Pichinde virus (PV) strain AN 3739 was determined to be sensitive to natural killer (NK) cells in vivo by enhanced replication in NK-cell-depleted mice. An NK-sensitive subclone (PV-NKs1) was serially passed in mice whose NK cells had previously been activated by an interferon inducer, and two plaque isolates were shown to be resistant to NK cells but not to interferon. Inoculation of severe-combined-immunodeficient mice with PV-NKs1 led to a persistent infection resulting in an NK-resistant viral population. This is the first demonstration of the isolation of viral "NK-escape" variants, as defined by the ability of the virus to replicate in vivo.  相似文献

Oncogenic mink cell focus-forming (MCF) viruses, such as MCF 247, show a positive correlation between the ability to replicate efficiently in the thymus and a leukemogenic phenotype. Other MCF viruses, such as MCF 30-2, replicate to high titers in thymocytes and do not accelerate the onset of leukemia. We used these two MCF viruses with different biological phenotypes to distinguish the effect of specific viral genes and genetic determinants on thymotropism and leukemogenicity. Our goal was to identify the viral sequences that distinguish thymotropic, nonleukemogenic viruses such as MCF 30-2 from thymotropic, leukemogenic viruses such as MCF 247. We cloned MCF 30-2, compared the genetic hallmarks of MCF 30-2 with those of MCF 247, constructed a series of recombinants, and tested the ability of recombinant viruses to replicate in the thymus and to induce leukemia. The results established that (i) MCF 30-2 and MCF 247 differ in the numbers of copies of the enhancer sequences in the long terminal repeats. (ii) The thymotropic phenotype of both viruses is independent of the number of copies of the enhancer sequences. (iii) The oncogenic phenotype of MCF 247 is correlated with the presence in the virus of duplicated enhancer sequences or with the presence of an enhancer with a specific sequence. These results show that the pathogenic phenotypes of MCF viruses are dissociable from the thymotropic phenotype and depend, at least in part, upon the enhancer sequences. On the basis of these results, we suggest that the molecular mechanisms by which the enhancer sequences determine thymotropism are different from those that determine oncogenicity.  相似文献

The quality of water sources and its potential health implications to adults and children populations of respective major communities in Northern Cross-River was assessed. Water samples (n = 10/water source/site) were collected from three (Okpoma, Okuku and Ugaga) communities and heavy metal concentrations (Lead (Pb), Cadmium (Cd), Chromium (Cr), Manganese (Mn), Nickel (Ni), Copper (Cu), Cobalt (Co), and Zinc (Zn)) were evaluated using Atomic Absorption Spectrometer (AAS). Overall, Pb, Cd, Ni, and Co were higher than drinking water guidelines, while only Cr, Mn, Cu, and Zn were within the permissible limits. The estimated average daily intake (EADI) and target hazard quotient (THQ) were used to determine risk implications for adult and children consumer populations. The EADI for Pb in adults for borehole water, Pb and Cr by child consumer population for borehole and shallow well water exceeded the reference dose (RfD) by USEPA. The THQ for adult population were >1 for Pb in borehole water and >1 for Pb and Cr across all sites for the child consumer population. Overall, our findings indicate toxicity and higher hazard risk for both adult (Pb) and children (Pb and Cr) populations that source drinking water from borehole and shallow well water in these communities.  相似文献

A new type of (reduced) point average molecular weight A1, is described. Several interesting properties are developed: (i) A1 = reduced weight average molecular weight over the whole cell, Awo A1 (meniscus) = Aw (meniscus); (iii) A1 (zero concentration) = reduced number average molecular weight, An (meniscus). In addition, its usefulness in extracting the meniscus concentration, J(a), and in examining heterogeneous systems such as mucus glycoproteins, are discussed. The evaluation and application of A1 requires only simple computational facilities, without the use for large-scale multiple data acquisition and recycling techniques.  相似文献

The growth properties of twelve different amber (am) mutants of bacteriophage T4 gene 43 (DNA polymerase) were examined by using nonpermissive (su(-)) as well as permissive (su(+)) Escherichia coli hosts. It was found that most of these mutants were measurably suppressed in su(-) hosts by translational ambiguity (misreading of codons during protein synthesis). The ability of these mutants to grow in response to this form of weak suppression probably means that the T4 gene 43 DNA polymerase can be effective in supporting productive DNA replication when it is supplied in small amounts. By similar criteria, studies with other phage mutants suggested that the products of T4 genes 62 (uncharacterized), 44 (uncharacterized), 42 (dCMP-hydroxymethylase), and 56 (dCTPase) are also effective in small amounts. Some T4 gene products, such as the product of gene 41 (uncharacterized), seem to be partially dispensable for phage growth since am mutants of such genes do propagate, although weakly, in streptomycin-resistant su(-) hosts which appear to have lost the capacity to suppress am mutations by ambiguity.  相似文献

Comparison of a number of murine leukemia virus clones by sodium dodecyl sulfate-polyacrylamide gel electrophoresis revealed extensive protein polymorphism among B-tropic, but not N-tropic, isolates from BALB/c mice, particularly in migration of p30 proteins. A type-specific radioimmunoassay for p30 was developed which uniformly discriminated all B-tropic viruses from N-tropic viruses of BALB/c origin. N- and B-tropic viruses of C57BL/6 and AKR Fv-1b/b origin could also be distinguished by this assay.  相似文献

Global increases in the occurrence of large, severe wildfires in forested watersheds threaten drinking water supplies and aquatic ecology. Wildfire effects on water quality, particularly nutrient levels and forms, can be significant. The longevity and downstream propagation of these effects as well as the geochemical mechanisms regulating them remain largely undocumented at larger river basin scales. Here, phosphorus (P) speciation and sorption behavior of suspended sediment were examined in two river basins impacted by a severe wildfire in southern Alberta, Canada. Fine‐grained suspended sediments (<125 μm) were sampled continuously during ice‐free conditions over a two‐year period (2009–2010), 6 and 7 years after the wildfire. Suspended sediment samples were collected from upstream reference (unburned) river reaches, multiple tributaries within the burned areas, and from reaches downstream of the burned areas, in the Crowsnest and Castle River basins. Total particulate phosphorus (TPP) and particulate phosphorus forms (nonapatite inorganic P, apatite P, organic P), and the equilibrium phosphorus concentration (EPC0) of suspended sediment were assessed. Concentrations of TPP and the EPC0 were significantly higher downstream of wildfire‐impacted areas compared to reference (unburned) upstream river reaches. Sediments from the burned tributary inputs contained higher levels of bioavailable particulate P (NAIP) – these effects were also observed downstream at larger river basin scales. The release of bioavailable P from postfire, P‐enriched fine sediment is a key mechanism causing these effects in gravel‐bed rivers at larger basin scales. Wildfire‐associated increases in NAIP and the EPC0 persisted 6 and 7 years after wildfire. Accordingly, this work demonstrated that fine sediment in gravel‐bed rivers is a significant, long‐term source of in‐stream bioavailable P that contributes to a legacy of wildfire impacts on downstream water quality, aquatic ecology, and drinking water treatability.  相似文献

Temperate forest 15N isotope trace experiments find nitrogen (N) addition‐driven carbon (C) uptake is modest as little additional N is acquired by trees; however, several correlations of ambient N deposition against forest productivity imply a greater effect of atmospheric nitrogen deposition than these studies. We asked whether N deposition experiments adequately represent all processes found in ambient conditions. In particular, experiments typically apply 15N to directly to forest floors, assuming uptake of nitrogen intercepted by canopies (CNU) is minimal. Additionally, conventional 15N additions typically trace mineral 15N additions rather than litter N recycling and may increase total N inputs above ambient levels. To test the importance of CNU and recycled N to tree nutrition, we conducted a mesocosm experiment, applying 54 g N/15N ha?1 yr?1 to Sitka spruce saplings. We compared tree and soil 15N recovery among treatments where enrichment was due to either (1) a 15N‐enriched litter layer, or mineral 15N additions to (2) the soil or (3) the canopy. We found that 60% of 15N applied to the canopy was recovered above ground (in needles, stem and branches) while only 21% of 15N applied to the soil was found in these pools. 15N recovery from litter was low and highly variable. 15N partitioning among biomass pools and age classes also differed among treatments, with twice as much 15N found in woody biomass when deposited on the canopy than soil. Stoichiometrically calculated N effect on C uptake from 15N applied to the soil, scaled to real‐world conditions, was 43 kg C kg N?1, similar to manipulation studies. The effect from the canopy treatment was 114 kg C kg N?1. Canopy treatments may be critical to accurately represent N deposition in the field and may address the discrepancy between manipulative and correlative studies.  相似文献

Bud mite, blister mite (Colomerus vitis Pagenstecher), and rust mite (Calepitrimerus vitis Nalepa) (Acari: Eriophyoidae) are recognized grapevine pests. Much of the biology and ecology of these pests is poorly understood. We used two types of molecular markers to gain further insight into the breeding biology and population structure of these mites, using individuals collected from sites around south‐eastern Australia. Patterns of genetic variation observed using PCR‐RFLP of ITS 1 (Internal Transcribed Spacer 1) confirmed the separate species status of the rust mite, and resolved the species status of bud and blister mites, revealing two closely related but distinct species. Microsatellite markers revealed extensive genetic differentiation between bud mite populations and blister mite populations even at micro‐geographical levels, suggesting low movement in these species. The findings indicate that separate control strategies are needed against bud and blister mites, and that localized control strategies are likely to be effective given their limited dispersal.  相似文献
