目的对视网膜锥体细胞退行性变(retinal cone degeneration,RCD)大鼠的主要脏器及血液、生化指标进行检测。方法随机选择BN(Brown Norway)-RCDF2代大鼠,视网膜电图鉴定,取静脉血检测生化指标,并对重要脏器进行准确称量。结果此大鼠脏器质量在雌雄之间除体重、大脑和肾差异有显著性外(P〈0.05),其余均无差异;脏器系数脑干和小脑雌性显著大于雄性(P〈0.01);生化指标碱性磷酸酶、葡萄糖、白蛋白、尿素氮、总蛋白雌性高于雄性,总胆固醇、天冬氨酸转氨酶、丙氨酸转氨酶、甘油三酯雌性低于雄性,其中雌性碱性磷酸酶活性显著高于雄性;雌性离子钙浓度显著高于雄性。结论BN-RCD大鼠基础生理值和生化参数均在正常范围内,其中一些参数在性别之间有差异,但与正常SD大鼠相比,天冬氨酸转氨酶、丙氨酸转氨酶、肌酐、钾离子差异显著,葡萄糖、甘油三酯、离子钙有差异。RCD大鼠为遗传性视网膜疾病的研究提供了一种新的突变模式动物。  相似文献

采用富集培养法从工业油污土壤中分离到1株能以石油为惟一碳源而生长的细菌菌株,采用正交设计实验对该菌株的降解条件进行了初步研究。结果表明,最佳降解条件为NH_4Cl 4.0 g/LL,K_2HPO_4 1.5 g/L,pH 8.0,NaCl 15.0 g/L。在最佳条件下,浓度为1 mL/L的原油可在4 d内降解50%以上。  相似文献

目的探讨何首乌(Polygonum multiflorum Thunb.)不同营养器官和组织中蒽醌类和茋类化合物的分布,为合理利用何首乌提供科学依据.方法采用数码显微鉴定和TLC对何首乌的不同器官和组织部位进行了比较研究.结果从器官来看,何首乌藤茎、地下茎和块根中均含有蒽醌(大黄素和大黄素甲醚)和芪类(2,3,5,4'-四羟基二苯乙烯-2-O-β-D-葡萄糖苷)而叶和嫩茎则不舍;从组织部位来看,蒽醌类成分主要分布于韧皮部,而茋类成分主要分布于周皮和韧皮部,木质部最少.结论蒽醌类和茋类成分的产生与周皮的形成有一定的关系.蒽醌类化合物的含量可能与韧皮部的发达程度有关,茋类化合物主要产生和贮藏于薄壁细胞中.  相似文献

目的:以日本血吸虫基因SjFABP和SjGST原核表达产物检测二价DNA疫苗pVIVO2-SjFABP-SjGST在体内诱发的特异性抗体。方法:克隆日本血吸虫抗原基因SjFABP和sjGST,构建重组原核表达载体pET30a-SjFABP、pET30a-SjGST及真核表达载体pVIVO2-SjFABP-SjGST;将pET30a-SjFABP和pET30a-sjGST进行原核表达,并将表达产物用镍亲和柱分离纯化;采用Western印迹对日本血吸虫DNA疫苗pVIVO2-SjFABP-SjGST免疫4周后的BALB/c小鼠血清进行特异性抗体检测。结果:克隆了日本血吸虫抗原基因SjFABP(399bp)和町GST(657bp),并构建了pET30a-SjFABP、pET30a-SjGST及pVIVO2-SjFABP-SjGST重组质粒;经Western印迹检测,pET30a-SjFABP及pET30a-SjGST原核表达的抗原蛋白均能够与经日本血吸虫二价DNA疫苗pVIVO2-SjFABP-SjGST免疫的小鼠的血清产生特异性免疫反应。结论:日本血吸虫町尉即和町GST基因的原核表达系统成功建立;原核表达的抗原蛋白具有免疫原性;以原核表达产物可检测日本血吸虫DNA疫苗pVIVO2-SiFABP-SiGST在体内诱发的特异性抗体。  相似文献

将狂犬病病毒中和性单链抗体基因克隆入原核表达载体pET-PE40,经酶切鉴定及序列测定,成功构建了重组免疫毒素原核表达载体。IPTG诱导后目的蛋白获得高效表达,SDS-PAGE分析目的蛋白主要以不溶性包涵体的形式存在于菌体中,表达量占菌体总蛋白的32.29%。包涵体蛋白经体外复性及离子交换色谱柱、疏水作用色谱柱、Sephadex G200凝胶过滤层析柱三步纯化后获得纯度大于96%的目的蛋白,间接免疫荧光染色检测表明重组免疫毒素与狂犬病病毒感染细胞具有抗原结合活性,MTT试验显示,重组免疫毒素对狂犬病病毒感染细胞具有明显的杀伤作用,而对正常细胞无杀伤作用。  相似文献

蒙古栎和紫椴幼苗对光环境转变的光合作用响应   总被引:2,自引:1,他引:1  
比较研究了从温室5%光强转到10%、30%和100%光强处理下,蒙古栎(Quercus mongolica)和紫椴(Tilia amurensis)幼苗的光合能力和叶绿素荧光的响应,揭示了两个树种对光环境变化的不同适应情况及其光保护机制。结果表明,光强转换后两种幼苗都发生了严重光抑制,蒙古栎幼苗的最大光化学效率(Fv/Fm)在光强转变后第3天降到最低(0.52),紫椴幼苗在光强转变后第1天就降到了最低(0.67),蒙古栎降低幅度明显高于紫椴。之后随着光适应时间的延长逐渐恢复到原有水平,说明短时期的光抑制没有对两种幼苗的光合机构造成光损伤;从不同光照条件来看,无论是最大净光合速率(Pmax),还是实际光化学效率(ФPSⅡ),2种幼苗均为30%光强下的值高于10%和100%光强,说明过低或过高的光强都不利于幼苗的生长发育,只有适当的中光才利于幼苗的生长发育;与30%光强相比,蒙古栎幼苗100%光强下PmaxFv/FmФPSⅡ、NPQ的变化幅度远大于紫椴幼苗,表明高光强对蒙古栎幼苗的影响要大于紫椴;100%光强下,2种幼苗均通过大量增加非光化学淬灭(NPQ)、类胡萝卜素和叶绿素之比(Car/Chl)耗散过剩光能,降低单位鲜重叶绿素含量(Chl)以减少光能吸收,避免了光合机构光破坏。  相似文献

Hippocampal neuronal apoptosis accompanied by impairment of cognitive function occurs in primary diabetic encephalopathy. In this study, we investigated the neuroprotective mechanism of the iridoid glycoside, aucubin, using rats (n=8). Diabetes mellitus was induced in the rats by intraperitoneal (i.p.) injection of streptozotocin (60 mg/kg body weight). After 65 d, half of the DM rats were administered aucubin (5 mg/kg; i.p.) for 15 d, yielding treatment DM A. A third group of rats received no strepto- zotocin or aucibin, and served as controls (CON). Encephalopathy was assessed using Y-maze be- havioral testing. Rats were euthanized on Day 87, and hippocampi were excised for visual (light and transmission electron microscopic) and immunochemical (Western blot; immunohistochemical) as- sessments of the CA1 subfield for apoptosis and expression of regulatory proteins Bcl-2 and Bax. Treatment responses to all the parameters examined (body weight, plasma glucose, Y-maze error rates, pyramidal cell ultrastructure, proportions of apoptotic cells, levels of expression of Bcl-2 and Bax, and survivability of neuronal cells) were identical: there were highly significant differences between DM and CON groups (P<0.001), but the effects were significantly moderated (P<0.01) in DM A compared with DM. These findings confirm the association of apoptosis with the encephalopathic effects of diabetes mellitus, and suggest a major role of the expression levels of Bcl-2 and Bax in the regulation of apop- totic cell death. All of the results suggest that aucubin could effectively inhibit apoptosis by modulating the expressions of Bcl-2 and Bax genes.  相似文献

We recently discovered that vascular responsiveness to adrenomedullin (AM), a vasoactive hormone, decreases after hemorrhage, which is markedly improved by the addition of its binding protein AMBP-1. One obstacle hampering the development of AM/AMBP-1 as resuscitation agents in trauma victims is the potential immunogenicity of rat proteins in humans. Although less potent than rat AM, human AM has been shown to increase organ perfusion in rats. We therefore hypothesized that administration of human AM/AMBP-1 improves organ function and survival after severe blood loss in rats. To test this, male Sprague-Dawley rats were bled to and maintained at an MAP of 40 mmHg for 90 min. They were then resuscitated with an equal volume of shed blood in the form of Ringer's lactate (i.e., low-volume resuscitation) over 60 min. At 15 min after the beginning of resuscitation, human AM/AMBP-1 (12/40 or 48/160 microg/kg BW) were administered intravenously over 45 min. Various pathophysiological parameters were measured 4h after resuscitation. In additional groups of animals, a 12-day survival study was conducted. Our result showed that tissue injury as evidenced by increased levels of transaminases, lactate, and creatinine, was present at 4h after hemorrhage and resuscitation. Moreover, pro-inflammatory cytokines TNF-alpha and IL-6 were also significantly elevated. Administration of AM/AMBP-1 markedly attenuated tissue injury, reduced cytokine levels, and improved the survival rate from 29% (vehicle) to 62% (low-dose) or 70% (high-dose). However, neither human AM alone nor human AMBP-1 alone prevented the significant increase in ALT, AST, lactate and creatinine at 4h after the completion of hemorrhage and resuscitation. Moreover, the half-life of human AM and human AMBP-1 in rats was 35.8 min and 1.68 h, respectively. Thus, administration of human AM/AMBP-1 may be a useful approach for attenuating organ injury, and reducing mortality after hemorrhagic shock.  相似文献

The lin-12/Notch signaling pathway is conserved from worms to humans and is a master regulator of metazoan development. Here, we demonstrate that lin-12/Notch gain-of-function (gf) animals display precocious alae at the L4 larval stage with a significant increase in let-7 expression levels. Furthermore, lin-12(gf) animals display a precocious and higher level of let-7 gfp transgene expression in seam cells at L3 stage. Interestingly, lin-12(gf) mutant rescued the lethal phenotype of let-7 mutants similar to other known heterochronic mutants. We propose that lin-12/Notch signaling pathway functions in late developmental timing, upstream of or in parallel to the let-7 heterochronic pathway. Importantly, the human microRNA let-7a was also upregulated in various human cell lines in response to Notch1 activation, suggesting an evolutionarily conserved cross-talk between let-7 and the canonical lin-12/Notch signaling pathway.  相似文献
