The rice U-box/ARM E3 ubiquitin ligase SPL11 negatively regulates programmed cell death (PCD) and disease resistance, and controls flowering time through interacting with the novel RNA/DNA binding KH domain protein SPIN1. Overexpression of Spin1 causes late flowering in transgenic rice under short-day (SD) and long-day (LD) conditions. In this study, we characterized the function of the RNA-binding and SPIN1-interacting 1 (RBS1) protein in flowering time regulation. Rbs1was identified in a yeast-two-hybrid screen using the full-length Spin1 cDNA as a bait and encodes an RNA binding protein with three RNA recognition motifs. The protein binds RNA in vitro and interacts with SPIN1 in the nucleus. Rbs1 overexpression causes delayed flowering under SD and LD conditions in rice. Expression analyses of flowering marker genes show that Rbs1 overexpression represses the expression of Hd3a under SD and LD conditions. Rbs1 is upregulated in both Spin1 overexpression plants and in the spl11 mutant. Interestingly, Spin1 expression is increased but Spl11 expression is repressed in the Rbs1 overexpression plants. Western blot analysis revealed that the SPIN1 protein level is increased in the Rbs1 overexpression plants and that the RBS1 protein level is also up-regulated in the Spin1 overexpression plants. These results suggest that RBS1 is a new negative regulator of flowering time that itself is positively regulated by SPIN1 but negatively regulated by SPL11 in rice. 相似文献

Background
Autoimmune thyroid disease (AITD) comprises diseases including Hashimoto''s thyroiditis and Graves'' disease, both characterized by reactivity to autoantigens causing, respectively, inflammatory destruction and autoimmune stimulation of the thyroid-stimulating hormone receptor. AITD is the most common thyroid disease and the leading form of autoimmune disease in women. Cytokines are key regulators of the immune and inflammatory responses; therefore, genetic variants at cytokine-encoding genes are potential risk factors for AITD.Methods
Polymorphisms in the IL6-174 G/C (rs1800795), TNFA-308 G/A (rs1800629), IL1B-511 C/T (rs16944), and IFNGR1-56 T/C (rs2234711) genes were assessed in a case-control study comprising 420 Hashimoto''s thyroiditis patients, 111 Graves'' disease patients and 735 unrelated controls from Portugal. Genetic variants were discriminated by real-time PCR using TaqMan SNP genotyping assays.Results
A significant association was found between the allele A in TNFA-308 G/A and Hashimoto''s thyroiditis, both in the dominant (OR = 1.82, CI = 1.37–2.43, p-value = 4.4×10−5) and log-additive (OR = 1.64, CI = 1.28–2.10, p-value = 8.2×10−5) models. The allele C in IL6-174 G/C is also associated with Hashimoto''s thyroiditis, however, only retained significance after multiple testing correction in the log-additive model (OR = 1.28, CI = 1.06–1.54, p-value = 8.9×10−3). The group with Graves'' disease also registered a higher frequency of the allele A in TNFA-308 G/A compared with controls both in the dominant (OR = 1.85, CI = 1.19–2.87, p-value = 7.0×10−3) and log-additive (OR = 1.69, CI = 1.17–2.44, p-value = 6.6×10−3) models. The risk for Hashimoto''s thyroiditis and Graves'' disease increases with the number of risk alleles (OR for two risk alleles is, respectively, 2.27 and 2.59).Conclusions
This study reports significant associations of genetic variants in TNFA and IL6 with the risk for AITD, highlighting the relevance of polymorphisms in inflammation-related genes in the etiopathogenesis of AITD. 相似文献93.

Amongst post‐Li‐ion battery technologies, lithium–sulfur (Li–S) batteries have captured an immense interest as one of the most appealing devices from both the industrial and academia sectors. The replacement of conventional liquid electrolytes with solid polymer electrolytes (SPEs) enables not only a safer use of Li metal (Li°) anodes but also a flexible design in the shape of Li–S batteries. However, the practical implementation of SPEs‐based all‐solid‐state Li–S batteries (ASSLSBs) is largely hindered by the shuttling effect of the polysulfide intermediates and the formation of dendritic Li° during the battery operation. Herein, a fluorine‐free noble salt anion, tricyanomethanide [C(CN)3?, TCM?], is proposed as a Li‐ion conducting salt for ASSLSBs. Compared to the widely used perfluorinated anions {e.g., bis(trifluoromethanesulfonyl)imide anion, [N(SO2CF3)2)]?, TFSI?}, the LiTCM‐based electrolytes show decent ionic conductivity, good thermal stability, and sufficient anodic stability suiting the cell chemistry of ASSLSBs. In particular, the fluorine‐free solid electrolyte interphase layer originating from the decomposition of LiTCM exhibits a good mechanical integrity and Li‐ion conductivity, which allows the LiTCM‐based Li–S cells to be cycled with good rate capability and Coulombic efficiency. The LiTCM‐based electrolytes are believed to be the most promising candidates for building cost‐effective and high energy density ASSLSBs in the near future. 相似文献

Background
The number of copies of the HLA-DRB1 shared epitope, and the minor alleles of the STAT4 rs7574865 and the PTPN22 rs2476601 polymorphisms have all been linked with an increased risk of developing rheumatoid arthritis. In the present study, we investigated the effects of these genetic variants on disease activity and disability in patients with early arthritis.Methodology and Results
We studied 640 patients with early arthritis (76% women; median age, 52 years), recording disease-related variables every 6 months during a 2-year follow-up. HLA-DRB1 alleles were determined by PCR-SSO, while rs7574865 and rs2476601 were genotyped with the Taqman 5′ allelic discrimination assay. Multivariate analysis was performed using generalized estimating equations for repeated measures. After adjusting for confounding variables such as gender, age and ACPA, the TT genotype of rs7574865 in STAT4 was associated with increased disease activity (DAS28) as compared with the GG genotype (β coefficient [95% confidence interval] = 0.42 [0.01–0.83], p = 0.044). Conversely, the presence of the T allele of rs2476601 in PTPN22 was associated with diminished disease activity during follow-up in a dose-dependent manner (CT genotype = −0.27 [−0.56– −0.01], p = 0.042; TT genotype = −0.68 [−1.64– −0.27], p = 0.162). After adjustment for gender, age and disease activity, homozygosity for the T allele of rs7574865 in STAT4 was associated with greater disability as compared with the GG genotype.Conclusions
Our data suggest that patients with early arthritis who are homozygous for the T allele of rs7574865 in STAT4 may develop a more severe form of the disease with increased disease activity and disability. 相似文献96.

Most individuals throughout the Americas are admixed descendants of Native American, European, and African ancestors. Complex historical factors have resulted in varying proportions of ancestral contributions between individuals within and among ethnic groups. We developed a panel of 446 ancestry informative markers (AIMs) optimized to estimate ancestral proportions in individuals and populations throughout Latin America. We used genome-wide data from 953 individuals from diverse African, European, and Native American populations to select AIMs optimized for each of the three main continental populations that form the basis of modern Latin American populations. We selected markers on the basis of locus-specific branch length to be informative, well distributed throughout the genome, capable of being genotyped on widely available commercial platforms, and applicable throughout the Americas by minimizing within-continent heterogeneity. We then validated the panel in samples from four admixed populations by comparing ancestry estimates based on the AIMs panel to estimates based on genome-wide association study (GWAS) data. The panel provided balanced discriminatory power among the three ancestral populations and accurate estimates of individual ancestry proportions (R2 > 0.9 for ancestral components with significant between-subject variance). Finally, we genotyped samples from 18 populations from Latin America using the AIMs panel and estimated variability in ancestry within and between these populations. This panel and its reference genotype information will be useful resources to explore population history of admixture in Latin America and to correct for the potential effects of population stratification in admixed samples in the region. 相似文献

Repetitive aquaculture-related protocols may act as cyclic stressors that induce chronic stress in cultured fish. The sea
bass is particularly sensitive to stressful conditions and the mere presence of humans will disturb feeding behavior. In this
paper, we study whether chronic stress induced by repetition of acute stress protocols affects long-term feeding behavior
and growth performance in sea bass and whether exogenous cortisol may induce stress-like changes in these parameters. We demonstrate
that both chronic stress and dietary cortisol decrease food intake and have a negative effect on feed conversion efficiency,
severely impairing sea bass performance. Both experimental approaches induced changes in the daily feeding activity by lengthening
the active feeding periods. Fish subjected to a cyclic stressor modify their daily feeding pattern in an attempt to avoid
interference with the time of the stressor. The delay in feeding when fish are acutely and repeatedly stressed could be of

The term "monoclonal gammopathy" (MG) includes a group of clonal plasma cell disorders, which show heterogeneous clinical behavior. While multiple myeloma (MM) and plasma cell leukemia (PCL) are incurable malignant diseases, most patients with MG of undetermined significance (MGUS) show an indolent/benign clinical course. Evidence has accumulated which supports the role of the bone marrow microenvironment in MG. Accordingly, the survival, drug-resistance and proliferation of MM cells have been shown to be largely dependent on a supportive microenvironment. Among the different environment-associated parameters, those related to the status/activity of the immune system are particularly relevant. This review focuses on the different ways clonal plasma cells (PC) interact with the immune system in different models of MG, to characterize crucial events in the development and progression of MG. Recurrent anthropogenic and natural perturbations, resource limitations and heterogeneous environments contribute to the maintenance of a remarkable biodiversity in Mediterranean plant communities. Yet, the essential mechanisms of community assembly in these systems remain largely unexplored. In the current paper we investigate the coexistence of Aleppo pine (Pinus halepensis Mill.) and Holm oak (Quercus ilex L.), two of the most widely distributed species in the Iberian Peninsula, in relation to gradients in water availability and disturbance. A spatial model of landscape forest dynamics was implemented, calibrated with experimental data, with stands arranged on a heterogeneous lattice and coupled by dispersal. It was found that pine population persistence in the landscape can be explained independently by a competition-colonization tradeoff (in mesic homogeneous environments) and by a tradeoff between shade and drought tolerance (in heterogeneous low disturbed environments). Both mechanisms reinforce mutually to maintain a shifting mosaic of both taxa along disturbance and aridity gradients. This view is consistent with palynological, historical and forest inventory records. In turn, equilibrium theories of vegetation dynamics (phytosociological), that neglect the role of heterogeneity and disturbances may be inadequate for Mediterranean forests, and have been shown to result in mismanagement practices. We claim that biologically informed models of forest dynamics are desperately needed as diagnosis tools for sustainable forest management. 相似文献
