Aleurone tissue from undried immature developing wheat grains (Triticum aestivum L. cv. Sappo), normally insensitive to gibberellic acid, can be made to respond to the hormone by a series of temperature treatments. Incubation of the de-embryoed grains at temperatures above 27° C for at least 8 h causes the tissue to become sensitive. Prolonged incubation at temperatures below 27° C does not effect a change in sensitivity. In addition to the requirement for exposure to an elevated temperature for a period of several hours the tissue must also subsequently be subjected to a period at a lower temperature for just a few seconds for the response to be observed. Once sensitized, the tissue remains responsive to gibberellic acid for substantial periods of time. Exposure of the tissue to temperatures which induce sensitivity to gibberellic acid also results in an increased leakage of amino acids. It is suggested that the increase in sensitivity to gibberellin requires two separate processes to take place. One could be a homeoviscous adaptation of the cell membranes in response to elevated temperature, the other a subsequent, permanent change in conformation of membrane components.  相似文献

Olfactory bulbs contain dendrodendritic synapses, which occur between granule cells and mitral cells, and gamma-aminobutyric acid (GABA) is thought to act as an inhibitory neurotransmitter at these synapses. Synaptosomes derived from the dendrodendritic synapses of the olfactory bulb were shown previously to contain considerable L-glutamate decarboxylase activity. The subcellular distribution and binding parameters of [3H]GABA and [3H]muscimol binding sites have now been determined in the rat olfactory bulb. Of all fractions examined, crude synaptic membranes (CSM) prepared from the dendrodendritic synaptosomes were shown to have the highest specific binding activity and accounted for nearly all of the total binding activity for both ligands. The specific binding activities for [3H]GABA and for [3H]muscimol were greatly increased after treating the CSM with 0.05% Triton X-100. Binding was shown to be Na+-independent, reversible, pharmacologically specific, and saturable. High- and low-affinity sites were detected for both ligands, and both classes of sites had appreciably lower KD values for muscimol (KD1 = 3.1 nM, KD2 = 25.1 nM) than for GABA (KD1 = 8.6 nM; KD2 = 63.7 nM). The amounts of the high-affinity binding sites for muscimol and GABA were similar (Bmax = 1.7 and 1.5 pmol/mg protein, respectively). The results of the present experiments indicate that the GABA and muscimol binding sites represent the GABA postsynaptic receptor, presumably on mitral cell dendrites, and provide further support for the hypothesis that GABA functions as a neurotransmitter at the dendrodendritic synapses in the olfactory bulb.  相似文献

The synthesis, high affinity uptake and degradation of GABA were compared with GABA content measured simultaneously in the nigras of unoperated, hemitransected and kainate-treated rats. In intact animals nigral GABA concentration was linearly related to glutamate decarboxylase activity, but not to GABA aminotransferase (GABA-T) or GABA uptake. One week after hemitransection all three parameters showed a linear reduction with GABA content, suggesting striatonigral GABA neurones may account for as much as 90% of the GABA and GAD, 31% of the GABA-T and 58% of GABA uptake sites in whole nigra. Nigral kainate treatment unexpectedly reduced all four parameters in parallel by as much as 50%, consistent with the formation of an unselective lesion.  相似文献

Summary The structure and metabolism of a soft-sediment estuarine macrofaunal community were measured over an annual cycle at two depth-contours in mesohaline Chesapeake Bay. Additional data for plankton productivity and respiration, as well as seston and sediment organics are also summarized for these communities. Benthic community respiration ranged from 0.24–3.38 g O2 m-2 d-1, and significant differences were detected between the two depths. Similarly, macroinfaunal standing stocks reached 11.2 and 32.3 g (ash free) m-2 for 3 m and 6 m depth communities, respectively, and both exhibited mid-summer declines in abundance. Inferences drawn from these data facilitated a partitioning of benthic community respiration into macrofaunal and meiofaunal/microbial components with a residual term, much of which could be explained statistically by interactions between these two components. A multi-variate statistical model developed from these data matched benthic respiration measurements within 1–2 S.E. Mass-balances of organic carbon were estimated for water column and benthos at the two depthcontours for early and late summer, as well as for an entire, time-weighted year. These various analyses led to the tentative conclusions that this benthic community was regulated by such internal factors as macrofaunal/meiofaunal grazing and microbial gardening, and by external factors such as temperature and predation by nekton. A mutant (CLP-8) of Saccharomyces cerevisiae possesses an abnormal ratio of native ribosomal subunits since it has an apparent deficiency of cytoplasmic 40 S subparticles. The mutant also has an abnormal anti-association factor activity. The lesion(s) responsible for the ribosomal subunit inbalance is not temperature-sensitive and is incomplete since the mutant still grows, albeit at a reduced rate compared to that of its parent. The lesion(s) in CLP-8 is, however, expressed at the level of 20 S ribosomal precursor RNA maturation. Thus, relative to the wild-type strain, there is both a slowed transport of 20 S ribosomal precursor RNA from the nucleus to the cytoplasm and a slowed cytoplasmic conversion of this RNA component into the mature 18 S form.  相似文献

The time evolution of slowly evolving discrete dynamical systems x i + 1 = T(r i ,x i ), defined on an interval [0, L], where a parameter r ichanges slowly with respect to i is considered. For certain transformations T, once r i reaches a critical value the system faces a non-zero probability of extinction because some x j [0, L]. Recent ergodic theory results of Ruelle, Pianigiani, and Lasota and Yorke are used to derive a simple expression for the probability of survival of these systems. The extinction process is illustrated with two examples. One is the quadratic map, T(r, x) = rx(1 – x), and the second is a simple model for the growth of a cellular population. The survival statistics for chronic myelogenous leukemia patients are discussed in light of these extinction processes. Two other dynamical processes of biological importance, to which our results are applicable, are mentioned.  相似文献

Graphite furnace atomic absorption spectrophotometry is a method used for the measurement of low concentrations of manganese (ppb range). Despite the widespread use of this technique, there is considerable inconsistency concerning sample preparation and choice of instrumental parameters. In this paper, we determined manganese concentrations of National Bureau of Standards (NBS) bovine liver by both graphite furnace (Instrumentation Laboratory IL 555B) and flame atomic absorption following wet digestion of the sample with nitric acid. The following instrumental parameters for the graphite furnace were found optimal for the measurement of manganese in digested NBS bovine liver: inert gas flow=14 SCFH, drying temperature 100°C/15 s (step 1), 125°C/15 s (step 2), pyrolysis temperature 500°C/15 s (step 3), and 1000°C/15 s (step 4); atomization temperature 2250°C/10 s (step 5). For optimal results, the nitric acid concentration of the sample should be between 2 and 4M. There were no significant differences found for manganese concentrations determined by either peak height or peak area measurement. Additionally, no significant differences were found in manganese concentrations determined by flame or furnace methods. Assuming proper sample preparation and choice of instrumental parameters, values obtained for manganese concentration by graphite furnace and flame atomic absorption spectrophotometry are similar. Therefore, data obtained by these two methods can be compared directly.  相似文献

The haploid genome size of Artemia is determined to be about 0.9·1012, as evidenced both by Feulgen microspectrophotometry of individual diploid class nuclei, which are but one of five polyploid classes present within the larvae, and by analysis of the reassociation kinetics of the isolated single copy DNA component. Polysomes isolated from 24-h incubation stage larvae contain an average of 10 ribosomes per messenger RNA molecule. Their rRNAs are found to have sedimentation coefficients of 18 S and 26 S, corresponding to molecular weights of 0.70·106 and 1.40·106, respectively, as determined by polyacrylamide electrophoresis and also by sucrose density centrifugation. Denaturation in glyoxal followed by agarose gel electrophoresis shows that unlike deuterostome rRNAs, Artemia 26 S rRNA contains a cryptic nick about midway in the molecule, which is not found in the 18 S molecule. Isolated rRNAs were labelled in vitro with 125I and hybridized with filter-immobilized DNA to saturation, which occurred at 0.051% for Xenopus, and at 0.074% for Artemia. From these results, it is calculated that in the haploid Artemia genome there are about 320 copies of the (18 S + 26 S) ribosomal RNA genes. Reciprocal heterologous hybridizations between these two species show that they share about 30% homology between their rDNA coding sequences.  相似文献

The tissue localization patterns of radiolabeled dividing cells obtained from gut lamina propria (LP), mesenteric (MLN) and peripheral (PLN) lymph nodes, and Peyer's patches (PP) were studied in guinea pigs using an adoptive lymphocyte transfer method. Within 24 hr 125I-deoxyuridine-labeled cells from donor LP and MLN, but not PLN, selectively localized in recipient gut and MLN. In contrast, donor PLN lymphoblasts returned to their sites of origin while labeled PP donor cells exhibited no specific tissue localization. These findings suggest that the gut LP, like the MLN, contains a population of cells which, unlike those in the PP and PLN, has the capacity to selectively localize in mucosal tissues. From this and other published work, we conclude that within the LP there is a population of cells at different stages of differentiation with a propensity to populate mucosae.  相似文献
