Eight human isoforms of phosphoinositide 3-kinases (PI3Ks) exist, but their individual functions remain poorly understood. Here, we show that different human small cell lung carcinoma (SCLC) cell lines overexpress distinct subsets of class I(A) and II PI3Ks, which results in striking differences in the signalling cascades activated by stem cell factor (SCF). Over expression of class I(A) p85/p110alpha in SCLC cells increased SCF-stimulated protein kinase B (PKB) activation and cell growth, but did not affect extracellular signal-regulated kinase (Erk) or glycogen synthase kinase-3 (GSK-3). This effect was selective, since it was not observed in SCLC cell lines overexpressing p85/p110beta or p85/p110delta. The SCF receptor associated with both class I(A) p85 and class II PI3KC2beta, and both enzymes contributed to SCF-stimulated PKB activity. A dominant-negative PI3KC2beta blocked both PKB activation and SCLC cell growth in response to SCF. Together our data provide novel insights into the specificity and functional significance of PI3K signalling in human cancer. 相似文献

Embryo dormancy in flowering plants is an important dispersal mechanism that promotes survival of the seed through time. The subsequent transition to germination is a critical control point regulating initiation of vegetative growth. Here we show that the Arabidopsis COMATOSE (CTS) locus is required for this transition, and acts, at least in part, by profoundly affecting the metabolism of stored lipids. CTS encodes a peroxisomal protein of the ATP binding cassette (ABC) transporter class with significant identity to the human X-linked adrenoleukodystrophy protein (ALDP). Like X-ALD patients, cts mutant embryos and seedlings exhibit pleiotropic phenotypes associated with perturbation in fatty acid metabolism. CTS expression transiently increases shortly after imbibition during germination, but not in imbibed dormant seeds, and genetic analyses show that CTS is negatively regulated by loci that promote embryo dormancy through multiple independent pathways. Our results demonstrate that CTS regulates transport of acyl CoAs into the peroxisome, and indicate that regulation of CTS function is a major control point for the switch between the opposing developmental programmes of dormancy and germination. 相似文献

New anti-cancer agents are being developed that specifically recognise tumour cells. Recognition is dependent upon the enhanced
expression of antigenic determinants on the surface of tumour cells. The tumour exposure and the extracellular accessibility
of the mucin MUC-1 make this marker a suitable target for tumour diagnosis and therapy. We isolated and characterised six
human scFv antibody fragments that bound to the MUC-1 core protein, by selecting a large naive human phage display library
directly on a MUC-1-expressing breast carcinoma cell line. Their binding characteristics have been studied by ELISA, FACS
and indirect immunofluorescence. The human scFv antibody fragments were specific for the tandem repeat region of MUC-1 and
their binding is inhibited by soluble antigen. Four human scFv antibody fragments (M2, M3, M8, M12) recognised the hydrophilic
PDTRP region of the MUC-1 core protein, which is thought to be an immunodominant region. The human scFv antibody fragments
were stable in human serum at 37 °C and retained their binding specificity.
For imaging or targeting to tumours over-expressing MUC-1, it might be feasible to use these human scFv, or multivalent derivatives,
as vehicles to deliver anti-cancer agents.

Evidence has been obtained that the product of ethylene biosynthesis from 1-aminocyclopropane carboxylic acid (ACC), aside from CO2, is cyanide. This has been accomplished by synthesis of a carbon-13-labeled ACC, feeding to apple tissue, and isolation of the free amino acids. By enzymatic degradation to CO2, followed by mass spectrometry, label is shown to be incorporated into the 4-carbon of asparagine. This occurs via well-known pathways for the metabolism of cyanide, and is confirmed by incorporation experiments using labeled cyanide. In conjunction with previous stereochemical studies, evidence for a sequential single-electron transfer pathway for ethylene biosynthesis has been considerably strengthened. 相似文献

An ultrastructural investigation was conducted of xylem parenchyma cells of peach (Prunus persica [L.] Batsch.) cv. Harbrite and red oak (Quercus rubra L.) in response to a freezing stress. Freezing curves of xylem tissues, as determined by differential thermal analysis, were used to predict temperatures at which both living and dead cells would be observed. Tissues were exposed to low temperatures (-15 to -35 C) and fixed in a frozen state at -10C and at thawing. Current models of the freezing behavior of supercooled plant cells suggest that xylem parenchyma cells behave as individual water droplets. This implies that cells are unresponsive to the presence of low temperature and extracellular ice until internal nucleation triggers lethal, intracellular freezing. For these reasons, deep supercooling has been described as an avoidance mechanism. Results of this study confirmed earlier reports that xylem parenchyma cells freeze as individuals or in small groups. Individual cells, however, did not exhibit a neutral response. Instead, a range of responses was observed that included internal and external vesiculation, deep invaginations of the plasma membrane, and the formation of electron-dense deposits external to the plasmalemma. In general, our observations suggested that the cells responded to a dehydrative stress. Results are discussed in context of the biophysical data associated with deep supercooling phenomena and compared to responses of cells that exhibit extracellular freezing. 相似文献

The aerial parts of Ophryosporus chilca afforded several prenylated p-hydroxyacetophenones, two labdane derivatives and a cinnamyl ester.
