The present study estimates herpes simplex virus type 2 (HSV-2) seroprevalence and evaluates its association with age, sex, human herpesvirus type 8 (HHV-8) and human immunodeficiency virus (HIV) among adults in Croatia. A cross-sectional survey included 166 HIV-infected patients and 219 blood donors. Antibodies against HSV-2 were determined by enzyme immunoassays based on gG2 recombinant glycoprotein. HSV-2 seroprevalence was 45.8% in HIV-infected patients and 8.7% in blood donors (p < 0.0001; OR 8.8; 95% CI 5.05-15.49). Independent predictors of HSV-2 seropositivity were HIV infection (OR 11.0; 95% CI 5.93-20.41), female gender (OR 2.28; 95% CI 1.22-4.26), older age (OR 3.93; 95% CI 2.74-7.11), and HHV-8 seropositivity (OR 2.72; 95% CI 1.09-6.75). Understanding the epidemiology of HSV-2 is a critical first step in designing interventions to decrease HSV-2 and HIV transmission. The association of HSV-2 with HIV infection and HHV-8 antibodies suggests a similar transmission route.  相似文献

The aim of the study was to determine the relation between motor abilities and belly dance performance in elementary school fifth- and sixth-grade female students. A battery of 19 motor tests was used in a sample of 96 students twice, i.e. at the beginning (initial measurement) and at the end (final measurement) of the academic year. On initial measurement, five factors were isolated by the motor space factor analysis: first factor of muscular-aerobic endurance; second factor integrating the strength of legs, coordination of foot and hand movement, and agility; third factor integrating explosive strength of the arms with speed and body coordination; fourth factor defined by flexibility (muscle tone regulation); and fifth factor integrating explosive strength of legs with equilibrium. On final measurement, five factors were isolated as well: first factor as a general one integrating coordination abilities, explosive strength of legs and flexibility; second factor defined by repetitive strength of the trunk and legs; third factor defined by rhythm coordination accompanied by flexibility; fourth factor predominantly defined by equilibrium (accompanied by explosive strength of throwing type and speed); and fifth factor predominantly defined by static strength of arms and legs (accompanied by arm movement frequency). On initial measurement, fourth factor responsible for muscle tone regulation and second factor integrating the strength of legs, coordination of movement frequency of arms and legs, and agility were found to be the best predictors of belly dance performance. In this setting, the tests of forward bow (flexibility) and sit-ups (repetitive strength of abdominal musculature) proved superior in differentiating high performance students and those less successful in belly dance. On final measurement, third factor named rhythm coordination (accompanied by muscle tone regulation) and second factor defined by repetitive strength of the trunk and legs were the best predictors of belly dance performance. In this setting, the tests of rhythm coordination, flexibility tests, tests of repetitive strength of the trunk and legs, and test of aerobic endurance proved superior in differentiating high performance and less successful students in belly dance.  相似文献

Before the secretion of hard dental tissues, tooth germs undergo several distinctive stages of development (dental lamina, bud, cap and bell). Every stage is characterized by specific proliferation patterns, which is regulated by various morphogens, growth factors and homeodomain proteins. The role of MSX homeodomain proteins in odontogenesis is rather complex. Expression domains of genes encoding for murine Msx1/2 during development are observed in tissues containing highly proliferative progenitor cells. Arrest of tooth development in Msx knockout mice can be attributed to impaired proliferation of progenitor cells. In Msx1 knockout mice, these progenitor cells start to differentiate prematurely as they strongly express cyclin-dependent kinase inhibitor p19INK4d. p19INK4d induces terminal differentiation of cells by blocking the cell cycle in mitogen-responsive G1 phase. Direct suppression of p19INK4d by Msx1 protein is, therefore, important for maintaining proliferation of progenitor cells at levels required for the normal progression of tooth development. In this study, we examined the expression patterns of MSX1, MSX2 and p19INK4d in human incisor tooth germs during the bud, cap and early bell stages of development. The distribution of expression domains of p19INK4d throughout the investigated period indicates that p19INK4d plays active role during human tooth development. Furthermore, comparison of expression domains of p19INK4d with those of MSX1, MSX2 and proliferation markers Ki67, Cyclin A2 and pRb, indicates that MSX-mediated regulation of proliferation in human tooth germs might not be executed by the mechanism similar to one described in developing tooth germs of wild-type mouse.  相似文献

Tellurium (Te) and selenium (Se) belong chemically to the VIa group of elements. Se represents an essential element closely related to thyroid function. Te has growing application in industrial processes. Little is known about the Te biological activity, particularly with respect to potential chemical interactions with Se-containing components in the organism. In this study, female Wistar rats (body weight: 115-120 g) received sodium selenite pentahydrate (10 mg/L) or sodium tellurite (9.4 mg/L) in drinking water for 6 wk. Additional groups of rats received their combination with zinc sulfate heptahydrate (515 mg/L). The stimulation of 5’-DI-I activity due to selenite (to 158%, p<0.01) or tellurite treatment (to 197%, p<0.01) was seen; however, no effect on glutathione peroxidase was demonstrated in this experiment. An elevation of T4, T3, and rT3 serum levels was measured in the Se+Te-treated group; T4 and rT3 levels were elevated in the Te+Zn-treated group. Te accumulates in the thyroid gland and influences the zinc thyroid level. Te treatment alone and in combination with Se or Zn decreased the iodine thyroid concentration to 65-70% of the control value. Further studies are needed to clarify the nature and effects of these events.  相似文献

Bifidobacteria are important members of the human gut flora, especially in infants. Comparative genomic analysis of two Bifidobacterium animalis subsp. lactis strains revealed evolution by internal deletion of consecutive spacer-repeat units within a novel clustered regularly interspaced short palindromic repeat locus, which represented the largest differential content between the two genomes. Additionally, 47 single nucleotide polymorphisms were identified, consisting primarily of nonsynonymous mutations, indicating positive selection and/or recent divergence. A particular nonsynonymous mutation in a putative glucose transporter was linked to a negative phenotypic effect on the ability of the variant to catabolize glucose, consistent with a modification in the predicted protein transmembrane topology. Comparative genome sequence analysis of three Bifidobacterium species provided a core genome set of 1,117 orthologs complemented by a pan-genome of 2,445 genes. The genome sequences of the intestinal bacterium B. animalis subsp. lactis provide insights into rapid genome evolution and the genetic basis for adaptation to the human gut environment, notably with regard to catabolism of dietary carbohydrates, resistance to bile and acid, and interaction with the intestinal epithelium. The high degree of genome conservation observed between the two strains in terms of size, organization, and sequence is indicative of a genomically monomorphic subspecies and explains the inability to differentiate the strains by standard techniques such as pulsed-field gel electrophoresis.Actinobacteria, Firmicutes, Proteobacteria, and Bacteroidetes are dominant microbial phyla widely distributed in diverse ecosystems on the planet (10, 13, 20, 23, 33, 40, 51). Metagenomic analyses of the microbial landscape inhabiting various mammalian environments, notably the human gastrointestinal tract (GIT) and skin, have specifically identified Actinobacteria as an important and occasionally dominant phylum (18, 21, 33). Among the members of the large, diverse, and dynamic microbial community residing in the human GIT, Bifidobacterium is a dominant genus considered beneficial to humans and includes probiotic strains (live microorganisms which, when administered in adequate amounts, confer a health benefit on the host) (11). The population of bifidobacteria in the human intestine varies over time. Following vaginal delivery, the GIT of healthy newborns is typically colonized by bifidobacteria, especially in breast-fed infants, during the first few days of life (12). Interindividual variation, however, is remarkable in the human infant intestinal flora (41), and dominant genera are not always consistent across metagenomic analyses of the human gut flora (18, 30, 33, 41). Over time, the infant intestinal ecosystem becomes more complex as the diet becomes more diverse, with bifidobacteria typically remaining dominant until weaning (30).Bifidobacterium animalis subsp. lactis is a gram-positive lactic acid bacterium commonly found in the guts of healthy humans and has been identified in the infant gut biota, particularly in ileal, fecal, and mucosal samples (52, 56). Some strains of B. animalis subsp. lactis are able to survive in the GIT, to adhere to human epithelial cells in vitro, to modify fecal flora, to modulate the host immune response, or to prevent microbial gastroenteritis and colitis (4, 15, 20, 40, 52, 56). Additionally, B. animalis subsp. lactis has been reported to utilize nondigestible oligosaccharides, which may contribute to the organism''s ability to compete in the human gut. Carbohydrates resistant to enzymatic degradation and not absorbed in the upper intestinal tract are a primary source of energy for microbes residing in the large intestine. The benefits associated with probiotic strains of B. animalis subsp. lactis have resulted in their inclusion in the human diet via formulation into a large array of dietary supplements and foods, including dairy products such as yogurt. Deciphering the complete genome sequences of such microbes will provide additional insight into the genetic basis for survival and residence in the human gut, notably with regard to the ability to survive gastric passage and utilize available nutrients. Also, these genomes provide reference sequences for ongoing metagenomic analyses of the human environment, including the gut metagenome.Bifidobacterium animalis subsp. lactis is the most common bifidobacterium utilized as a probiotic in commercial dairy products in North America and Europe (22, 38). However, despite this commercial and probiotic significance, strain-level differentiation of B. animalis subsp. lactis strains has been hindered by the high genetic similarity of these organisms, as determined by pulsed-field gel electrophoresis and other nucleic acid-based techniques (6, 55, 56), and the lack of available genomic sequence information. The genome sequence of strain BB-12 (17) is not currently publicly available, and only a draft genome sequence in 28 contigs is available for strain HN019 (GenBank project 28807). The complete B. animalis subsp. lactis genome for strain AD011 (28) was only recently (2009) published. While this was an important first step, a single genome does not allow identification of unique targets for strain differentiation or comparative analyses within the subspecies.The objectives of this study were to determine the complete genome sequences of two B. animalis subsp. lactis strains, the type strain and a widely used commercial strain, to provide insights into the functionality of this species and into species identification and strain specialization.  相似文献

The aim of this study is to find dialysis relevant sonographic parameters of painful shoulder of the symptomatic dialyzed patients comparing them with parameters in asymptomatic dialyzed patients and healthy volunteers. Significant difference in all metric parameters (thickness of supraspinatus tendon, diameter of biceps tendon sheet and capsula-bone distance) were noticed between all groups and the symptomatic had the highest values. Asymptomatic had the higher values then volunteers. Inhomogenicity of the tendon and biceps tendon sheet effusion in the symptomatic patients were the most often occurred. Subdeltoid effusion, deposits and tendon rupture were found only in symptomatic patients. No difference in presence of calcifications between symptomatic and asymptomatic was found. Metric parameters are relevant and associated with dialysis, as well as biceps tendon effusion tendon inhomogenicity, deposits and subdeltoid effusion. Tendon ruptures are relatively rare and nonspecific.  相似文献

Star-shaped poly(ethylene glycol)-block-polyethylenimine [star-(PEG-b-PEI)] significantly enhance plasmid DNA condensation of low molecular weight (MW) PEIs. The star-block copolymers were prepared via a facile synthesis route using hexamethylene diisocyanate as linker between PEG and PEI blocks. NMR and FT-IR spectroscopy confirmed the structures of intermediately activated PEG and final products. Furthermore, the copolymers were characterized by size exclusion chromatography, static light scattering, and viscosimetry. Their molecular weights (M(w) 19-26 kDa) were similar to high MW PEI (25 kDa). Thermoanalytical investigations (thermogravimetric analysis, differential scanning calorimetry) were also performed and verified successful copolymer synthesis. DNA condensation with the low MW PEIs (800 and 2000 Da) and their 4- and 8-star-block copolymers was studied using atomic force microscopy, dynamic light scattering, zeta-potential measurements, and ethidium bromide (EtBr) exclusion assay. It was found that low MW PEIs formed huge aggregates (500 nm to 2 microm) in which DNA is only loosely condensed. By contrast, the star-block copolymers yielded small (80-110 nm), spherical and compact complexes that were stable against aggregation even at high ionic strength and charge neutrality. Furthermore, as revealed in the EtBr exclusion assay these star-block copolymers exhibited a DNA condensation potential as high as high MW PEI. Since these star-(PEG-block-PEI) copolymers are composed of relatively nontoxic low MW PEI and biocompatible PEG, their potential as gene delivery agents merits further investigations.  相似文献

In the past decade, extreme hydrological events were expressed with extreme droughts and floods in temperate regions. The aim of this paper is to explain how such changes in hydrology can influence cyanobacterial populations in floodplain ecosystems. We therefore analyzed a 6-year (2003–2008) study of the phytoplankton in the Kopački Rit floodplain, one of the largest natural floodplains in the middle section of the Danube River (Europe). During the studied period, the shallow floodplain lake shifted between a state of turbid water, characterized by high phytoplankton biomass and regular appearance of cyanobacteria blooms, to a state of clear water with very low phytoplankton biomass and absence of cyanobacteria, and back to the turbid state. Apparently, the major forces driving the cyclic shift were closely related to extremely high and long-lasting flood events. Significant increase in water level, low hydraulic residence time of water, decrease in transparency and low-light climate, together with mass developed aquatic macrophyte vegetation in the whole inundated floodplain were unfavorable conditions for growth and proliferation of cyanobacteria. With the establishment of the flood regime characterized by long-lasting periods without flooding, in-lake processes prevailed leading to cyanobacterial bloom. The most successful were filamentous non-N-fixing cyanobacteria tolerant to mixed and low-light conditions (Planktothrix and Limnothrix) and invasive species Cylindrospermopsis raciborskii. Their massive development led to the establishment of a phytoplankton steady state. All our results demonstrate that the altered intensity and frequency of flood events will have pronounced effects on the appearance of cyanobacterial blooms and generally on alternative stable states in the floodplain. Relating to this, management objectives should be focused on qualifications of changes in hydrology and projecting those effects for potential floodplain restoration.  相似文献
