The effects produced on bacteriorhodopsin by low concentrations of several detergents have been studied by absorption and fourth-derivative spectrophotometry. Sodium dodecyl sulfate induces the appearance of the blue form of bacteriorhodopsin (λmax = 600 nm) at pH values up to 7.0 in a reversible manner. The apparent pK of the purple-to-blue transition raised with increasing concentration of SDS. Of the other detergents tested, only sodium dodecyl-N-sarcosinate showed a slight red-shift of the absorption band to 580 nm, whereas sodium taurocholate, Triton X-100 and cetyltrimethylammonium bromide did not favour the appearance of the blue form. The effect of SDS was found to be consistent with a localized conformational change that moves away the counter-ion of the protonated Schiff base.  相似文献

Summary Lactic acid produced by continuous culture of L.casei in an upflow packed bed reactor, was recovered with Amberlite IRA 400 in a fluidized bed column. Bed expansions of 1.25 and 2.25 were applied. Reutilization did not alter the capability of net recovery of 0.048 ± 0.01 g lactic acid/g resin. When 2200 cm/h of ascensional velocity was used, (bed expansion of 2.25), the resin adsorbed 39.3% of the initial lactic acid and 63.5% was eluted. This resin supported the highest exchange capacity of 0.126 g lactic acid/g resin. Applying high flow rates, the process has potential industrial applications due to the short time employed.  相似文献

This paper describes a simulation problem, motivated by the study of glaucoma, a very serious and widespread ocular illness. To ascertain whether a patient suffers from glaucoma, a perimetric test is done, but the evolution of the disease is very slow, and large longitudinal sets of tests taken on the same patient are needed to study its evolution, to analyze the efficiency of existing methods to detect the progression of glaucoma and to develop new ones. Simulation can be a very useful procedure to get appropriate data sets to work with. Our aim in this work is to simulate several VFs in a healthy patient to reflect his evolution in time. We use a spatio‐temporal model to simulate from, taking into account the correlation existing between the observed (or simulated) values in space and time. Abstract.  A phylogenetic analysis, at a tribal and subtribal level, of the subfamily Hydrophilinae was conducted. The analysis was based on twenty-nine taxa (twenty-three genera) and 148 characters (fifty-eight from immature stages and bionomics, and ninety from adults). According to the present study, Hydrophilinae is monophyletic, and except for the tribe Hydrophilini which appears as polyphyletic (it includes the subtribes Hydrophilina, Hydrobiina, and Acidocerina), the remaining tribes are monophyletic. The tribes Berosini and Chaetarthriini form the basal clade of Hydrophilinae. One unexpected result is the relationship between the subtribe Hydrobiina and the tribe Sperchopsini, which form a well-supported clade. The final tree has the following structure: (((((Sperchopsini Hydrobiina) (Anacaenini Laccobiini)) Acidocerina) Hydrophilina) (Berosini Chaetarthriini)). The results partially disagree with the phylogeny presented by Hansen, in 1991, which was based mostly on adult characters. Several evolutionary trends are briefly discussed: the types of egg case, the morphology of the clypeolabrum, mouthparts, legs, and breathing adaptations in larvae.  相似文献

Mean square optical anisotropies and molar Kerr constants were calculated for homopolypeptides of the 20 natural amino acids and of several enzymes and proteins in the random-coil state. The effect of hydration was taken into account in constructing the molecular potential that gives the conformational energies as a function of the rotational angles phi and psi of the backbone and chi(1) of the side chain. The Rotational Isomeric State model was used in calculated energies, the Valence Optical Scheme and the matrix calculus technique of Flory being employed in the evaluation of the optical properties. The results are compared with calculations for the same substances that were performed without taking into account the solvent, as well as with other similar studies. The Kerr constant is confirmed as being one of the most sensitive properties of a given polypeptide to the residue class and to the sequence of those residues.  相似文献

As part of an epidemiological study on leishmaniasis in Merida, Venezuela, the diversity and dispersion of sandflies species found in 15 localities between 175 m and 1,960 m.a.s.l., are presented. From 7,126 collected sandflies (5,132 female and 1,994 male), 24 species were identified, 10 of them recognized as anthropophilic. The relation species-altitude is presented, and the species composition found in human dwellings, periodomestic and sylvatic areas, are recorded. The possible role of the identified species on the transmission of leishmaniasis in the andean region, is discussed.  相似文献

The kinetics of anaerobic degradation of a molasses wastewater were measured under constant pH conditions in a laboratory scale packed bed reactor. In continuous and batch experiments the formation and degradation rates of the organic acids (butyric, propionic and acetic) have been followed. The influence of hydrogen gas on the acid degradation rates has been measured and, contrary to the literature and the thermo-dynamic calculations, no inhibition was detected, biofilm diffusional effects may be the reason. Two dynamic simulation models were tested, a heterogeneous model, which considered the biofilm diffusion-reaction phenomena and a quasihomogeneous model with the same kinetics. Except for hydrogen, the diffusion effects were found to be negligible. Otherwise both models gave essentially the same results and the time profiles of acids, hydrogen, carbon dioxide and methane agreed relatively well with dynamic startup experiments. Batch experiments showed the acid concentrations to be highly sensitive to the initial molasses concentration. This aspect was not included in the model but is being investigated further.  相似文献

Quantitation and characterization of tau factor in porcine tissues   总被引:2,自引:0,他引:2  
Using a monospecific antibody against brain tau factor purified by affinity chromatography, we have studied the distribution of tau factor or related polypeptides in different cells. The presence of tau in all cell types tested was demonstrated by a radioimmunoassay. Tau factor-related proteins were found in liver, spleen, pancreas, kidney and lung, although at much lower levels than that found in neural cells. In all cases, they copolymerized with tubulin and were heat-resistant. When the distribution of tau factor-related proteins was studied by Western blotting, tau factor antiserum reacted against peptides with an electrophoretic mobility that was similar to those of brain tau factor peptides. Immunofluorescence studies have also been performed with the same antibody to determine the distribution of tau factor-related peptides in PK15 cells. Our results indicated that these peptides were associated to the microtubule network.  相似文献

The kind of fixative and duration of fixation modify the affinity of plant cell structures, as shown by a 10-15 hr impregnation at 70 C in 2% aqueous AgNO2, and a 1-2 hr reduction at room temperature by a 1:1 mixture of 10% formalin and 1% hydroquinone. Cytoplasmic staining was enhanced by fixing in salts of heavy metals, in buffered 6.5% glutaraldehyde, and in 0.5% picric acid. Nuclear staining was prominent after mixtures of glutaraldehyde and hydroquinone, after formalin and pyrogallol, and after acetone, propylene glycol or ether. Nucleolar staining was favored by fixing in 10% formalin, in 5% formalin containing 0.5% hydroquinone, in 50% ethanol containing 0.5% pyrogallol, or in ethylene glycol. Chromosome staining was favored by fixation in 50% acetic or propionic acid, in 2% trichloroacetic acid, and in methanol or ethanol. The best morphological preservations were seen after 50% acetic acid, 6.5% glutaraldehyde, or the 5% formalin-0.5% hydroquinone mixture.  相似文献
