The immunohistochemical localization of CRF- and neurophysin-containing neurons in the hypothalamus of the Mongolian gerbil was studied by means of the PAP technique. The CRF-immunoreactive fibers were detected mainly in the outer layer of the median eminence of intact adult male gerbils. The CRF-positive neurons respond to aminoglutethimide (Elipten, Ciba) administration by showing increased immunoreactivity and an increase in the number of stained cell bodies in the parvocellular division of the paraventricular nucleus. Aminoglutethimide treatment results also in an increase in the number of neurophysin-immunoreactive nervous fibers localized in the internal layer of the median eminence. However, CRF-immunoreactive fibers are observed mainly in the outer layer of the median eminence while neurophysin-immunopositive axons are seen predominantly in the internal layer of this region. Since the axons of paraventricular neurons run to the median eminence and their staining ability is changed due to aminoglutethimide, their involvement in the endocrine control of hypophysial ACTH release is postulated. 相似文献

125I-Wheat germ agglutinin (WGA) binding parameters of human urothelial cell lines of different grades of transformation (TGrll and TGrlll) were compared. The values of association constant (Ka) and the number of binding sites/cell for HCV29 (TGrll) cell line were about 3×106M–1 and over 4×107, respectively. Two TGrlll cell lines, HCV29T and Hu549 revealed lower values for Ka, and considerably higher numbers of binding sites/cell (about 3×108 and 2×108, respectively). Binding of125I-WGA to total cellular proteins resolved by SDS-PAGE and transferred to nitrocellulose showed multiple diffused bands in the range of 58–180 kDa. Some of these bands were characteristic for TGrll cells (124 kDa) or TGrlll cells (135 and 148 kDa).Abbreviations TGr

The HpaII methylase (M.HpaII) recognizes the sequence CCGG and methylates the inner cytosine residue. The MspI methylase (MspI) recognizes the same sequence but methylates the outer cytosine residue. Both methylases have the usual architecture of 10 well-conserved motifs surrounding a variable region, responsible for sequence specific recognition, that is quite different in the two methylases. We have constructed hybrids between these two methylases and studied their methylation properties. A hybrid containing the variable region and C-terminal sequences from M.MspI methylates the outer cytosine residue. A second hybrid identical to the first except that the variable region derives from the M.HpaII methylates the inner cytosine residue. Summary Seven white-rot fungi were examined for the production of ligninase, manganese peroxidase and laccase. All these enzymes were found inTrametes gibbosa andTrametes hirsuta. Only manganese peroxidase and laccase were produced byPycnoporus cinnabarinus,Coriolopsis polyzona,Stereum hirsutum,Dichomitus squalens andGanoderma valesiacum. All fungi decolorized Poly B-411 and Indulin AT plates with low-N medium. The profile-to-face chloroplast movement in the green alga Mougeotia has been induced by strong blue and near-ultraviolet light pulses (6 J m-2). Simultaneously, strong red or far-red light (10 W m-2) was applied perpendicularly to the inducing beam. The response was measured photometrically. Against the far-red background the reciprocity law was found to hold for pulse durations varying two orders of magnitude. The action spectrum exhibited a maximum near 450 nm and a distinct increase in near-ultraviolet. The time-course and the spectral dependence of pulse responses of chloroplasts in Mougeotia were similar to those recorded for other plants which are sensitive only to blue. This points to an alternative sensor system active in the short-wavelength region in addition to the phytochrome system.Abbreviations FR

The effect of hydrocortisone and dexamethasone on superovulation was examined in 12 cows. On the day PMSG was given, each animal received either the first of five daily doses of 250 mg succinate hydrocortisone or one injection of 30 mg dexamethasone. In the 48-hr interval between the injection of PMSG and PGF(2)alpha, the concentration of progesterone rose from 6.97 to 10.22 ng/ml in the experimental groups and only to about 2.8 ng/ml in the control group. In the following days progesterone increased even more, from 15.7 to 26.0 ng/ml seven days after estrus in the experimental group and to 19.25 ng/ml in the control group. The group which received dexamethasone had an average of 4.7 corpora lutea and one embryo flushed per animal. The hydrocortisone group had an average of 2.5 corpora lutea and one cow had two embryos. Summary Additon of pyruvate or leucine was found to be efficient for increasing the intracellular ratios of NADH/NAD and NADPH/NADP while reducing the coenzyme A concentration during the cultivation of Alcaligenes eutrophus. Microbiological transformation of benzaldehyde accomplished by the fungus Rhodotorula mucilaginosa immobilized in the ultrafiltration cell was studied. A polysulfone membrane formed on a sintered PVC support was used for the separation of the transformation product from the cellular material. Kinetic investigations have led to results which are typical of continuously fed stirred tank reactors (CFSTR)-the value of the maximum reaction rate (V(max)) and apparent Michaelis constant (K'(m)) are practically independent of the substrate retention time (calculated in terms of the flow intensity value). A strong relationship was found to occur between V(max) and biomass concentration in the reactor. Study of the apparent enzyme stability shows that the decrease in the biocatalyst activity is chiefly caused by penetration of the cells through the membrane. The experimental results were approximated in terms of the adopted mathematical model. Based on this model, the half-lives (t(1/2)) of enzyme activities were determined. The t(1/2) value varies from 35 to 82 days and depends both on the permeate flux through the membrane and on the separation properties of the membrane. 相似文献
