Human blood was sheared between rotating polyethylene disks and plasma hemoglobin measured at intervals to produce kinetic
hemolysis curves (KHC), plotted as free hemoglobin concentration vs time. The KHC produced by blood samples incubated in the
presence of penicillin, streptomycin, gentamicin, and amikacin lie always below those for control samples, indicating a reduction
in hemolysis; this reduction was greater as the drug concentration was increased. Explanations in terms of alterations in
red cell structure were sought by several characterization tests of amikacin-loaded blood samples. Drug-localization studies
demonstrated that significant fractions of the total dosage were associated with the red-cell membrane. Resistive pulse spectroscopy
was used to show how amikacin affected cell size, deformability, and osmotic fragility; results were sensitive to storage
age of the blood. In all cases, the effect of shearing was to reduce cell size, deformability, and osmotic fragility. Mechanisms
for hemolytic protection by drugs are proposed. 相似文献

Summary Abnormalities in DNA metabolism have been found in third-instar females of Drosophila melanogaster that are heteroallelic or homoallelic for X-chromosomal giant (gt) mutations. Analysis of DNA metabolism in larval brain ganglia was carried out using alkaline sucrose gradient centrifugation, incorporation assays and a neutral filter elution assay. These analyses show that gt stocks synthesize DNA of a reduced molecular weight, have an unusually high frequency of spontaneous single and doublestrand breaks, and exhibit a reduction in the normal inhibition of DNA synthesis following treatment with UV and the carcinogen AAAF. These phenomena are not associated with a defect in the repair of X-ray induced DNA breaks nor are they accompanied by any alterations in chromosome stability. Analysis of homozygous 1(2)gl larvae also reveal that these phenomena are specific to the gt locus and are thus not attributable solely to an extended developmental program. These findings strengthen the suggestion that the genetic instability associated with gt is related to perturbations in chromosome metabolism (Green 1982).Abbreviations used UV

Cell suspension cultures of the Madagascan Periwinkle Catharanthus roseus (L) G. Don were maintained on Gamborg's B5 medium and their growth monitored by measuring cellular fresh and dry weight, cell number and mitotic activity. Samples of cells of different ages and physiological states were subcultured onto an alkaloid production medium and their rates of growth and alkaloid accumulation measured over a period of 30–45 days. In two experiments the rate of biomass accumulation was directly related to the rate of cellular serpentine accumulation. Possible mechanisms underlying this phenomenon are discussed in relation to the properties of cells comprising the inocula. 相似文献

Synopsis Aspects of the life history of Barbus anoplus were studied in Lake le Roux, a turbid man-made lake on the Orange River, South Africa. This minnow underwent a population
explosion and successfully colonized the shoreline of the newly-formed lake during the early phases of reservoir filling.
Male and female B. anoplus reach sexual maturity in one year at about 40 mm fork length. They have a multiple spawning habit with the first spawning
in November–January and the second in February–March. The growth of the two resulting cohorts is discussed. It is proposed
that the offspring from the second spawning not only acts as a ‘back-up’ but is capable of prolonging the life of that year-class
into an additional reproductive season. Most of the minnows die after their second summer, but more offspring from the second
spawning, especially females, live into a third summer. Females attain a larger maximum size (73 mm FL) and age (3–4 years)
than males (60 mm FL, 2–3 years). B. anoplus is small and short-lived with a high seasonal reproductive potential, which is in contrast to the larger Barbus species in the Orange River system. These life-history traits enable the species to colonize and successfully inhabit unstable

The absolute rate for the repair reaction of radiation-induced, oxygen-dependent lesions in bacterial transforming DNA with the sulphydryl (SH)-containing compound dithiothreitol (DTT) has been determined using a fast response method, the gas explosion technique, to be 1.6 X 10(6) mol-1 s-1. Glutathione reacts ten times slower than DTT with the irradiated transforming DNA. It can also be calculated that transforming DNA radicals react with O2 in a damage-fixing reaction with a rate of about 3 X 10(8) dm3 mol-1 s-1. These rates are compared with values in the literature for reaction rates of SH, compounds and O2 with irradiated DNA constituents and with bacterial cells. 相似文献

A polypeptide which inhibits the growth of human carcinoma cells has been characterized from Novikoff rat ascites fluid. This tumor inhibitory factor co-purified with transforming growth factor activity through acid/ethanol extraction and Bio-Gel chromatography. The two activities were completely separated by reverse phase HPLC. The tumor inhibitory factor is heat stable and requires disulfide bonds for bioactivity. This factor inhibited the anchorage independent growth of the more differentiated human colon carcinoma cell lines but did not affect the less differentiated carcinoma cells. The presence of stimulatory and inhibitory activities in the same extracts suggests that the relative concentrations of these factors may be important in the control of cell growth. 相似文献

Preincubation of murine neuroblastoma cells (clone N1E-115) with terbium chloride resulted in a significant potentiation of carbachol-mediated increase in cyclic GMP formation. This effect was accompanied by a shift of the peak response from 30 s to 120 s and a 6-fold decrease in carbachol concentration producing half-maximal responses, in addition to a significant increase in the Hill coefficient. Terbium ions also caused a significant decrease in the affinity and an increase in the maximum binding of [3H]quinuclidinyl benzilate to muscarinic receptors, the change in affinity being mainly due to a decrease in the association rate. Preincubation of cells with 1 mM carbachol for 4 h (the desensitized state of the muscarinic receptor) resulted in a decrease in the ability of terbium to alter [3H]quinuclidinyl benzilate binding. The effects of terbium reported here might be due to its affecting muscarinic receptor-effector coupling, which is considered to be lost upon receptor desensitization. 相似文献

Scrapie is a transmissible disease that results in progressive degeneration of the central nervous system and death. Although scrapie has been studied histopathologically, relatively little is known concerning neurotransmitter alterations. Specific [3H]muscimol binding to whole brain crude synaptic membranes (CSM) from mice clinically affected with scrapie was significantly (p less than 0.01) reduced, to approximately 73% of that of the controls. Of the brain regions examined, binding to only cerebral CSM was significantly (p less than 0.0001) decreased. Scatchard analyses of saturation curves revealed that the high-affinity (KD = 8 +/- 3 nM) site for muscimol was abolished in cerebral CSM from scrapie-infected mice, while the low-affinity site was unaffected. Binding of [3H]flunitrazepam to cerebral CSM was unaffected by scrapie and was stimulated by GABA to the same extent in both scrapie and control mice. These results suggest that scrapie agent 139A in C57BL/6J mice manifests a portion of its CNS pathology via a high-affinity GABA binding site that is unassociated with the benzodiazepine receptor. 相似文献

A branch and bound algorithm is described for searching rapidlyfor minimal length trees from biological data. The algorithmadds characters one at a time, rather than adding taxa, as inprevious branch and bound methods. The algorithm has been programmedand is available from the authors. A worked example is givenwith 33 characters and 15 taxa. About 8 x 1012 binary treesare possible with 15 taxa but the branch and bound program findsthe minimal tree in <5 min on an IBM PC.
