Morphological changes induced by the melittin tetramer on bilayers of egg phosphatidylcholine and dipalmitoylphosphatidylcholine have been studied by quasi-elastic light scattering, gel filtration and freeze-fracture electron microscopy. It is concluded that melittin similarly binds and changes the morphology of both single and multilamellar vesicles, provided that their hydrocarbon chains have a disordered conformation, i.e., at temperatures higher than that of the transition, Tm. When the hydrocarbon chains are ordered (gel phase), only small unilamellar vesicles are morphologically affected by melittin. However after incubation at T greater than Tm, major structural changes are detected in the gel phase, regardless of the initial morphology of the lipids. Results from all techniques agree on the following points. At low melittin content, phospholipid-to-peptide molar ratios, Ri greater than 30, heterogeneous systems are observed, the new structures coexisting with the original ones. For lipids in the fluid phase and Ri greater than 12, the complexes formed are large unilamellar vesicles of about 1300 +/- 300 A diameter and showing on freeze-fracture images rough fracture surfaces. For lipids in the gel phase, T less than Tm after passage above Tm, and for 5 less than Ri less than 50, disc-like complexes are observed and isolated. They have a diameter of 235 +/- 23 A and are about one bilayer thick; their composition corresponds to one melittin for about 20 +/- 2 lipid molecules. It is proposed that the discs are constituted by about 1500 lipid molecules arranged in a bilayer and surrounded by a belt of melittin in which the mellitin rods are perpendicular to the bilayer. For high amounts of melittin, Ri less than 2, much smaller and more spherical objects are observed. They are interpreted as corresponding to lipid-peptide co-micelles in which probably no more bilayer structure is left. It is concluded that melittin induces a reorganization of lipid assemblies which can involve different processes, depending on experimental conditions: vesicularization of multibilayers; fusion of small lipid vesicles; fragmentation into discs and micelles. Such processes are discussed in connexion with the mechanism of action of melittin: the lysis of biological membranes and the synergism between melittin and phospholipases.  相似文献

A full length dopamine-beta-hydroxylase (DBH) cDNA clone was isolated from a human pheochromocytoma lambda gt11 library. Both structural and functional evidence confirms the authenticity of the clone: (i) antibodies selected with fusion proteins generated by positive clones precipitate DBH activity, (ii) the sequence of three internal DBH tryptic peptides are included in the deduced DBH sequence, (iii) the previously reported N-terminal 15 amino acids of bovine DBH exhibits a nearly complete identity with that predicted for human DBH. The polypeptide chain of DBH comprises 578 amino acids corresponding to an unmodified protein of 64 862 daltons and is preceded by a cleaved signal peptide of 25 residues. DBH exists in both membrane-bound and soluble forms. The hydropathy plot reveals no obvious hydrophobic segment, except the signal peptide. S1 mapping analysis indicates no diversity in the 5' and 3' extremities of the DBH mRNA. Taken together with available biochemical data, these observations suggest that the membrane attachment of DBH probably results from a post-translational modification, glypiation being the most likely candidate. Comparative amino acid sequence analysis establishes that DBH shares no homology with the other catecholamine synthesizing enzymes, tyrosine hydroxylase and phenylethanolamine-N-methyl transferase.  相似文献

Effects of mine acid on longevity and reproductive rate of the parthenogenic gastrotrich Lepidodermella squammata were studied under laboratory culture conditions. Water from unpolluted and polluted streams was used, directly or mixed, to establish a series of test conditions at pH 8.1, 7.1, 6.4, 5.2, 4.6 and 3.3. Eggs, cultured individually under each test condition, were observed at 12 h intervals for hatching, daughter egg laying and death. Data representing 50 animals under each test condition were used in the construction of a series of life tables. From these were calculated maximal life expectancy (ex), net reproductive rate per individual lifetime (Ro) and intrinsic rate of natural increase (rmax).Values of ex, Ro and rmax were maximal at pH 7.1 and were reduced slightly at the higher pH; ex was greatly reduced and Ro zero at pH 6.4 and 5.2; and ex was zero at pH 4.6 and 3.3. Analysis of variance tests indicate significant differences between ex values of L. squammata cultured at pH 8.1 and those cultured at pH 7.1, but no differences between Ro or rmax.Associated with the decrease in pH was an increase in total conductivity and a decrease in carbonate alkalinity and hence in carbonate conductivity. It appears that L. squammata is capable of living and reproducing at pH 6.0 to 6.5 under field conditions low in carbonates, providing non-carbonate ions are not abundant, or under field conditions high in non-carbonate ions, providing sufficient carbonates are present.  相似文献

Crepidorhopalon perennis is a metallophyte critically endangered by mining activities and currently known from only one site on copper‐rich soils in Katanga (Dem. Rep. Congo). It is closely related to the annual C. tenuis, also a rare metallophyte, but with a broader geographical range. We investigated the variation in morphometric traits and ecological niches (based on edaphic conditions and pollinator assemblages) of C. perennis and C. tenuis, to evaluate the risk of inter‐specific competition, and their potential for hybridization to ascertain if C. perennis might be at risk of genetic swamping by its more widespread congener. We examined whether species were found under sympatric or parapatric settings with opportunity for hybridization (or gene exchange). Such knowledge is essential for implementing restoration management protocols, including the introduction of C. perennis into substitution sites where C. tenuis might be already present. Fourteen morphological characters and 11 soil variables were measured and visiting pollinator species were identified at the site where the two species co‐occur. Our results show that the two species can be distinguished based on their morphological traits, show niche overlap based on edaphic properties, and share the same pollinator assemblage. In addition, no morphologically intermediate individuals could be detected, suggesting no hybridization, and that the two species may be reproductively isolated. We conclude that C. perennis conservation and restoration operations can be realized in substitution sites where C. tenuis may be present, with the need, however, to evaluate the potential effect of sharing a pollinator assemblage on reproductive success of both species.  相似文献

Plant communities of soils naturally enriched in copper and cobalt in Katanga (D. R. Congo) are critically threatened in the short term due to mining activities. For biodiversity conservation and ecosystem restoration purposes, there is an urgent need to acquire more knowledge on those plant communities including their diversity and their relationships to environmental factors. The classification of 62 vegetation plots located in 6 metal‐rich rocky hills in the Tenke Fungurume mining area resulted in 3 well‐defined steppic and steppic savanna communities. Canonical analysis showed that the community comprising the largest proportion of strictly endemic metallophytes (i.e. species that only occur on metal‐rich soils) developed in the soils with the most elevated concentrations of Cu and Co. However, contrasting species assemblages in the two other plant communities were explained by soil nutrients and percentage rocks in addition to heavy metal concentrations. The results of this study will assist with restoration efforts because they (1) provide a rigorous assessment of communities before a disturbance and (2) define essential edaphic conditions needed for the reestablishment of critical communities.  相似文献

Positive supercoils are introduced in cellular DNA in front of and negative supercoils behind tracking polymerases. Since DNA purified from cells is normally under-wound, most studies addressing the relaxation activity of topoisomerase I have utilized negatively supercoiled plasmids. The present report compares the relaxation activity of human topoisomerase I variants on plasmids containing equal numbers of superhelical twists with opposite handedness. We demonstrate that the wild-type enzyme and mutants lacking amino acids 1–206 or 191–206, or having tryptophane-205 replaced with a glycine relax positive supercoils faster than negative supercoils under both processive and distributive conditions. In contrast to wild-type topoisomerase I, which exhibited camptothecin sensitivity during relaxation of both negative and positive supercoils, the investigated N-terminally mutated variants were sensitive to camptothecin only during removal of positive supercoils. These data suggest different mechanisms of action during removal of supercoils of opposite handedness and are consistent with a recently published simulation study [Sari and Andricioaei (2005) Nucleic Acids Res., 33, 6621–6634] suggesting flexibility in distinct parts of the enzyme during clockwise or counterclockwise strand rotation.  相似文献

Melittin is known to self-associate as tetramers in solutions of high ionic strength. Here, an N-bromosuccinimide oxidized-Trp19 melittin is prepared. This derivative can act as an acceptor of the fluorescence of native melittin and is used in order to observe a possible self-association of melittin in phospholipid bilayers.Resonance energy transfer was shown to occur in solutions of high ionic strength, showing that oxidized melittin can associate with native melittin.In phospholipid bilayers, no association is detected in the absence of NaCl. In its presence, an equilibrium between monomeric melittin and oligomeric species is observed. These species are not dimers, but any other degree of association may account for our experimental results. Significant differences in characteristic transfer efficiency reveal differences in the structure of these oligomers according to the length or state of phospholipids (fluid or at the transition temperature).
