The aggregation of amyloid β-peptide (Aβ) into β-sheet-rich aggregates is a crucial step in the etiology of Alzheimer’s disease. Helical forms of Aβ have been suggested to be intermediates in the aggregation process of the peptide in aqueous phase, micelles and membranes. A stable helical Aβ analog would be useful to investigate the role of helical intermediates in fibrillization by Aβ. Here we designed a helical analog by simply cross-linking the Cys residues of A30C, G37C-Aβ1-42 with 1,6-bismaleimidohexane. The analog assumed a weak α-helical conformation in model membranes mimicking lipid raft microdomains of neuronal membranes under conditions in which the wild-type Aβ1-42 formed a β-sheet, indicating the cross-linking locally induced a helical conformation. Furthermore, addition of equimolar helical Aβ analog significantly reduced the amyloid formation and cytotoxicity by Aβ1-42. Thus, our helical Aβ1-42 is not only a model peptide to investigate the role of helical intermediates in fibrillization by Aβ, but also an inhibitor of Aβ-induced cytotoxicity.  相似文献

A herd of 28–33 Japanese Black cows (Bos taurus) were allowed to graze on an experimental plot comprising monoculture swards of centipedegrass (Eremochloa ophiuroides) and bahiagrass (Paspalum notatum) (0.39 and 0.61 ha, respectively) for 3–5 days each month (from 0850 to 1600 hours) between May (late spring) and October (mid-autumn). On a monthly basis, the animals showed an increasing trend to select centipedegrass in preference to bahiagrass as the relative crude protein (CP) concentration of the former increased relative to the latter. On a daily basis, the animals showed a decreased selectivity for centipedegrass with the progression of grazing days, as centipedegrass sward lost both quantity and quality faster than bahiagrass sward under higher degrees of defoliation. Animals maintained similar bite rates on the two swards by modifying feeding station behavior as soon as they switched between the swards, i.e., they increased the number of bites per feeding station and decreased the number of feeding stations selected per unit grazing time soon after switching to centipedegrass, with the reverse process occurring soon after the switch to bahiagrass. The results show CP concentration to be a partial forage factor influencing animals’ choice between tropical grasses growing as patches. The results also demonstrate that animals have an ability to adapt their foraging behavior flexibly and rapidly to varying types of vegetation.  相似文献

The third repeat fragment (R3) in the four-repeat microtubule-binding domain of the water-soluble tau protein has been considered to play an essential role in the protein's filamentous assembly. To clarify the associational and conformational features that differentiate R3 from the second repeat, R2, the heparin-induced assembly profiles of these peptide fragments were monitored by the thioflavin fluorescence method and electron microscopy. The trifluoroethanol-induced reversible conformational change from a random structure to an alpha-helical structure, in an aqueous solution, was monitored by CD measurement, and the structure of R2 in trifluoroethanol solution was analyzed by a combination of two-dimensional 1H-NMR measurements and molecular modeling calculations to facilitate comparison with the structure of R3. The speed of R3 assembly was remarkably faster than that of R2, in spite of their similar amino acid sequences. The averaged NMR conformers of R2 exhibited the whole-spanning alpha-helical structure. Similar features observed in R2 and R3 conformers in trifluoroethanol were that the Leu10-Leu20/Lys20 sequence takes a helical structure with the amphipathic-like distribution of the respective side-chains, whereas the C-terminal moieties are both flexible. In contrast, a notable difference was observed at the N-terminal Val1-Lys6 sequence, namely, a helical conformation for R2 and an extended conformation for R3. These conformational behaviors would be associated with the different self-aggregation speeds and seeding reactions between R2 and R3.  相似文献

Lactococcus lactis subsp. lactis strains show glutamate decarboxylase activity, whereas L. lactis subsp. cremoris strains do not. The gadB gene encoding glutamate decarboxylase was detected in the L. lactis subsp. cremoris genome but was poorly expressed. Pup ultrasonic vocalizations (USVs) are emitted from maternally separated pups and are thought to be a trigger for eliciting maternal behavior in mice. We investigated the effects of social experience and gonadectomy on the retrieving behavior of mice and their responses to pup USVs produced by a nanocrystalline silicon thermo-acoustic emitter. In each experiment, virgin, gonadectomized, sham-operated, sexually experienced, and parenting mice of both sexes were used, and the effects of these manipulations were compared in each sex. The retrieving behavior of both sexes increased with social experience or gonadectomy. In particular, mothers showed the highest retrieving activity among female groups, while castrated male mice showed the highest retrieving activity among male groups. All groups of female mice responded to pup USVs, with the responsiveness of sexually experienced female mice being the most enhanced. Unlike the females, virgin male mice did not respond to pup USVs, although socially experienced or castrated males showed this response; fathers exhibited the highest responsiveness. These results suggest that not only parenting experience, but also mating experience, may enhance retrieving activity and response to pup USVs in mice of both sexes. Nevertheless, the degree to which parenting experience contributed to the enhancement of both activities differed between the sexes. Furthermore, gonadectomy enhanced both activities in both sexes, although its effect was more prominent in males. Overall, our findings suggest that alteration in responsiveness of mice to pup USVs might be one of the changes in parental behavior caused by social experiences or gonadal hormones.  相似文献

Many carnivorous mammals consume fruits and disperse the intact seeds to specific sites. Few studies have attempted to quantify this seed dispersal or evaluate its effectiveness, despite its potential importance and functional uniqueness. In the study reported here, we found that a frugivorous carnivore, the common palm civet (Paradoxurus hermaphroditus), generated seed shadows that are distinct from those of the sympatric frugivore, the pig-tailed macaque (Macaca nemestrina), and played a unique and important role in the regeneration of Leea aculeata (Leeaceae). We found that macaques dispersed the seeds randomly, while civets dispersed them non-randomly to sites such as the banks of small rivers, rain-flow paths, abandoned trails, and treefall gaps, which are characterized by low stem density and canopy cover. Seeds of L. aculeata that were dispersed by civets to the banks of rivers and gaps had significantly higher survival and growth rates than those dispersed to rain-flow paths or abandoned trails. Seeds dispersed by macaques or to random locations also had low survival. Although the effects of the civets on seed fate were not straightforward, compared with macaques and random dispersal, civets significantly enhanced the survival and growth of L. aculeata seeds after 1 year. These results indicate that non-random dispersal by civets is important for the persistence of L. aculeata. Civets may disperse other plant species and thus could have profound effects on forest dynamics.  相似文献

Interspecific hybridization in plants is known to have ecological effects on associated organisms. We examined the differences in insect herbivore community structure and grazing pressure on tree canopy leaves among natural hybrids and their parental oak species. We measured leaf traits, herbivore community structure, and grazing pressure on leaves of two oak species, Quercus crispula and Q. dentata, and their hybrids. The concentration of nitrogen in canopy leaves was greater in hybrids and in Q. dentata than in Q. crispula. The concentration of total phenolics was lower in hybrids than in Q. crispula. The concentration of condensed tannin was greater in hybrids than in Q. crispula. Relative herbivore abundance and species richness were greater on oak hybrids than on either parental species; herbivore species diversity and composition on hybrids were close to those on Q. crispula. Herbivore grazing pressure was lower on hybrids and Q. dentata than on Q. crispula. There was a negative correlation between herbivore grazing pressure and leaf nitrogen, suggesting that interspecific variation among oak taxa in herbivore pressure may be explained by leaf nitrogen; variation in herbivore community structure among oak taxa is likely to be controlled by polygenic leaf traits. Differing responses of (1) herbivore community structure and (2) herbivore grazing pressure to host plant hybridization may play important roles in regulating herbivore biodiversity in cool‐temperate forest canopies.  相似文献
