The preparative method of polyacrylamide gel electrophoresis was adapted for purification and characterization of isotopically labeled L-cell interferons. Re-covery of interferon activity was quantitative, and purification and resolution were comparable to those obtained by analytical polyacrylamide gel electrophoresis. Ultimate specific activities attainable ranged from 2 x 10(6) to 3 x 10(6) international units per mg of protein.  相似文献

The antibiotic activity of the antifungal substance mucidin was compared with the activity of nystatin and pimaricin. The antibiotics were tested by the plate method using 19 fungal species, mainly phytopathogenic ones. Toward 14 species, mucidin was ten times more active than nystatin and pimaricin, toward 5 species the activities were roughly the same. The antibiotics differed also in the sharpness of the inhibition zone boundaries.  相似文献

Ethylene was collected in methanol solution of mercuric acetate and the addition compound formed was then separated by means of paper chromatography. The spot area and colour intensity after detection were determined using a densitometer. The amount of collected ethylene was calculated from a calibration curve. The ethylene liberated from plant samples was collected during one or two days. During this period the amputation of the whole plant organs did not influence ethylene production. Changes in ethylene production were found after segmentation of the tissue or after the treatment with auxin and Co2+ ions. The above-ground parts of investigated herbs released 0.3 to 3.5 μl of ethylene per kg fresh weight per hour. The leaves of investigated trees released 1 to 20 μl of ethylene per kg f.w. per hour. The rate of the production of ethylene seems to be specific for a given species.  相似文献

It was found that ethionine partially inhibits the transport of the inducer (TMG) of β-galactosidase into the cells ofEscherichia coli ML-30. The synthesis of β-galactosidase-specific messenger RNA is not inhibited. Ethionine appears to be incorporated into proteins synthesized by the strains used. The incorporation of ethionine into the molecule of β-galactosidase results in the synthesis of an enzymically inactive, immunologically cross-reacting protein.  相似文献

Summary The morphology of postnatal differentiation of the Golgi apparatus, the nucleus, the perikaryon, and the dendrites was studied in Purkinje cells of the rat cerebellum for 30 days after birth using histochemical, histological, and electron microscopic methods.The Golgi apparatus during differentiation undergoes morphological and positional changes. From the 1st to 7th postnatal day, the Golgi apparatus is found in a supranuclear position, and is connected with the axes of differentiating primary dendrites by beam-like processes. From days 8 to 11 this connection disappears, and most of the Golgi apparatus assumes a lateronuclear and infranuclear position. After the 11th or 12th day, the Golgi apparatus is found in perinuclear and peripheral cytoplasmic positions. The formation of granular endoplasmic reticulum occurs in the vicinity of the perinuclear Golgi apparatus. The differentiation of cell and nuclear forms requires approximately 20 days. The morphological changes of differentiation are discussed in relation to the participation of the Golgi apparatus in the differentiation of dendrites and in the formation of the granular endoplasmic reticulum.  相似文献

In the experiment with 18 winter varieties the genotypic, phenotypic and environmental correlations were estimated in all combinations between the pairs of 11 quantitative characters. Both yield of grain with its components and further characters were evaluated, of which flag leaf size and leaf angle of the last but one leaf are of importance for the utilization of light energy and consequently for receiving maximum rates of photosynthesis. In simple genotypic and phenotypic correlations their different stability with regard to mineral nutrition, eventually to the date of sowing was proved. In the discussion the limited signification of simple correlation for the statement of complicated processes in plants caused by multiplied associations of characters was pointed out. In the example of yield components of the plant at the application of 60 kg N per hectare it was shown that also further unknown variable factors might participate in the genotypic correlation. It may be concluded that the inclusion of them when constructing the selection indexes will give further foundations for handling yield as a breeding or genetic character, and for the causal analysis of yield formation in wheat.  相似文献

Ethionine at concentrations of 10−3M, 5×10−3M and 10−2M inhibits growth, both of β-galactosidase inducible ML-30 and constitutive ML-308Escherichia coli strains. The protein synthesis (measured by the incorporation of l-leucine-14C and l-aspartic-14C acid into proteins) of these strains is inhibited to the same extent as their growth. The synthesis of inducible and constitutive β-galactosidase produced by the strains ML-30 and ML-308, respectively, is considerably inhibited by ethionine.  相似文献
