Effects of C2H4 and CO2 on respiration of pre-soaked upper cocklebur(Xanthium pennsylvanicum Wallr.) seeds during a pre-germinationperiod were examined in relation to effects of the two gaseson germination. At 33?C, cocklebur seed germination was greatlystimulated. This high temperature-stimulated germination wasseverely inhibited by C2H4, but not by CO2, although both gasesstimulated germination at 23?C. C2H4 promoted seed respirationat 23?C, but its promotive effect decreases with increasingtemperature and disappeared at about 35?C, while CO2 stimulatedrespiration regardless of temperature. CO2 augmented the operationof the CN-sensitive, cytochrome path (CP) regardless of temperature,resulting in an increase in the ratio of the CP flux to a CN-resistant,alternative path (AP) flux. On the other hand, C2H4 augmentedthe operation of both paths, particularly of the AP, at 23?C,where it promoted germination. However, at 33?C where germinationis suppressed by C2H4, C2H4 preferentially stimulated respirationvia the AP, thus leading to an extremely high ratio of AP toCP. The inhibitory effect of C2H4 on germination at 33?C disappearedcompletely in enriched O2, under which conditions CP is knownto be augmented. At 23?C, CO2 and C2H4 acted independently incontrolling seed respiration, but they were antagonistic at33?C. The independent action appeared when the AP flux was verylow relative to the CP flux, while the antagonism appeared whenthe AP flux had risen. This differential action of the two gasesat different temperatures was also observed in the ATP level,adenylate pool size and energy charge of the axial tissues.These results suggest that the germination-controlling actionsof both CO2 and C2H4 are fundamentally manifested through themodification of respiratory systems. However, the germination-inhibitingeffect of C2H4 at 33 ?C was not removed by inhibitors of AP,and there was little difference in the adenylate compounds betweenthe C2H4-treated and non-treated seeds at 33?C. Many clinically useful pharmaceuticals are semi-synthesized from natural products produced by actinobacteria and fungi. The synthetic protocols usually contain many complicated reaction steps and thereby result in low yields and high costs. It is therefore important to breed microorganisms that produce a compound most suitable for chemical synthesis. For a long time, desirable mutants have been obtained by random mutagenesis and mass screening. However, these mutants sometimes show unfavorable phenotypes such as low viability and low productivity of the desired compound. Fusicoccin (FC) A is a diterpene glucoside produced by the fungus Phomopsis amygdali. Both FC and the structurally-related cotylenin A (CN) have phytohormone-like activity. However, only CN exhibits anti-cancer activity. Since the CN producer lost its ability to proliferate during preservation, a study on the relationship between structure and activity was carried out, and elimination of the hydroxyl group at position 12 of FC was essential to mimic the CN-like activity. Based on detailed dissection of the biosynthetic machinery, we constructed a mutant producing a compound without a hydroxyl group at position 12 by gene-disruption. The mutant produced this compound as a sole metabolite, which can be easily and efficiently converted into an anti-cancer drug, and its productivity was equivalent to the sum of FC-related compounds produced by the parental strain. Our strategy would be applicable to development of pharmaceuticals that are semi-synthesized from fungal metabolites.  相似文献

Here we report the first application of amino acid-type selective (AATS) isotope labeling of a recombinant protein secreted by Brevibacillus choshinensis for a nuclear magnetic resonance (NMR) study. To prepare the 15N-AATS-labeled protein, the transformed B. choshinensis was cultured in 15N-labeled amino acid-containing C.H.L. medium, which is commonly used in the Escherichia coli expression system. The analyses of the 1H-15N heteronuclear single quantum coherence (HSQC) spectra of the secreted proteins with a 15N-labeled amino acid demonstrated that alanine, arginine, asparagine, cysteine, glutamine, histidine, lysine, methionine, and valine are suitable for selective labeling, although acidic and aromatic amino acids are not suitable. The 15N labeling for glycine, isoleucine, leucine, serine, and threonine resulted in scrambling to specific amino acids. These results indicate that the B. choshinensis expression system is an alternative tool for AATS labeling of recombinant proteins, especially secretory proteins, for NMR analyses.  相似文献

The high-affinity cation-binding sites of bacteriorhodopsin (bR) were examined by solid-state 13C NMR of samples labeled with [3-13C]Ala and [1-13C]Val. We found that the 13C NMR spectra of two kinds of blue membranes, deionized (pH 4) and acid blue at pH 1.2, were very similar and different from that of the native purple membrane. This suggested that when the surface pH is lowered, either by removal of cations or by lowering the bulk pH, substantial change is induced in the secondary structure of the protein. Partial replacement of the bound cations with Na+, Ca2+, or Mn2+ produced additional spectral changes in the 13C NMR spectra. The following conclusions were made. First, there are high-affinity cation-binding sites in both the extracellular and the cytoplasmic regions, presumably near the surface, and one of the preferred cation-binding sites is located at the loop between the helix F and G (F-G loop) near Ala196, consistent with the 3D structure of bR from x-ray diffraction and cryoelectron microscopy. Second, the bound cations undergo rather rapid exchange (with a lifetime shorter than 3 ms) among various types of cation-binding sites. As expected from the location of one of the binding sites, cation binding induced conformational alteration of the F-G interhelical loop.  相似文献

Excitable cells in the petiole of Mimosa pudica were locatedby microelectrode technique and stained with Procion YellowMx4R which was previously filled in the electrode and injectediontophoretically into the cells. Microscopic observations ofsections of the stained petioles revealed that protoxylem parenchymacells and narrow phloem cells were excitable. The protoxylemlocalized just inside the metaxylem was composed almost entirelyof the parenchyma cells which were 106.3±5.2 µmlong (mean±EM, n=15) and 14.2±0.6 µm indiameter (n =33). The excitable phloem cells were 76.4±4.1µm long (n=7) and 7.0±0.3 pan in diameter (n=37)and were thought to be companion cells or narrow parenchymacells or both. Amplitudes of action potentials recorded fromthe petiolar surface had a linear relation to those from theexcitable cells in the same petiole. The effect of human lung conditioned medium (HLCM) on the DNA synthesis in cultured human alveolar macrophages (HAM) was evaluated. The medium from human lung cultured for 3 days (3d-HLCM) promoted the DNA synthesis as well as the recombinant human GM-CSF does. On the other hand, that cultured for six days (6d-HLCM) did not promote the DNA synthesis but strongly suppressed GM-CSF induced DNA synthesis in HAM. This growth inhibitory effect was also observed when several macrophage like cell lines were cultured with 6d-HLCM. Partial purification of an inhibitory factor on gel permeation HPLC revealed two distinct peaks of activity with molecular weights of 38 kd and 110 kd.  相似文献

The effect of propyl-gallate (PG) and benzohydroxamic acid (BHAM),inhibitors of cyanide-resistant, alternative respiration path(AP), on germination were examined using after-ripened upperand lower cocklebur (Xanthium pennsylvanicum Wallr.) seeds pre-soakedat 23?C for various periods. Germination was strongly suppressedby PG or BHAM at concentration above 2 mM. However, germinationwas enhanced by low concentrations of PG or BHAM (0.25 or 0.5mM) which reduced some portions of AP operation. Similarly,the high temperature-induced germination of pre-soaked upperseeds was promoted by the same low concentration range of PGor BHAM, in which PG and BHAM were effective only when appliedat the start of high temperature incubation. The inhibitionof germination by C2H4 at high temperature occurred only whenseeds were exposed to C2H4 during the earlier period of hightemperature incubation, and delayed application tended to promotetheir germination, although most of germinated seeds did notexhibit the normal germination behaviour of predominant radicleprotrusion. If the upper seeds had been subjected to a short-timepre-soaking, the inhibition of high temperature-induced germinationby C2H4 was prevented by the low concentrations of PG or BHAM,although the germination restored was mostly an abnormal, predominantlycotyldonary growth, suggesting that the germination inhibitionby C2H4 may be involved in some step of axial growth or in thegrowth of some specific axial zone. The lower concentrationsof PG or BHAM were promotive to the axial growth even at 33?C.Based on these results, the involvement of AP in cocklebur seedgerminaton is discussed in relation to the differential growthof axial and cotyledonary tissues. Resting potentials of excitable cells in the petiole and mainpulvinus of Mimosa pudica depended on the external concentrationsof potassium ([K$]) and sodium ([Na$]), and peaks of the actionpotentials depended on the external concentration of chloride([Cl]) in the excitable protoxylem cells of the petioleand the upper-half cells of the main pulvinus, which suggeststhat the action potential in these cells is a Cl spike.In the lower-half cells of the main pulvinus, the peak of theaction potential did not show a clear dependence on [K$], [Na$]or [Cl]. This implies that there is a decrease in theion selectivity of the membrane toward the peak of the actionpotential. Effective membrane resistances of the excitable protoxylemand phloem cells of the petiole, and the lower-half cells ofthe main pulvinus were 3.9?3.1 M (mean?SD, n=7), 9.4?8.2 M (n=5)and 5.0?2.9 M (n=15), respectively. A study of weekly and seasonal variation of stroke onset   总被引:2,自引:0,他引:2  
A registry based study was conducted to assess the variation in first-onset stroke with weekdays and seasons, in relation to the effects of age. Between 1 December 1991 and 30 November 1998, 10,729 first-onset stroke patients aged 25 or more were registered in Toyama Prefecture, Japan. We compared the weekly and seasonal variation in first-onset stroke by a one-way goodness-of-fit chi(2)-test. The relationship between seasonal variation in stroke onset and age was also evaluated by the method of Kendall's tau-b R x C tables with ordered categories. The frequency of onset of all strokes and cerebral infarctions (CI) was significantly higher on weekdays than at weekends (P < 0.01). More men had strokes and CI on a Monday (P < 0.01), and more women had cerebral hemorrhage (CH) on a Monday and CI at the end of the week. Stroke incidence was higher in patients aged less than 60 years (20.6%) than in those aged 60 years or over (18.7%) on a Monday compared to the weekend. By chi(2)-test, comparing observed with expected numbers of stroke onsets, weighted by the number of days in each 3-month period, the incidence of all strokes, CI and CH was significantly higher in winter and spring than in summer. The seasonal variation in the onset of stroke declined with age: all strokes (P < 0.001) and CH (P < 0.001) in both genders; subarachnoid hemorrhage (P < 0.001) only in men. Our study shows that the onset of stroke is more frequent on weekdays than on weekends, and may be associated with changes in psychophysiological stresses between working days and the weekend. We also observed a clear negative dose response relationship between seasonal variations in occurrence and age. It may be speculated that younger people have more change to work outdoors, exposing themselves to the winter environment. Their lifestyle and physiological condition may be different from those of older people.  相似文献
