Computer simulation of hydrodynamic properties of semiflexible macromolecules: randomly broken chains, wormlike chains, and analysis of properties of DNA

The translational and rotational diffusion coefficients and the intrinsic viscosity of semiflexible, randomly broken, and wormlike chains have been obtained by Monte Carlo simulation in the context of the rigid-body treatment. Both approximate and rigorous rigid-body hydrodynamics are used, so that the error introduced by the approximate methods can be evaluated. A randomly broken chain and a wormlike chain having the same contour length and persistence length have the same radius of gyration but different values for any of the hydrodynamic properties. The two types of chains are compared in this regard. Considering that the cross section of the chain is represented by a cylinder better than by a string of spheres, we devise a cylindrical correction to be applied to the results simulated for chains of beads. Application is made to the analysis of experimental data for the translational and rotational coefficients of DNA fragments with up to 10(3) base pairs, obtaining the persistence length for each model. The values for the wormlike chain agree well with model-independent values obtained from radii of gyration and with other literature data at varying ionic strength. The randomly broken chain is equally able to reproduce the experimental length dependence of the properties, but the resulting persistence length may be too high.     

Transplantation of the chick early eye primordium into a lensectomized optic cup of a host embryo

Eye primordia of young chick embryos (stage XII) were transplanted into lensectomized optic cups of older embryos (stage XVII) to analyze the influence of the host retina on the degree of morphological differentiation attained by the donor lens. Embryos were sacrificed 24-96 h later. The donor lens primordium showed a differentiation more in correlation with the host eye cup (stage XXIII) after 24-96 h of incubation.     

Biochemical markers associated with two Mv chromosomes from Aegilops ventricosa in wheat-Aegilops addition lines

Summary The distribution of three biochemical markers, U-1, CM-4 and Aph_v-a, -b, among wheat-Aegilops addition lines carrying M^v chromosomes from Aegilops ventricosa (genomes D^vM^v) has been investigated. Addition lines which had been previously grouped together on the basis of common non-biochemical characters carried marker U-1, a protein component from the 2M urea extract. The added chromosome, in the appropriate genetic background, seems to confer a high level of resistance to the eyespot disease, caused by the fungus Cercosporella herpotrichoides. The other two markers were concomitantly associated with another similarly formed group of addition lines. Both CM-4, a protein component from the chloroform:methanol extract, and Aph_v-a, -b, alkaline phosphate isozymes, have been previously shown to be associated with homoeologous chromosome group 4, which suggests that the added chromosome in the second group of addition lines is 4M^v.     

Severe mental deficiency,proportionate dwarfism,and delayed sexual maturation

Summary Two 46,XY brothers were found to have a previously undescribed syndrome characterized by severe mental deficiency, proportionate dwarfism, and delayed sexual development. A recessive mode of inheritance, either autosomal or X-linked, is assumed.     

Review of the Neotropical Isertieae (Rubiaceae)

All neotropical genera previously included in the tribe Isertieae A. Rich. ex DC. are considered. The tribe Isertieae is here delimited by the following characters: absence of rhaphides, valvate aestivation of the corolla, cordiform or discoid placenta, fleshy indehiscent fruit, and seed coat cells with pitted walls. As so delimited, the tribe definitely includes the neotropical generaIsertia Schreb.,Gonzalagunia Ruiz & Pav.,Amphidasya Standl.,Raritebe Wernh.,Sabicea Aubl., andMussaenda L. as well as some paleotropical genera not here considered. The following neotropical genera are excluded from tribe Isertieae:Hippotis Ruiz & Pav.,Pentagonia Benth.,Sommera Schlecht.,Tammsia Karst.,Coccocypselum P. Browne,Schradera Vahl,Didymochlamys Hook, f., andXerococcus Oerst. The position of four genera,Striolaria Ducke,Patima Aubl.,Habroneuron Standl., andNeosabicea Wernh., remains uncertain due to a lack of material.     

A bead and spring model for the stiffness of DNA.

The chain stiffness of linear native DNA is represented by a generalized bead and spring model recently proposed. It incorporates molecular rigidity by means of springs between beads, which are second neighbors along the contour of the chain. These springs are equivalent to elastic forces having longitudinal and transversal contributions. The model is compared with existing experimental data of sedimentation and low-angle light scattering to obtain the statistical parameters of DNA. The value of the statistical length obtained with this model is 1300 Å. The same value is obtained with the wormlike chain. Throughout this analysis, excluded volume is left out as a simplifying assumption.
