Using a human placenta cDNA library, we cloned a novel member belonging to the scavenger receptor family. Complementary DNA of this clone encodes a type II transmembranous glycoprotein containing a collagen-like domain, which are typical structural characteristics of scavenger receptor class A. This protein also contains a C-type lectin/carbohydrate recognition domain (C-type CRD) located at the C-terminus. We designated this as Scavenger Receptor with C-type Lectin (SRCL) type I. We also isolated human SRCL type II, which lacks the C-type CRD. Northern blot analysis revealed that hSRCL type I and type II mRNAs are abundantly expressed in adult human tissues. When hSRCL type I and type II were expressed in CHO-K1 cells, they were localized in the plasma membrane forming clusters on the surface. Ligand-binding studies of CHO-K1 cells expressing hSRCL type I and type II demonstrated their specific binding capacity in Escherichia coli and Staphylococcus aureus. These results indicate that hSRCL is a novel bacteria-binding receptor containing a C-type CRD and this receptor may play an important role in host defense. 相似文献

From the reaction between dihydroxoplatinum(II) and l-ascorbic acid, two types of platinum(II) ascorbate complexes were obtained and structurally characterized with ethylenediamine (en), N,N-dimethylethylenediamine (dmen) and N,N,N′-trimethylethylenediamine (trimen) as stabilizing ligands. In [Pt(en)(asc-C,O)] (1), [Pt(dmen)(asc-C,O)] (2) and [Pt(trimen)(asc-C,O)] (4), the ascorbate dianion forms a five-membered chelate ring, coordinating to the Pt(II) ion at the 2-carbon and the 5-oxygen atoms (C,O-chelate). From the same mother solution, crystals of [Pt(trimen)(asc-O,O′)] (3) were obtained during the precipitation of 4; in 3 the ascorbate is bound to the Pt at the 2- and 3-oxygen atoms (O,O′-chelate). Compounds 3 and 4 are the first well-characterized linkage isomers among the transition-metal ascorbate complexes. The O,O′-chelated 3 slowly changes to the C,O-chelated 4 in an aqueous solution. Bulkiness of the stabilizing ligand, i.e. en, dmen and trimen has an influence on the formation of the C,O-chelated species, 1, 2 and 4. 相似文献

TNF-alpha, a potent proinflammatory cytokine, is synthesized as a membrane-anchored precursor and proteolytically released from cells. Soluble TNF is the primary mediator of pathologies such as rheumatoid arthritis, Crohn's disease, and endotoxin shock. The TNF-alpha converting enzyme (TACE), a disintegrin and metalloprotease 17 (ADAM17), has emerged as the best candidate TNF sheddase, but other proteinases can also release TNF. Because TACE-deficient mice die shortly after birth, we generated conditional TACE-deficient mice to address whether TACE is the relevant sheddase for TNF in adult mice. In this study, we report that TACE inactivation in myeloid cells or temporal inactivation at 6 wk offers strong protection from endotoxin shock lethality in mice by preventing increased TNF serum levels. These findings corroborate that TACE is the major endotoxin-stimulated TNF sheddase in mouse myeloid cells in vivo, thereby further validating TACE as a principal target for the treatment of TNF-dependent pathologies. 相似文献

POEM, also known as nephronectin, is an extracellular matrix protein considered to be a positive regulator of osteoblast differentiation. In the present study, we found that tumor necrosis factor-α (TNF-α), a key regulator of bone matrix properties and composition that also inhibits terminal osteoblast differentiation, strongly inhibited POEM expression in the mouse osteoblastic cell line MC3T3-E1. TNF-α-induced down-regulation of POEM gene expression occurred in both time- and dose-dependent manners through the nuclear factor kappa B (NF-κB) pathway. In addition, expressions of marker genes in differentiated osteoblasts were down-regulated by TNF-α in a manner consistent with our findings for POEM, while over-expression of POEM recovered TNF-α-induced inhibition of osteoblast differentiation. These results suggest that TNF-α inhibits POEM expression through the NF-κB signaling pathway and down-regulation of POEM influences the inhibition of osteoblast differentiation by TNF-α. 相似文献

The amount of quaternary ammonium ion (Bu4N+), which is believed not to be bound to general carboxylpolyelectrolytes, bound to poly(iso BVE-co-MA) was estimated by conductivity measurements. In the region of the density of polyion charge in which the polyions are thought to take a free drain'ng conformation, it has been confirmed that the activity coefficient of Bu4N+ ions is less than 0.5 in the presence of a small amount of Bu4NCl, showing that the force between the counterions and the polyion is probably due to the hydrophobic interaction. The growth-retarded (grt) mouse has an autosomal recessive, fetal-onset, severe thyroid hypoplasia related to TSH hyporesponsiveness. Through genetic mapping and complementation experiments, we show that grt is a missense mutation of a highly conserved region of the tyrosylprotein sulfotransferase 2 (Tpst2) gene, encoding one of the two Tpst genes implicated in posttranslational tyrosine O-sulfation. We present evidence that the grt mutation leads to a loss of TPST2 activity, and TPST2 isoform has a high degree of substrate preference for TSH receptor (TSHR). The expression of TPST2 can restore TSH-TSHR-mediated cAMP production in fibroblasts derived from grt mice. Therefore, we propose that the tyrosine sulfation of TSHR by TPST2 is crucial for TSH signaling and resultant thyroid gland function. 相似文献
