Antioxidant ebselen reduces oxidative damage in focal cerebral ischemia   总被引:13,自引:0,他引:13  
The antioxidant and neuroprotective potential of the glutathione peroxidase mimic ebselen has been investigated in experimental stroke. Intravenous ebselen (1 mg/kg/h) or vehicle infusion was started 45 min before permanent middle cerebral artery occlusion in the rat, and continued until the end of the experiment. The topography and extent of oxidative damage to the brain was assessed immunohistochemically using an antibody for DNA damage that identified hydroxylated products of 2'-deoxyguanosine (8-OHdG/8-oxodGuo) and an antibody for lipid peroxidation that identified the 4-hydroxynonenal histidine adduct (4-HNE). Ischemic damage was mapped and evaluated with standard histopathology. In the vehicle-treated rats immunopositive staining for both 8-oxodGuo and 4-HNE extended beyond the boundary of ischemic damage. In ebselen-treated rats, the extent of tissue immunopositive for 8-oxodGuo, and 4-HNE was less than that demonstrating ischemic damage confirming the antioxidant mechanism of action in vivo. In addition, ebselen treatment induced a 28% reduction in cortical ischemic damage (p <.02).  相似文献

Measurement of key morphological indices of chemostat cultures of Penicillium chrysogenum, by image analysis, was carried out manually at Strathclyde University and semi-automatically at Birmingham University using identical preserved samples. Using both methods, the value of the mean hyphal growth unit was found to decrease with decreasing dilution rate. Although similar trends were observed for data obtained at Strathclyde and Birmingham, the values of key morphological indices measured by semi-automated analysis were consistently higher than the values obtained by manual analysis. This discrepancy was as a result of the different analysis methods, particularly with respect to clump analysis. An electronic image transfer method is discussed which would allow analysis of a given image set at either site by either method. © Rapid Science Ltd. 1998  相似文献

The potential of uranium recovery from the dilute uranium ore bioleach solutions of the Elliot Lake district of Canada was examined using immobilized microbial biomass. Batch and continuous laboratory scale pilot plant experiments were carried out. The results have shown that the immobilized microbial biomass can successfully recover all of the uranium from dilute (less than 300 mg U/L) solutions. The uranium can subsequently be eluted producing a high uranium concentration eluate perhaps exceeding 5000 mg U/L. The biomass maintained its biosorption capacity of about 50 mg U/g over 12 examined successive adsorption-elution cycles with no apparent indication of failure.  相似文献

An unique stable isotope labelling experiment was conducted whereby mixtures of sulfate and sulfite of different isotopic compositions were metabolized by Clostridium pasteurianum. The results showed during reduction of 1 mM SO 3 = plus 1 mM SO 4 = , essentially all evolved H2S arose from the sulfite whereas in the case of cellular sulfur, 85% was derived from sulfite and the remainder from sulfate.  相似文献

Growth of Saccharomyces cerevisiae in minimal salts-glucose-SO4(2-) medium with varying concentrations of pantothenate (0-1000 microgram/L) produced changes in the cellular lipid content and in the ratio of saturated to unsaturated fatty acids. Substantial differences in SO4(2-) diffusion were also observed with changes in pantothenate concentration. During sulfate reduction, the delta 34S value of the evolved sulfide varied with the pantothenate concentration ranging from -31% in the absence of pantothenate to 0% at 400-1000 microgram/L pantothenate. The isotope selectivity is related to the effect of pantothenate concentration on cellular metabolism.  相似文献

The consequences of an oscillatory fluid shear mechanism on nutrient transport in bone during physical activity and ultrasonic therapy are discussed. During movement, periodic stress on bone creates transient pressure gradients that circulate interstitial fluid through calcified bone. A transport model derived from oscillatory Taylor-Aris dispersion phenomena was used to predict a ratio of effective-to-molecular diffusivity, K/D, for solutes of varying sizes up to 50 nm in diameter, in pores filled with interstitial fluid and pericellular matrix. The magnitude of the estimated transport enhancement depended on the molecular size, pore dimension, applied frequency and the displacement of the fluid during pressurization. For oscillation frequencies and amplitudes corresponding to those experienced during normal human activity, transport enhancements of up to 100 fold are expected for molecules larger than 5 nm in diameter. Enhancements of up to one order of magnitude, due to ultrasound stimulations in the MHz frequency range, are also expected for 7-nm-sized solutes. No effects are anticipated for ions, whose molecular diffusion time is too fast relative to the oscillation frequency. This model is expected to be useful for understanding differences in bone growth as a function of type of movement or to develop new physical therapies.  相似文献

Plant functional traits are thought to drive variation in primary productivity. However, there is a lack of work examining how dominant species identity affects trait–productivity relationships. The productivity of 12 pasture mixtures was determined in a 3‐year field experiment. The mixtures were based on either the winter‐active ryegrass (Lolium perenne) or winter‐dormant tall fescue (Festuca arundinacea). Different mixtures were obtained by adding forb, legume, and grass species that differ in key leaf economics spectrum (LES) traits to the basic two‐species dominant grass–white clover (Trifolium repens) mixtures. We tested for correlations between community‐weighted mean (CWM) trait values, functional diversity, and productivity across all plots and within those based on either ryegrass or tall fescue. The winter‐dormant forb species (chicory and plantain) had leaf traits consistent with high relative growth rates both per unit leaf area (high leaf thickness) and per unit leaf dry weight (low leaf dry matter content). Together, the two forb species achieved reasonable abundance when grown with either base grass (means of 36% and 53% of total biomass, respectively, with ryegrass tall fescue), but they competed much more strongly with tall fescue than with ryegrass. Consequently, they had a net negative impact on productivity when grown with tall fescue, and a net positive effect when grown with ryegrass. Strongly significant relationships between productivity and CWM values for LES traits were observed across ryegrass‐based mixtures, but not across tall fescue‐based mixtures. Functional diversity did not have a significant positive effect on productivity for any of the traits. The results show dominant species identity can strongly modify trait–productivity relationships in intensively grazed pastures. This was due to differences in the intensity of competition between dominant species and additional species, suggesting that resource‐use complementarity is a necessary prerequisite for trait–productivity relationships.  相似文献

Invasion ecology urgently requires predictive methodologies that can forecast the ecological impacts of existing, emerging and potential invasive species. We argue that many ecologically damaging invaders are characterised by their more efficient use of resources. Consequently, comparison of the classical ‘functional response’ (relationship between resource use and availability) between invasive and trophically analogous native species may allow prediction of invader ecological impact. We review the utility of species trait comparisons and the history and context of the use of functional responses in invasion ecology, then present our framework for the use of comparative functional responses. We show that functional response analyses, by describing the resource use of species over a range of resource availabilities, avoids many pitfalls of ‘snapshot’ assessments of resource use. Our framework demonstrates how comparisons of invader and native functional responses, within and between Type II and III functional responses, allow testing of the likely population-level outcomes of invasions for affected species. Furthermore, we describe how recent studies support the predictive capacity of this method; for example, the invasive ‘bloody red shrimp’ Hemimysis anomala shows higher Type II functional responses than native mysids and this corroborates, and could have predicted, actual invader impacts in the field. The comparative functional response method can also be used to examine differences in the impact of two or more invaders, two or more populations of the same invader, and the abiotic (e.g. temperature) and biotic (e.g. parasitism) context-dependencies of invader impacts. Our framework may also address the previous lack of rigour in testing major hypotheses in invasion ecology, such as the ‘enemy release’ and ‘biotic resistance’ hypotheses, as our approach explicitly considers demographic consequences for impacted resources, such as native and invasive prey species. We also identify potential challenges in the application of comparative functional responses in invasion ecology. These include incorporation of numerical responses, multiple predator effects and trait-mediated indirect interactions, replacement versus non-replacement study designs and the inclusion of functional responses in risk assessment frameworks. In future, the generation of sufficient case studies for a meta-analysis could test the overall hypothesis that comparative functional responses can indeed predict invasive species impacts.  相似文献
