tested whether defoliation of a legume (Lotus corniculatus) can enhance N nutrition of the neighbouring grass (Holcus lanatus) by increasing growth of soil decomposer biota and the availability of soil organic matter N for grass uptake. We grew mixtures of L. corniculatus and H. lanatus in grassland soil that included 15N-labelled L. corniculatus litter. In half of the systems, we subjected L. corniculatus to a defoliation treatment mimicking insect larvae feeding. At destructive harvests 1, 3, 9 and 30 days after the last defoliation event, we determined how L. corniculatus defoliation affected decomposer microbes, protozoa and nematodes and whether these changes among decomposers created a feedback on the growth and 15N uptake of the neighbouring H. lanatus. Defoliation reduced the growth and litter-N uptake, but increased shoot N concentration of L. corniculatus. Of the soil variables measured, defoliation doubled the number of bacterial-feeding protozoa, but did not affect the abundance of decomposer microbes and bacterial- and fungal-feeding nematodes. Defoliation did not have statistically significant effects on H. lanatus shoot growth, shoot N concentration or litter-N uptake. Our results demonstrate how defoliation-induced changes in legume ecophysiology can affect the growth of decomposers in soil. However, these effects did not appear to lead to a significant change in the availability of soil organic N to the neighbouring grass. It seems that when positive effects of legume defoliation on grass N nutrition are found in grassland ecosystems, these are more likely to be explained by direct transfer of fixed N rather than changes in the availability of soil organic matter N. 相似文献

Transgenic (TG) female mice, expressing a chimeric bovine luteinizing hormone (LH) beta-subunit/human chorionic gonadotropin beta-subunit COOH-terminal extension (bLHbeta-CTP) gene, produce high levels of circulating LH and serve as a model for functional ovarian hyperandrogenism and follicular cysts. We report here that obesity is a typical feature of these female mice. The mean body weight of the bLHbeta-CTP females was significantly higher than in controls at, and beyond 5 wk of age, and at 5 mo, it was 32% increased. At this age, the amount of white adipose tissue in the bLHbeta-CTP females was significantly increased, as reflected by the weight difference of the retroperitoneal fat pad. In addition, the expression of leptin mRNA in white adipose tissue of the TG females was elevated about twofold. Serum leptin and insulin levels, and food intake, were also increased significantly in the TG females. Brown adipose tissue (BAT) thermogenic activity, as measured by GDP binding to BAT mitochondria, was reduced (P < 0.05). Ovariectomy at the age of 3 wk totally prevented the development of obesity. In summary, the present results show that intact female bLHbeta-CTP mice are obese, have increased food consumption, and reduced BAT thermogenic activity. The weight gain can be explained partly by elevated androgens but is probably also contributed to the increased adrenal steroidogenesis. Hence, the bLHbeta-CTP mice provide a useful model for studying obesity related to elevated LH secretion, with consequent alterations in ovarian and adrenal function. 相似文献
