Selective, double irradiation allows the assignment of most 13C-n.m.r. signals in a series of per-O-acetyl disaccharides composed of two D-glucose residues linked α-(1→3), β-(1→3), α-(1→4), β-(1→4), α-(1→6), β-(1→6), and α,α-(1→1). The main influences that affect the chemical shifts are discussed and the spectra of β-cellobiose octaacetate and β-maltose octaacetate are compared to those of cellulose and amylose triacetate, respectively, to show the possibilities and limitations of a disaccharide model for the interpretation of the 13C-spectrum of a polymer.  相似文献

A comparison of methods used in isolating microsomes and in measuring microsomal cytochrome P-450 demonstrated that separation following protoplast lysis gave the best results. By this latter technique a high amount of cytochrome P-450 (0.2–0.3 nmol/mg) was recovered but cytochrome P-420, considered as the denatured form, was absent.The alkanes specifically induce cytochromes P-450 and b5 localized on the microsomes. The denaturation in vivo of cytochrome P-450 into cytochrome P-420 even occurs during storage at 1 °C. This degradation is increased during preparation of subcellular fractions if no preventive measures are taken.  相似文献

4-Methylumbelliferyl 2-acetamido-2-deoxy-β-D-glucopyranoside, 2-acetamido-4-O-(2-acetamido-2-deoxy-β-D-glucopyranosyl)-2-deoxy-β-D-glucopyranoside (di-N-acetyl-β-chitobioside), and O-(2-acetamido-2-deoxy-β-D-glucopyranosyl)-(1→4)-O-(2-acetamido-2-deoxy-β-D-glucopyranosyl)-(1→4)-2-acetamido-2-deoxy-β-D-glucopyranoside (tri-N-acetyl-β-chitotrioside) were obtained in good yield from the corresponding peracetylated glycosyl chlorides by condensation with the sodium salt of 4-methylumbelliferone in N,N-dimethylformamide. The trisaccharide glycoside is hydrolyzed by lysozyme and is, therefore, a convenient substrate for this enzyme; the 4-methylumbelliferone produced can be determined by the increase of the fluorescence intensity at 442 nm. The intensity of the fluorescence of 4-methylumbelliferyl tri-N-acetyl-β-chitotrioside is enhanced upon binding with lysozyme without modification of the position of the absorption maximum. The binding constant and the rate of hydrolysis of the trisaccharide glycoside by lysozyme are higher than those obtained with p-nitrophenyl tri-N-acetyl-β-chitotrioside.  相似文献

Summary Two spontaneous mutants of Escherichia coli strain KMBL-146 selected for resistance to the aminoglycoside antibiotic neamine show severe restriction of amber suppressors in vivo. Purified ribosomes from the mutant strains exhibit low neamine-induced misreading in vitro and a decreased affinity for the related antibiotic streptomycin.Biochemical analysis shows that the mutants each have two modified 30S ribosomal proteins, S12 and S5. In agreement with these results, genetic analysis shows that two mutations are present, neither of which confers resistance to neamine by itself; the mutation located in gene rpxL (the structural gene for protein S12) confers streptomycin dependence but this dependence is suppressed in the presence of the second mutation, located in gene rpxE (the structural gene for protein S5).  相似文献
