In a previous study (M. Sasaki, J. Maki, K. Oshiman, Y. Matsumura, and T. Tsuchido, Biodegradation 16:449-459, 2005), the cytochrome P450 monooxygenase system was shown to be involved in bisphenol A (BPA) degradation by Sphingomonas sp. strain AO1. In the present investigation, we purified the components of this monooxygenase, cytochrome P450 (P450bisd), ferredoxin (Fd(bisd)), and ferredoxin reductase (Red(bisd)). We demonstrated that P450bisd and Fd(bisd) are homodimeric proteins with molecular masses of 102.3 and 19.1 kDa, respectively, by gel filtration chromatography analysis. Spectroscopic analysis of Fd(bisd) revealed the presence of a putidaredoxin-type [2Fe-2S] cluster. P450(bisd), in the presence of Fd(bisd), Red(bisd), and NADH, was able to convert BPA. The K(m) and kcat values for BPA degradation were 85 +/- 4.7 microM and 3.9 +/- 0.04 min(-1), respectively. NADPH, spinach ferredoxin, and spinach ferredoxin reductase resulted in weak monooxygenase activity. These results indicated that the electron transport system of P450bisd might exhibit strict specificity. Two BPA degradation products of the P450(bisd) system were detected by high-performance liquid chromatography analysis and were thought to be 1,2-bis(4-hydroxyphenyl)-2-propanol and 2,2-bis(4-hydroxyphenyl)-1-propanol based on mass spectrometry-mass spectrometry analysis. Glucocorticoid (GC) excess promotes adipose tissue accumulation, and 11β-hydroxysteroid dehydrogenase type 1 (11β-HSD1) plays an important role in the local amplification of GC. Therefore, in this study, we investigated the effects of carbenoxolone (CBX), an 11β-HSD1 inhibitor, on morphological changes in visceral fat, and the expression of genes involved in adipogenesis and lipid metabolism in high-fat (HF) diet-fed mice. Mice were fed a HF diet from 5 weeks of age. At 10 weeks of age, the mice received an intraperitoneal injection of CBX or vehicle every day for 2 weeks. CBX decreased body weight and visceral fat mass, and improved insulin sensitivity in HF-fed mice. This was accompanied by reduced adipocyte size and a decrease in large-sized adipocytes in visceral fat. The expression of adipogenesis (PPARγ and C/EBPα), glucose transport (GLUT4) and lipid metabolism (LPL, ATGL, and HSL)-related genes were suppressed in CBX mice. CBX treatment induced beneficial morphological changes in visceral fat and decreased the expression of adipogenesis, glucose transport and lipid metabolism-related genes. These findings reveal a potential mechanism underling the effects of CBX on reduced fat accumulation and improved insulin sensitivity. 相似文献

To clarify the feeding habits and major food sources of sandy beach fishes, the gut contents of 55 fish species collected
on a sandy beach at Fukiagehama, South-West Kyushu Island, Japan, were examined. Ontogenetic changes in food preference were
recognized in nine species (Hypoatherina valenciennei, Lateolabrax japonicus, Trachurus japonicus, Sillago japonica, Sphyraena japonica, Paralichthys olivaceus, Heteromycteris japonica, Paraplagusia japonica, and Takifugu niphobles). A cluster analysis based on dietary overlaps showed that the sandy beach fish assemblage comprised six trophic groups (mysid,
amphipod, zooplankton, juvenile fish, terrestrial insect, and mollusk feeders). Of these, the first three groups were the
most abundantly represented, whereas the last two were represented by only a single species. These results indicated that
epibenthic macrofauna, such as mysids and gammaridean amphipods, and zooplankton, were important food resources for the fish

Inhalation of hydrogen (H2) gas has been demonstrated to limit the infarct volume of brain and liver by reducing ischemia-reperfusion injury in rodents. When translated into clinical practice, this therapy must be most frequently applied in the treatment of patients with acute myocardial infarction, since angioplastic recanalization of infarct-related occluded coronary artery is routinely performed. Therefore, we investigate whether H2 gas confers cardioprotection against ischemia-reperfusion injury in rats. In isolated perfused hearts, H2 gas enhances the recovery of left ventricular function following anoxia-reoxygenation. Inhaled H2 gas is rapidly transported and can reach ‘at risk’ ischemic myocardium before coronary blood flow of the occluded infarct-related artery is reestablished. Inhalation of H2 gas at incombustible levels during ischemia and reperfusion reduces infarct size without altering hemodynamic parameters, thereby preventing deleterious left ventricular remodeling. Thus, inhalation of H2 gas is promising strategy to alleviate ischemia-reperfusion injury coincident with recanalization of coronary artery. 相似文献

Background
Automatic measurement becomes a preference, and indeed a necessity, when analyzing 1000 s of ECGs in the setting of either drug-inducing QT prolongation screening or genome-wide association studies of QT interval. The problem is that individual manufacturers apply different computerized algorithms to measure QT interval. We conducted a comparative study to assess the outcomes with different automated measurements of QT interval between ECG machine manufacturers and validated the related heart rate correction methods.Methods and Results
Herein, we directly compared these different commercial systems using 10,529 Fukuda Denshi ECGs and 72,754 Nihon Kohden ECGs taken in healthy Japanese volunteers. Log-transformed data revealed an equal optimal heart rate correction formula of QT interval for Fukuda Denshi and Nihon Kohden, in the form of QTc = QT/RR−0.347. However, with the raw data, the optimal heart rate correction formula of QT interval was in the form of QTc = QT+0.156×(1-RR) for Fukuda Denshi and QTc = QT+0.152×(1-RR) for Nihon Kohden. After optimization of heart rate correction of QT interval by the linear regression model using either log-transformed data or raw data, QTc interval was ∼10 ms longer in Nihon Kohden ECGs than in those recorded on Fukuda Denshi machines. Indeed, regression analysis revealed that differences in the ECG machine used had up to a two-fold larger impact on QT variation than gender difference. Such an impact is likely to be of considerable importance when ECGs for a given individual are recorded on different machines in the setting of multi-institutional joint research.Conclusions
We recommend that ECG machines of the same manufacturer should be used to measure QT and RR intervals in the setting of multi-institutional joint research. It is desirable to unify the computer algorithm for automatic QT and RR measurements from an ECG. 相似文献988.

The intracellular parasite Salmonella enterica serovar Typhimurium causes a typhoid-like systemic disease in mice. Whereas the survival of Salmonella in phagocytes is well understood, little has been documented about the exit of intracellular Salmonella from host cells. Here we report that in a population of infected macrophages Salmonella induces “oncosis,” an irreversible progression to eukaryotic cell death characterized by swelling of the entire cell body. Oncotic macrophages (OnMs) are terminal deoxynucleotidyltransferase-mediated dUTP-biotin nick end labeling negative and lack actin filaments (F-actin). The plasma membrane of OnMs filled with bacilli remains impermeable, and intracellular Salmonella bacilli move vigorously using flagella. Eventually, intracellular Salmonella bacilli intermittently exit host cells in a flagellum-dependent manner. These results suggest that induction of macrophage oncosis and intracellular accumulation of flagellated bacilli constitute a strategy whereby Salmonella escapes from host macrophages. 相似文献

The role of the periodontal ligament (PDL) is to support the tooth during function and resist external forces applied to it. The dominant vertical component of these forces is associated with shear in the PDL. Little information, however, is available on the dynamic behavior of the PDL in shear. Therefore, the present study was designed to determine the dynamic shear properties of the PDL in the porcine molar (n=10). From dissected mandibles transverse sections of the mesial root of the first molar were obtained at the apical and coronal levels and used for dynamic shear tests. Shear strain (0.5%, 1.0%, and 1.5%) was applied in superoinferior direction parallel to the root axis with a wide range of frequencies (0.01-100 Hz). The dynamic complex and storage moduli increased significantly with the loading frequency, the dynamic loss modulus showed only a small increase. The dynamic elasticity was significantly larger in the coronal region than in the apical region although the dynamic viscosity was similar in both regions. The present results suggest that non-linearities, compression/shear coupling, and intrinsic viscoelasticity affect the shear material behavior of the PDL, which might have important implications for load transmission from tooth to bone and vice versa. 相似文献

In vitro cultures of plant cells have often been utilized to generate genetic variations, which are designated somaclonal
variations. Little is known about the major genetic alterations in the cultured cells and the nature of these genetic changes.
Here, we examined different lines of rice Oc cells that have been cultured for more than 20 years on agar media or in liquid
media. We surveyed 35 clones obtained from PCR amplification of the 3-kb EPSPs–RPS20 region. The sequence divergence among the Oc cells was even greater than that between Japonica and Indica rice cultivars.
The divergent sequences appeared to be maintained as multiple copies in a single cell. Surprisingly, the nucleotide substitutions
in the Oc cells were characterized by an extremely high frequency of transition mutations of A/T-to-G/C, a feature which is
similar to that of the mutations caused by chemical mutagens such as 5-bromouracil and 2-aminopurine. Although no replacements
in the exons of this region were observed among the AA-genome Oryza species, our results revealed that the nucleotide substitutions of the cultured cell lines occurred more frequently at replacement
sites in the exons than at synonymous sites. These distinct mutation biases found in rice in vitro cultures might contribute
importantly to somaclonal variations.
