While trying to achieve their nutritional requirements, foraging herbivores face the costs of plant defenses, such as toxins. Teasing apart the costs and benefits of various chemical constituents in plants is difficult because their chemical defenses and nutrient concentrations often co-vary. We used an approach derived from predator–prey studies to quantitatively compare the foraging response of a free-ranging mammalian herbivore, the swamp wallaby (Wallabia bicolor), through three feeding trials with artificial diets that differed in their concentrations of (1) the terpene 1,8-cineole, (2) primary constituents (including nitrogen and fiber), and (3) both the terpene and the primary constituents. Applying the giving-up density (GUD) framework, we demonstrated that the foraging cost of food patches increases with higher dietary cineole concentration and decreases with higher dietary nutrient concentration. The effect of combined differences in nutrients and cineole concentrations on GUD was interactive, and high nutrient food required more cineole to achieve the same patch value as low nutrient food. Our results indicate that swamp wallabies equate low nutrient, poorly defended food with high nutrient, highly defended food, providing two contrasting diets with similar cost–benefit outcomes. This behavior suggests that equal concentrations of chemical defenses provide nutrient-poor plants with relatively greater protection as nutrient-rich plants. Nutrient-rich plants may therefore face the exacerbated problem of being preferred by herbivores and therefore need to produce more defense compounds to achieve the same level of defense as nutrient-poor plants. Two-color spatio-temporal image cross-correlation spectroscopy (STICCS) is a new, to our knowledge, image analysis method that calculates space-time autocorrelation and cross-correlation functions from fluorescence intensity fluctuations. STICCS generates cellular flow and diffusion maps that reveal interactions and cotransport of two distinct molecular species labeled with different fluorophores. Here we use computer simulations to map the capabilities and limitations of STICCS for measurements in complex heterogeneous environments containing micro- and macrostructures. We then use STICCS to analyze the co-flux of adhesion components in migrating cells imaged using total internal reflection fluorescence microscopy. The data reveal a robust, time-dependent co-fluxing of certain integrins and paxillin in adhesions in protrusions when they pause, and in adhesions that are sliding and disassembling, demonstrating that the molecules in these adhesions move as a complex. In these regions, both α6β1- or αLβ2-integrins, expressed in CHO.B2 cells, co-flux with paxillin; an analogous cotransport was seen for α6β1-integrin and α-actinin in U2OS. This contrasts with the behavior of the α5β1-integrin and paxillin, which do not co-flux. The distribution range of fishes along an upstream–downstream gradient within a river basin is determined by the ecological requirements of each fish species. This differential pattern of distribution may suggest the occurrence of fish zones, where different functional guilds would prevail. The earliest concept of fish zonation highlighted the preferential position of several species in European rivers. Nowadays, there is a consensus that for a broad application of this concept, it is necessary to consider the division in functional groups along the downstream profile as related to reproductive, feed and population dynamics traits. However, for a number of tropical systems, such information does not exist. In this case, it is possible to use the fish family as a surrogate metric, by assuming that it fully covers common aspects of morphology, feeding, and behavior. Using this surrogate metric, we analyzed the distribution range of fish species along an elevation gradient in a typical Atlantic Forest basin in southeast Brazil. Our objective was to delimit fish zones based on the representativeness of some dominant fish families. Between 2006 and 2008, we sampled 42 sites from the estuary to the headwater creeks in elevations that varied from 3 to 783 m. The data were explored using the species (or family) occurrence, relative numerical abundance (N′), relative biomass (B′), and the importance index (IP). We applied the non-metric multidimensional scaling (NMDS) to ordinate the sampling sites using the Jaccard distance calculated on the species occurrence matrix. Data on family importance were superimposed to the NMDS to check how the distribution and the abundance of the families were correlated to the ordination. We caught 64 species distributed among 25 fish families. Based on the dominance of some families, we propose that the fish assemblage in the Itanhaém river basin can be organized in four fish zones (FZ) and three transitory zones (TZ), which replace each other along the longitudinal gradient. The marine/estuarine fishes dominated in FZ1 and TZ1. The freshwater fishes were dominated by Characidae (FZ2), Heptapteridae (FZ3), and Gymnotidae (FZ4). TZ2 was equally dominated by Characidae, Loricariidae, and Heptapteridae. The distinct assemblage composition along the FZ’s may relate to complex interactions involving seasonal patterns of variation in salinity levels (next to estuary), slope, presence of downstream barriers, and availability of food resources. We expect that the patterns observed in our study will also be presented in other coastal Atlantic Forest basins in Southeast Brazil. 相似文献

The use of heat shock protein 90 (Hsp90) inhibitors is an attractive antineoplastic therapy. We wanted to compare the effects of the benzoquinone 17-allylamino-17-demethoxygeldanamycin (17-AAG, tanespimycin) and the novel isoxazole resorcinol–based Hsp90 inhibitor NVP-AUY922 in a panel of pancreatic and colorectal carcinoma cell lines and in colorectal primary cultures derived from tumors excised to patients. PANC-1, CFPAC-1, and Caco-2 cells were intrinsically resistant to 17-AAG but sensitive to NVP-AUY922. Other cellular models were sensitive to both inhibitors. Human epidermal growth factor receptor receptors and their downstream signaling pathways were downregulated in susceptible cellular models, and concurrently, Hsp70 was induced. Intrinsic resistance to 17-AAG did not correlate with expression of ATP-binding cassette transporters involved in multidrug resistance. Some 17-AAG-resistant, NVP-AUY922–sensitive cell lines lacked NAD(P)H:quinone oxidoreductase 1 (NQO1) enzyme and activity. However, colorectal LoVo cells still responded to both drugs in spite of having undetectable levels and activity of NQO1. Pharmacological and biologic inhibition of NQO1 did not confer resistance to 17-AAG in sensitive cell lines. Therefore, even though 17-AAG sensitivity is related to NQO1 protein levels and enzymatic activity, the absence of NQO1 does not necessarily convey resistance to 17-AAG in these cellular models. Moreover, NVP-AUY922 does not require NQO1 for its action and is a more potent inhibitor than 17-AAG in these cells. More importantly, we show in this report that NVP-AUY922 potentiates the inhibitory effects of chemotherapeutic agents, such as gemcitabine or oxaliplatin, and other drugs that are currently being evaluated in clinical trials as antitumor agents. 相似文献

Aim To determine if it is possible to generate analytically derived regionalizations for multiple groups of European plants and animals and to explore potential influences on the regions for each taxonomic group. Location Europe. Methods We subjected range maps of trees, butterflies, reptiles, amphibians, birds and mammals to k‐means clustering followed by v‐fold cross‐validation to determine the pattern and number of regions (clusters). We then used the mean range sizes of species in each group as a correlate of the number of regions obtained for each taxon, and climate and species richness gradients as correlates of the spatial arrangement of the group‐specific regions. We also included the pattern of tree clusters as a predictor of animal clusters in order to test the ‘habitat templet’ concept as an explanation of animal distribution patterns. Results Spatially coherent clusters were found for all groups. The number of regions ranged from three to eight and was strongly associated with the mean range sizes of the species in each taxon. The cluster patterns of all groups were associated with various combinations of climate, underlying species richness gradients and, in the case of animals, the arrangement of tree clusters, although the rankings of the correlates differed among groups. In four of five groups the tree pattern was the strongest single predictor of the animal cluster patterns. Main conclusions Despite a long history of human disturbance and habitat modification, the European biota retains a discernable biogeographic structure. The primary driver appears to be aspects of climate related to water–energy balance, which also influence richness gradients. For many animals, the underlying habitat structure, as measured by tree distributions, appears to have a strong influence on their biogeographic structure, highlighting the need to preserve natural forest formations if we want to preserve the historical signal found in geographic distributions. 相似文献

One difficulty in the conservation of endangered wildlife is the lack of reliable information on its status. This lack of
knowledge can often be attributed to financial and logistic constraints as well as the lack of trained personnel to collect
data. We test a simple method to study bears in the southern Balkans by inspecting power poles, which are used by bears for
marking and rubbing purposes. We created a network of barbed-wire fitted poles for the collection of hair samples, evenly
distributed throughout six study areas. During 87 sampling sessions in the main study area, we collected 191 samples and identified
six microsatellite loci that were variable enough for individual bear identification. The most and best-quality hair samples
were collected during the mating period, and DNA was most successfully extracted from samples remaining <4 weeks in the field.
In the six study areas, we identified 47 bears. An advantage of using power poles for hair sampling is their availability
and accessibility; no bait is required, and the network can be easily set up. A drawback may be an unequal capture probability
of sex and age classes of bears. Despite this limitation, using power poles proved to be a simple and cheap method for the
noninvasive genetic study of bears that did not require any prior knowledge on habitat use and activity patterns. The method
is suitable for large-scale surveys to estimate distribution and relative densities of bears and could also be applied for

Acca sellowiana (Berg.) Burr. is a native Myrtaceae from southern Brazil and Uruguay, now the subject of a domestication and breeding program.
Biotechnological tools have been used to assist in this program. The establishment of a reliable protocol of somatic embryogenesis
has been pursued, with a view to capturing and fixing genetic gains. The rationale behind this work relies on the fact that
deepening comprehension of the general metabolism of zygotic embryogenesis may certainly improve the protocol for somatic
embryogenesis. Thus, in the present work we studied the accumulation of protein, total sugars, starch, amino acids, polyamines
(PAs), IAA and ABA, in different stages of A. sellowiana zygotic embryogenesis. Starch is the predominant storage compound during zygotic embryo development. Increased synthesis
of amino acids in the cotyledonary stage, mainly of asparagine, was observed throughout development. Total free PAs showed
increased synthesis, whereas total conjugated PAs were mainly observed in the early developmental stages. IAA decreased and
ABA increased with the progression from early to late embryogenesis. Besides providing basic information on the morphophysiological
and biochemical changes of zygotic embryogenesis, the results here obtained may provide adequate strategies towards the modulation
of somatic embryogenesis in this species as well as in other woody angiosperms. 相似文献

Plants orient their growth depending on directional stimuli such as light and gravity, in a process known as tropic response. Tropisms result from asymmetrical accumulation of auxin across the responding organ relative to the direction of the stimulus, which causes differential growth rates on both sides of the organ. Here, we show that gibberellins (GAs) attenuate the gravitropic reorientation of stimulated hypocotyls of dark-grown Arabidopsis (Arabidopsis thaliana) seedlings. We show that the modulation occurs through induction of the expression of the negative regulator of auxin signaling INDOLE-3-ACETIC ACID INDUCIBLE19/MASSUGU2. The biological significance of this regulatory mechanism involving GAs and auxin seems to be the maintenance of a high degree of flexibility in tropic responses. This notion is further supported by observations that GA-deficient seedlings showed a much lower variance in the response to gravity compared to wild-type seedlings and that the attenuation of gravitropism by GAs resulted in an increased phototropic response. This suggests that the interplay between auxin and GAs may be particularly important for plant orientation under competing tropic stimuli. 相似文献
