Persistent infection with hepatitis C virus (HCV) is a major cause of liver diseases such as chronic hepatitis, liver cirrhosis, and hepatocellular carcinoma. Here we report that inhibition of heat shock protein 90 (Hsp90) is highly effective in suppressing HCV genome replication. In HCV replicon cells, HCV replication was reduced by Hsp90 inhibitors and by knockdown of endogenous Hsp90 expression mediated by small-interfering RNA (siRNA). The suppression of HCV replication by an Hsp90 inhibitor was prevented by transfection with Hsp90 expression vector. We also tested the anti-HCV effect of Hsp90 inhibition in HCV-infected chimeric mice with humanized liver. Combined administration of an Hsp90 inhibitor and polyethylene glycol-conjugated interferon (PEG-IFN) was more effective in reducing HCV genome RNA levels in serum than was PEG-IFN monotherapy. These results suggest that inhibition of Hsp90 could provide a new therapeutic approach to HCV infection.  相似文献

Most human neurodegenerative diseases are sporadic and appear later in life. Aging and neurodegeneration are closely associated, and recent investigations reveal that endoplasmic reticulum (ER) stress is involved in the progression of these features. Immunoglobulin heavy chain-binding protein (BiP) is an ER chaperone that is central to ER functions. We produced knock-in mice expressing a mutant BiP that lacked the retrieval sequence to elucidate the effect of a functional defect in an ER chaperone in multicellular organisms. The homozygous mutant BiP mice died within several hours after birth because of respiratory failure with an impaired biosynthesis of pulmonary surfactant by alveolar type II cells. The heterozygous mutant BiP mice grew up to be apparently normal adults, although some of them revealed motor disabilities as they aged. Here, we report that the synthesis of a mitochondrial protein, pyrroline-5-carboxylate reductase 1 (PYCR1), is enhanced in the brains of homozygous mutant BiP mice. We performed a two-dimensional gel analysis followed by liquid chromatography-tandem mass spectrometry. PYCR1 was identified as one of the enhanced proteins. We also found that sublethal ER stress caused by tunicamycin treatment induced the synthesis of PYCR1 in murine fibroblasts. PYCR1 has been shown to be related to the aging process. Mutations in the PYCR1 gene cause cutis laxa with progeroid features and mental retardation. These findings suggest a pathophysiological interaction between ER stress and a mitochondrial function in aging.  相似文献

Summary The relationships between the amounts of foliage and heights of trees were studied for the dominant understory tree species, including three evergreen and three deciduous species, in a secondary forest of Chamaecyparis obtusa Endl. The relationships showed two phases: leaf increasing and stationary phases. In the leaf-increasing phase, the height growth allowed these species to expand the canopy by increasing the number of leaves. In the stationary phase, the number of leaves was relatively constant number irrespective of tree height from 160 to 400 cm. The number of leaves in the stationary phase represents the maximum number of leaves that can be supported by trees under shady conditions. From the analyses of vertical distributions of leaves in six species, mono- and multi-layer foliage distributions were detected. Two evergreen species, Eurya japonica and Cleyera japonica, showed multi-layer foliage distributions, whereas three deciduous species, Lyonia ovalifolia, Rhododendron reticulatum and Vaccinium hirtum, and one evergreen species, Pieris japonica, showed mono-layer foliage distributions. The relationships between the weights of non-photosynthetic and photosynthetic organs of the six species were examined. The proportion of non-photosynthetic organs increased with tree height. The understory species attained the stationary phase and were maintained by minimizing their investment in non-photosynthetic organs, i.e. their height growth was arrested by the shady conditions under the crown trees.  相似文献

A novel NAD(P)H-quinone oxidoreductase (NQR) was isolated fromthe cyanobacterium Synechocystis PCC6803 by ion-exchange, affinityand gel-filtration chro-matographies. Isolated NQR was foundto be a drgA gene product that was a homodimer composed of 23-kDasub-units. It showed NAD(P)H-plastoquinone oxidoreductase activitywith Km values for NADPH and NADH of 12 and 48 µM respectively.The activity was inhibited by thiol-modifying reagents, butnot by rotenone, amobarbital, salicylhydroxamic acid, dicumarol,flavone, or diphenylene-iodonium chloride. Therefore, the Cys-147residue is probably involved in the catalytic reaction. The Tritium (T) uptake method for detecting hydrogenase (Hase) was applied to measure the Hase activity of aerobic nitrogen-fixing bacterium Azotobacter agile. The cell-free extract of this bacterium contains the ATP-stimulated T-uptake activity, and this activity was separated from the nitrogenase activity. In the supernatant obtained by centrifugation at 20,000 × g for 30 min, this ATP-stimulated T-uptake activity existed mainly in large molecular weight fraction and was distributed to precipitate at 184,000 × g for 1 hr. After this ultra-centrifugation, the distribution patterns of methylene blue (MB) reduction and T-uptake activities were significantly different from each other, and MB reduction activity remained much more in the supernatant. The Hase activity detected by both T-uptake and MB reduction was mainly in the particle fraction precipitated at 20,000 × g for 30 min from the cell-free extract. When the activities of the praticle fraction were solubilized with Triton X–100, the ATP-stimulated T-uptake activity was effectively solubilized. These results imply that the cell-free extract of Azotobacter agile contained some different kinds of hydrogenases which catalyzed MB reduction, T-uptake and ATP-stimulated T-uptake activities at different intensities from each other.  相似文献

Acute exacerbation of idiopathic pulmonary fibrosis (AE-IPF) is characterized by severe worsening dyspnea of unknown etiology and high mortality without effective treatment. Recently, direct hemoperfusion with polymyxin B (PMX)-immobilized fiber cartridge (PMX-DHP) has been reported to improve pulmonary oxygenation and survival in patients with AE-IPF although its mechanism of action remains unknown. To gain insights into the pathobiology of AE-IPF through the beneficial effects of PMX-DHP, we analyzed the profile of cytokines adsorbed onto PMX-fibers used in 9 AE-IPF patients. In addition, the sera of these AE-IPF patients collected immediately before and after PMX-DHP, 9 stable IPF patients and 8 healthy individuals were also analyzed. The serum levels of cytokines including IL-9, IL-12, IL-17, PDGF and VEGF were significantly decreased immediately after PMX-DHP (P < 0.02), and VEGF and IL-12 were most prominently reduced. In addition to PDGF and VEGF, IL-1β, IL-1ra, IL-8, IL-23, FGF basic, GM-CSF, IP-10, RANTES and TGF-β were eluted from used PMX-fibers. Interestingly, improved pulmonary oxygenation after PMX-DHP was correlated well with the quantities of eluted VEGF. These results suggest that adsorption of proinflammatory, profibrotic and proangiogenic cytokines onto PMX-fibers is one of the mechanisms of action of PMX-DHP in AE-IPF. Notably, removal of VEGF by PMX-DHP may contribute to the rapid improvement in oxygenation by suppressing vascular permeability in the lung.  相似文献

In acute CCL4 intoxication of rats significantly increased activities of hepatic low-Km hexokinases, glucose-6-phosphate dehydrogenase, phosphofructokinase, aldolase A and pyruvate kinase M2 with concurrently decreased activities of glucokinase, glucose-6-phosphatase, fructose-1,6-diphosphatase, aldolase B and pyruvate kinase L were observed. The resulting enzyme pattern was apparently different from that in dietary induction. Principal component analysis revealed that the degree of enzyme deviation in the injured liver was much greater than that in the regenerating liver after partial hepatectomy and was closer to that in fetal liver or hepatoma tissue.  相似文献
