This report aims to facilitate the implementation of the Three Rs (reduction, refinement and replacement) in the testing of vaccines for regulatory and other purposes. The focus is predominantly on identification of reduction and refinement opportunities in batch potency testing but the principles described are widely applicable to other situations that involve experimental infections of animals. The report should also help to interpret the requirements of the European Pharmacopoeia with regard to the use of alternative tests, humane endpoints and other refinements. Two specific worked examples, for batch potency testing of Clostridium chauvoei and canine leptospira, with recommendations for harmonisation of international test requirements for these and other vaccines, are provided as appendices online. 相似文献

Drought is a major limitation for rice production in rainfed ecosystems. Identifying quantitative trait loci (QTLs) linked
to drought resistance provides opportunity to breed high yielding rice varieties suitable for drought-prone areas. Although
considerable efforts were made in mapping QTLs associated with drought-resistance traits in rice, most of the studies involved
indica × japonica crosses and hence, the drought-resistance alleles were contributed mostly by japonica ecotypes. It is desirable to look for genetic variation within indica ecotypes adapted to target environment (TE) as the alleles from japonica ecotype may not be expressed under lowland conditions. A subset of 250 recombinant inbred lines (RILs) of F8 generation derived from two indica rice lines (IR20 and Nootripathu) with contrasting drought-resistance traits were used to map the QTLs for morpho-physiological
and plant production traits under drought stress in the field in TE. A genetic linkage map was constructed using 101 polymorphic
PCR-based markers distributed over the 12 chromosomes covering a total length of 1,529 cM in 17 linkage groups with an average
distance of 15.1 cM. Composite interval mapping analysis identified 22 QTLs, which individually explained 4.8–32.2% of the

Mitotic centromere-associated kinesin (MCAK)/Kif2C is the most potent microtubule (MT)-destabilizing enzyme identified thus far. However, MCAK's function at the centromere has remained mechanistically elusive because of interference from cytoplasmic MCAK's global regulation of MT dynamics. In this study, we present MCAK chimeras and mutants designed to target centromere-associated MCAK for mechanistic analysis. Live imaging reveals that depletion of centromere-associated MCAK considerably decreases the directional coordination between sister kinetochores. Sister centromere directional antagonism results in decreased movement speed and increased tension. Sister centromeres appear unable to detach from kinetochore MTs efficiently in response to directional switching cues during oscillatory movement. These effects are reversed by anchoring ectopic MCAK to the centromere. We propose that MCAK increases the turnover of kinetochore MTs at all centromeres to coordinate directional switching between sister centromeres and facilitate smooth translocation. This may contribute to error correction during chromosome segregation either directly via slow MT turnover or indirectly by mechanical release of MTs during facilitated movement. 相似文献

We used stable isotope (SI) methods in combination with pen feeding trials to determine the importance of eggs of the Atlantic horseshoe crab Limulus polyphemus to migratory fattening of red knots Calidris canutus rufa and ruddy turnstones Arenaria interpres morinella during spring stopover in Delaware Bay. By manifesting measurable fractionation (ca +3‰) and rapid turnover, blood plasma δ15 nitrogen proved a functional marker for SI diet tracking during the short 3-week stopover. Blood samples from free-ranging knots (3 data sets) and turnstones (1 data set) produced similar convergence of plasma δ15 N signatures with increasing body mass that indicated highly similar diets. Asymptotes deviated slightly (0.3‰ to 0.7‰) from that of captive shorebirds fed a diet of only crab eggs during stopover, thus confirming a strong crab egg-shorebird linkage. The plasma δ15 N crab-egg diet asymptote was enriched ca +4.5‰ and therefore readily discriminated from that of either blue mussels Mytilus edulis or coquina clams Donax variabilis , the most likely alternative prey of knots in Delaware Bay. Crab eggs were highly palatable to captive knots and turnstones which achieved rates of mass gain (3–11 g/d) comparable to that of free-ranging birds. Peak consumption rates during hyperphagic events were 23,940 and 19,360 eggs/bird/d, respectively. The empirical conversions of eggs consumed to body mass gained (5,017 eggs/g for knots and 4,320 eggs/g for turnstones) indicate the large quantities of crab eggs required for the maintenance of these shorebird populations during stopover. 相似文献
