Preliminary investigations of the diatom genus Asterionella ralfsii W. Smith from Kejimkujik National Park, Nova Scotia indicate that its morphology differs from other reported forms. Mean cell length increased between the spring and the fall. Bimodal distribution of length classes occurred in several lakes and was not related to measured environmental variables. Priming of random hexamers in cDNA synthesis is known to show sequence bias, but in addition it has been suggested recently that mismatches in random hexamer priming could be a cause of mismatches between the original RNA fragment and observed sequence reads. To explore random hexamer mispriming as a potential source of these errors, we analyzed two independently generated RNA-seq datasets of synthetic ERCC spikes for which the reference is known. First strand cDNA synthesized by random hexamer priming on RNA showed consistent position and nucleotide-specific mismatch errors in the first seven nucleotides. The mismatch errors found in both datasets are consistent in distribution and thermodynamically stable mismatches are more common. This strongly indicates that RNA-DNA mispriming of specific random hexamers causes these errors. Due to their consistency and specificity, mispriming errors can have profound implications for downstream applications if not dealt with properly. 相似文献

Points of origin and pathways of spread are often poorly understood for introduced parasites that drive disease emergence in imperiled native species. Co‐introduction of parasites with non‐native hosts is of particular concern in remote areas like the Hawaiian Islands, where the introduced nematode Camallanus cotti has become the most prevalent parasite of at‐risk native stream fishes. In this study, we evaluated the prevailing hypothesis that C. cotti entered the Hawaiian Islands with poeciliid fishes from the Americas, and spread by translocation of poeciliid hosts across the archipelago for mosquito control. We also considered the alternative hypothesis of multiple independent co‐introductions with host fishes originating from Asia. We inferred conduits of introduction and spread of C. cotti across the archipelago from geographic patterns of mtDNA sequence variation and allelic variation across 11 newly developed microsatellite markers. The distribution of haplotypes suggests that C. cotti spread across the archipelago following an initial introduction on O'ahu. Approximate Bayesian Computation modeling and allelic variation also indicate that O'ahu is the most likely location of introduction, from which C. cotti dispersed to Maui followed by spread to the other islands in the archipelago. Evidence of significant genetic structure across islands indicates that contemporary dispersal is limited. Our findings parallel historical records of non‐native poeciliid introductions and suggest that remediating invasion hotspots could reduce the risk of infection in native stream fishes, which illustrates how inferences on parasite co‐introductions can improve conservation efforts by guiding responses to emerging infectious disease in species of concern. 相似文献

Conservation practitioners face difficult choices in apportioning limited resources between rare species (to ensure their existence) and common species (to ensure their abundance and ecosystem contributions). We quantified the opportunity costs of conserving rare species of migratory fishes in the context of removing dams and retrofitting road culverts across 1,883 tributaries of the North American Great Lakes. Our optimization models show that maximizing total habitat gains across species can be very efficient in terms of benefits achieved per dollar spent, but disproportionately benefits common species. Conservation approaches that target rare species, or that ensure some benefits for every species (i.e., complementarity) enable strategic allocation of resources among species but reduce aggregate habitat gains. Thus, small habitat gains for the rarest species necessarily come at the expense of more than 20 times as much habitat for common ones. These opportunity costs are likely to occur in many ecosystems because range limits and conservation costs often vary widely among species. Given that common species worldwide are declining more rapidly than rare ones within major taxa, our findings provide incentive for triage among multiple worthy conservation targets. 相似文献

Summary Immunity relies on the circulation of lymphocytes through many different tissues including blood vessels, lymphatic channels,
and lymphoid organs. The ability of lymphocytes to traverse the interstitium in both nonlymphoid and lymphoid tissues can
be determined in vitro by assaying their capacity to locomote through Type I collagen. In an attempt to characterize potential causes of microgravity-induced
immunosuppression, we investigated the effects of simulated microgravity on human lymphocyte function in vitro using a specialized rotating-wall vessel culture system developed at the Johnson Space Center. This very low shear culture
system randomizes gravitational vectors and provides an in vitro approximation of microgravity. In the randomized gravity of the rotating-wall vessel culture system, peripheral blood lymphocytes
did not locomote through Type I collagen, whereas static cultures supported normal movement. Although cells remained viable
during the entire culture period, peripheral blood lymphocytes transferred to unit gravity (static culture) after 6 h in the
rotating-wall vessel culture system were slow to recover and locomote into collagen matrix. After 72 h in the rotating-wall
vessel culture system and an additional 72 h in static culture, peripheral blood lymphocytes did not recover their ability
to locomote. Loss of locomotory activity in rotating-wall vessel cultures appears to be related to changes in the activation
state of the lymphocytes and the expression of adhesion molecules. Culture in the rotating-wall vessel system blunted the
ability of peripheral blood lymphocytes to respond to polyclonal activation with phytohemagglutinin. Locomotory response remained
intact when peripheral blood lymphocytes were activated by anti-CD3 antibody and interleukin-2 prior to introduction into
the rotating-wall vessel culture system. Thus, in addition to the systemic stress factors that may affect immunity, isolated
lymphocytes respond to gravitational changes by ceasing locomotion through model interstitium. These in vitro investigations suggest that microgravity induces non-stress-related changes in cell function that may be critical to immunity.
Preliminary analysis of locomotion in true microgravity revealed a substantial inhibition of cellular movement in Type I collagen.
Thus, the rotating-wall vessel culture system provides a model for analyzing the microgravity-induced inhibition of lymphocyte
locomotion and the investigation of the mechanisms related to lymphocyte movement. 相似文献

Fishes of the characid genus Roeboides are documented as lepidophagous, but the degree of specialization on scales varies between species. This study examines ontogenetic changes in morphology and diet of Roeboides dayi and Roeboides affinis from Venezuela. Juveniles of both species feed on microcrustacea and insects during the wet season, when availability of these resources is greatest. As aquatic habitat decreases during the dry season, insect and microcrustacea availability decreases and fish densities increase. As the fish densities increase, scale consumption becomes more profitable. Interspecific differences in the degree of specialization of the teeth and jaws was associated with the degree of scale feeding. Adult R. dayi consumed scales in approximately equal proportions to insects. Adult R. affinis consumes approximately 100% scales, and has more specialized head morphology than R. dayi. 相似文献

Two patients suffering from cardiovascular beriberi presented with different clinical manifestations. One had the classical features of a high cardiac output with raised jugular venous pressure and gross oedema. The other was in fulminating heart failure with clinical evidence of a low cardiac output but no peripheral oedema. The latter type of beriberi (shoshin) is rare. Cardiovascular beriberi has a high mortality when untreated. Both patients responded dramatically to thiamine, and this emphasizes the importance of considering thiamine deficiency as a cause of heart failure even when the cardiac output is low. 相似文献
