Summary Explants from petioles, folioles or hypocotyls ofOnonis natrix have been used for calli initiation. Hypocotyls inoculated on MS medium supplemented with 2% sucrose and 0.5 mg.1–1 2,4-D / 1 mg.1–1 Kin showed to be the best primary explant. Cell suspension cultures were established in MS basal medium supplemented with 2% sucrose, 0.5 mg.1–1 NAA or 2,4-D and 1 mg.1–1 Kin. Different subculturing periods, inoculum density, hormonal supplementation and sucrose concentration were assayed in order to obtain the best culture growth conditions. The optimal conditions were achieved with cultures initiated with 40 g.1–1 of initial inoculum, growing in MS basal medium supplemented with 4% sucrose, 0.5 mg.1–1 NAA and 1 mg.1–1 Kin subcultured every twelve days. Under these experimental conditions, the cultures showed a doubling time of 36.3 hours. 相似文献

In the rural Mediterranean landscape, mosaics of patches of sclerophyllous scrubland and semi-natural grasslands are frequent. The plant communities of these patches, which are physiognomically easy to recognise, are very heterogeneous. The objective of this paper is to determine whether the patches of scrub-grassland represent an integrated response unit of the vegetation with regard to the physical environment (climatic, geographical and edaphic factors) and human use, or whether, on the contrary, this is an independent response. In order to do this a total of 50 sampling sites where scrubland and grassland patches were in contact were studied along a 370 km E-W mesoclimatic gradient from central Spain to Portugal (Iberian Peninsula). Two distinct zones, the east and the west halves of the study area, were identified according to the plant communities. Within each of these zones, each type of patch responded to the previously mentioned factors, differentially and independently. This determined a general lack of inter-community association or floristic correlation between scrub and grassland plant communities in contact. The spatial association is a random process probably related to the particular human management realised on every scrubland and grassland patch. The scrub–grassland pattern, which is characteristic of the Mediterranean landscape, does not represent an integrated response unit of the vegetation to a given environment, but rather the sum of the independent responses to the environment of the two patches in contact.Nomenclature for taxa: T. Introduction:
The detection of the human papillomavirus (HPV) through the combination of the HPV test and other techniques such as cytology has impacted the detection and timely treatment of lesions associated with cervical cancer.Objective:
To estimate the budgetary impact of the strategy of early detection of HPV with DNA test genotyping with reflex cytology versus conventional cytology in women aged 30 to 65 years attending the cervical cancer screening program at a health benefit managing entity in Colombia.Materials and methods:
Using a decision tree and a Markov model, the clinical implications and direct costs of screening, diagnosis, and treatment were estimated in a cohort of women. The analysis considered two screening cycles and their annual costs. The data on the prevalence of clinical results and the costs were taken from the health managing entity. The information on the progression, persistence, and regression of the health states were taken from the ATHENA study.Results:
The screening scheme with the HPV test, genotyping, and reflex cytology compared to conventional cytology was cost-saving. The average cost per screening cycle with the HPV test was estimated at COP $ 129,201,363 and with cytology at COP $ 186,309,952, i.e., a saving of COP $ 57,108,589 (30.7%).Conclusion:

Input–output tables (IOTs) provide a relevant picture of economic structure as they represent the composition and interindustry relationships of an economy. The technical coefficients matrix (A matrix) is considered to capture the technological status of an economy; so, it is of special relevance for the evaluation of long-term, structural transformations, such as sustainability transitions in integrated assessment models (IAMs). The A matrix has typically been considered either static or exogenous. Endogenous structural change has rarely been applied to models. The objective of this paper is to analyze energy intensity, a widely used variable in IAMs, and its role as a driver of structural change. We therefore identify the most relevant technical coefficients in the IOTs time series and estimate an econometric model based on the energy intensity of five different final end-use energy sources. The results of this analysis show that energy intensity has a significant influence on the evolution of the A matrix and should therefore be taken into consideration when analyzing endogenous structural change in models. 相似文献

Background
Previous studies have shown that prenatal exposure to the mutagen N-ethyl-N-nitrosourea (ENU), a N-nitroso compound (NOC) found in the environment, disrupts developmental neurogenesis and alters memory formation. Previously, we showed that postnatal ENU treatment induced lasting deficits in proliferation of neural progenitors in the subventricular zone (SVZ), the main neurogenic region in the adult mouse brain. The present study is aimed to examine, in mice exposed to ENU, both the structural features of adult neurogenic sites, incorporating the dentate gyrus (DG), and the behavioral performance in tasks sensitive to manipulations of adult neurogenesis.Methodology/Principal Findings
2-month old mice received 5 doses of ENU and were sacrificed 45 days after treatment. Then, an ultrastructural analysis of the SVZ and DG was performed to determine cellular composition in these regions, confirming a significant alteration. After bromodeoxyuridine injections, an S-phase exogenous marker, the immunohistochemical analysis revealed a deficit in proliferation and a decreased recruitment of newly generated cells in neurogenic areas of ENU-treated animals. Behavioral effects were also detected after ENU-exposure, observing impairment in odor discrimination task (habituation-dishabituation test) and a deficit in spatial memory (Barnes maze performance), two functions primarily related to the SVZ and the DG regions, respectively.Conclusions/Significance
The results demonstrate that postnatal exposure to ENU produces severe disruption of adult neurogenesis in the SVZ and DG, as well as strong behavioral impairments.
