Scanning electron microscopy, light microscopy, and histology were used to study growth in species of the pedunculate barnacle genus Octolasmis (O. angulata, O. cor, O. californiana, O. mülleri). These species are epizoic in the gill chamber of portunid crabs and have highly reduced capitular shell plates, with large areas of general cuticle in between. The external integument grows by means of a system of narrow growth zones, one encircling the peduncle and a Y-shaped system on either side of the capitulum. Growth is by a regular series of molts, but shedding of old cuticle and production of new layers is entirely restricted to the growth zones. Just prior to ecdysis, the new cuticle lies in a highly folded fashion beneath the old cuticle that is about to be shed. At ecdysis, the old cuticle breaks along the margins of the growth zones and the resulting scars remain as a system of "ecdysial lines" along either side of the zone. Once exposed after ecdysis, the new cuticle remains as a part of the permanent external integument. The growth zones divide the externa into five cuticular areas, two on the peduncle and three on the capitulum. The calcareous shell plates (carina, paired scuta, and, when present, paired terga) all lie within the capitular regions and the ecdysial lines pass across, not around, these mineralized areas. The number, relative spacing, and topology of the ecdysial lines form a record of the growth history of the specimen. These and other growth patterns demonstrate that size increase is due to the formation of new cuticle by molting in the growth zones, while expansion of the shell plates by mineralization follows only after production of the new cuticle. Thus, although specialized, growth in Octolasmis still complies with the general crustacean model, complicated only by the mineralization of parts of the capitular cuticle into shell plates. The results are compared with the very scarce information on molting in other barnacles. We argue that at least the circular peduncular growth zone is omnipresent in the Cirripedia Thoracica.  相似文献

Pit1 is the human receptor for gibbon ape leukemia virus (GALV) and feline leukemia virus subgroup B (FeLV-B), while the related human protein Pit2 is a receptor for amphotropic murine leukemia virus (A-MuLV). The A-MuLV-related isolate 10A1 can utilize both Pit1 and Pit2 as receptors. A stretch of amino acids named region A was identified in Pit1 (residues 550 to 558 in loop 4) as critical for GALV and FeLV-B receptor function. We have here investigated the role of region A in A-MuLV and 10A1 entry. Insertion of a single amino acid in region A of mouse Pit1 resulted in a functional A-MuLV receptor, showing that region A plays a role in A-MuLV infection. Moreover, the downregulation of 10A1 receptor function by changes in region A of human Pit1 indicates that this region is also involved in 10A1 entry. Therefore, region A seems to play a role in infection by all viruses utilizing Pit1 and/or Pit2 as receptors.  相似文献

Even though most bird species with a raptorial feeding habit express varying extents of reversed sexual dimorphism (RSD: females bigger than males), the evolutionary basis for its maintenance, as well as its possible secondary consequences for the ecological adaptations of the different sexes, is debated. We studied pairs of tawny owls, Strix aluco (females 20% heavier than males), throughout the year by telemetry to test whether any inter-sexual differences in movement patterns, resource partitioning and breeding effort correlated with RSD. Females were larger than males in all body size measures and were 16% heavier than would be expected from the difference in wing length alone. In accordance with predictions from flight economics, males moved longer distances per time unit than females, in particular during the post-fledging season, when they also fed chicks more often than the females. Males had larger home ranges than females during the post-fledging period, whereas the sexes had home ranges of equal size during the non-breeding season. Until 10 days after fledging, females foraged much closer to the offspring than males, apparently balancing their distance to offspring between the needs of offspring guarding and foraging. In males, the parent–offspring distance only increased with decreasing brood condition. The sexes did not differ in habitat use or feeding habits, rendering no indications of food niche partitioning. The study provides further evidence that selection for males to be light and energetically efficient foragers is the main evolutionary force behind RSD in raptorial birds, even when the prey base is confined by territoriality.  相似文献

Over the past few decades, there has been a growing demand for genome analysis of ancient human remains. Destructive sampling is increasingly difficult to obtain for ethical reasons, and standard methods of breaking the skull to access the petrous bone or sampling remaining teeth are often forbidden for curatorial reasons. However, most ancient humans carried head lice and their eggs abound in historical hair specimens. Here we show that host DNA is protected by the cement that glues head lice nits to the hair of ancient Argentinian mummies, 1,500–2,000 years old. The genetic affinities deciphered from genome-wide analyses of this DNA inform that this population migrated from north-west Amazonia to the Andes of central-west Argentina; a result confirmed using the mitochondria of the host lice. The cement preserves ancient environmental DNA of the skin, including the earliest recorded case of Merkel cell polyomavirus. We found that the percentage of human DNA obtained from nit cement equals human DNA obtained from the tooth, yield 2-fold compared with a petrous bone, and 4-fold to a bloodmeal of adult lice a millennium younger. In metric studies of sheaths, the length of the cement negatively correlates with the age of the specimens, whereas hair linear distance between nit and scalp informs about the environmental conditions at the time before death. Ectoparasitic lice sheaths can offer an alternative, nondestructive source of high-quality ancient DNA from a variety of host taxa where bones and teeth are not available and reveal complementary details of their history.  相似文献

In the mid-1980s, two versions of Timm's original immersion sulfide silver method were published. The authors used immersion of tissue in a sulfide solution as opposed to Timm, who used immersion of tissue blocks in hydrogen sulfide-bubbled alcohol. The autometallography staining resulting from the "sulfide only immersion" was not particularly impressive, but the significance of this return to an old approach became obvious when Wenzel and co-workers presented their approach in connection with introduction by the Palmiter group of zinc transporter 3 (ZnT3). The Wenzel/Palmiter pictures are the first high-resolution, high-quality pictures taken from tissues in which free and loosely bound zinc ions have been captured in zinc-sulfur nanocrystals by immersion. The trick was to place formalin-fixed blocks of mouse brains in a solution containing 3% glutaraldehyde and 0.1% sodium sulfide, ingredients used for transcardial perfusion in the zinc-specific NeoTimm method. That the NeoTimm technique results in silver enhancement of zinc-sulfur nanocrystals has been proved by proton-induced X-ray multielement analyses (PIXE) and in vivo chelation with diethyldithiocarbamate (DEDTC). The aims of the present study were (a) to make the immersion-based capturing of zinc ions in zinc-sulfur nanocrystals work directly on sections and slices of fixed brain tissue, (b) to work out protocols that ensure zinc specificity and optimal quality of the staining, (c) to apply "immersion autometallography" (iZnSAMG) to other tissues that contain zinc-enriched (ZEN) cells, and (d) to make the immersion approach work on unfixed fresh tissue.  相似文献

A high sensitivity for the detection of inflammatory and destructive changes in inflammatory joint diseases makes magnetic resonance imaging potentially useful for assigning specific diagnoses, such as rheumatoid arthritis and psoriatic arthritis in arthritides, that remain undifferentiated after conventional clinical, biochemical and radiographic examinations. Pink-footed geese ( Anser brachyrhynchus ) breed in the Arctic, where their populations have doubled since the 1980s. There is concern that nesting geese disturb the fragile tundra and lead to a trophic cascade with strong top-down effects on vegetation and soil processes. A better understanding of the distribution of geese and factors that influence nest site selection is needed to highlight potential problem areas and assess the potential for further population expansion. To help infer the importance of environmental variables on nest site selection, we built generalized additive models using nest observations collected in 2003 and 2004 from the Sassendalen valley, Svalbard, along with a suite of geographical information system explanatory predictors. The fit of the models was very high (explaining over 72% of the deviance), and predictive power to independent samples indicated useful predictions that could discriminate between presences and absence of nests very well (area under the receiver operating characteristic curves exceeded 0.88). Significant predictors of nest site selection included elevation, slope, aspect, percentage of snow cover, percentage of foraging habitat cover, and a spatial autocovariate. Spatial predictions were applied to the broader Nordenskiöldsland region of Svalbard and highlighted the importance of previously unsurveyed locations for nesting.  相似文献
