OBJECTIVE: To investigate the reported increase in the expression of the glucose transporter GLUT-1 in borderline and malignant ovarian epithelial tumors and its relationship to prognosis. STUDY DESIGN: In this study, areas in which immunohistochemical membranous staining with GLUT-1 were most evident were selected, and the proportions of GLUT-1 expression in 46 benign, 11 borderline and 42 malignant cases of ovarian epithelial tumors were determined quantitatively with a computer and Zeiss Vision KS 400 3.0 (G?ttingen, Germany) for Windows (Microsoft, Redmond, Washington, U.S.A.) image analysis. RESULTS: GLUT-1 expression was determined in all borderline tumors (11 of 11) and in 97.6% of malignant tumors (41 of 42). No GLUT-1 expression was observed in benign tumors. The intensity of GLUT-1 staining was lower in borderline tumors than in malignant cases. This was statistically significant (p = 0.005). As differentiation in malignant tumors increased, proportions of GLUT-1 expression showed a relative increase, but this difference was not statistically significant (p = 0.68). CONCLUSION: When GLUT-1 expression in borderline and malignant ovarian epithelial tumors was analyzed against prognosis, no statistically significant difference was identified. Assessment of GLUT-1 expression using the image analysis program was more reliable, with higher reproducibility than in previous studies.  相似文献

Our purpose in this study was to investigate the protective effects of selenium and vitamin E on the blood–brain barrier (BBB) permeability in rats with convulsion under hyperthermic conditions. To eliminate the effect of sex on BBB, we performed our study on 4- to 5-week-old prepubertal rat pups. Evans-blue was used as a BBB tracer. Convulsions were induced by administration of i.p. pentylenetetrazol. In the selenium group, 4 ppm selenium was added to the drinking water for 4–5 weeks. Vitamin E was administered at 700 mg/kg ip. It was shown that the convulsions, both under normothermic and hyperthermic conditions, caused widespread increase in the BBB permeability (p < 0.05). In addition, a significant difference was observed among female and male rats (f [1, 102] = 6.387, p < 0.05). In convulsions under normothermic conditions, there was a further increase in the BBB permeability (F[3, 102] = 43.534, p < 0.001) and a greater increase of permeability in males compared to females (F[1, 102] = 6.387, p < 0.05). Selenium and vitamin E significantly decreased the BBB destruction caused by convulsions under hyperthermic conditions in males (p < 0.05). Treatment with selenium or vitamin E has beneficial effects on the BBB breakdown during convulsions. But gender differences are very important in BBB permeability under pathological conditions and antioxidant treatments.  相似文献

Acinetobacter sp. strain ADP1 is a nutritionally versatile soil bacterium closely related to representatives of the well-characterized Pseudomonas aeruginosa and Pseudomonas putida. Unlike these bacteria, the Acinetobacter ADP1 is highly competent for natural transformation which affords extraordinary convenience for genetic manipulation. The circular chromosome of the Acinetobacter ADP1, presented here, encodes 3325 predicted coding sequences, of which 60% have been classified based on sequence similarity to other documented proteins. The close evolutionary proximity of Acinetobacter and Pseudomonas species, as judged by the sequences of their 16S RNA genes and by the highest level of bidirectional best hits, contrasts with the extensive divergence in the GC content of their DNA (40 versus 62%). The chromosomes also differ significantly in size, with the Acinetobacter ADP1 chromosome <60% of the length of the Pseudomonas counterparts. Genome analysis of the Acinetobacter ADP1 revealed genes for metabolic pathways involved in utilization of a large variety of compounds. Almost all of these genes, with orthologs that are scattered in other species, are located in five major 'islands of catabolic diversity', now an apparent 'archipelago of catabolic diversity', within one-quarter of the overall genome. Acinetobacter ADP1 displays many features of other aerobic soil bacteria with metabolism oriented toward the degradation of organic compounds found in their natural habitat. A distinguishing feature of this genome is the absence of a gene corresponding to pyruvate kinase, the enzyme that generally catalyzes the terminal step in conversion of carbohydrates to pyruvate for respiration by the citric acid cycle. This finding supports the view that the cycle itself is centrally geared to the catabolic capabilities of this exceptionally versatile organism.  相似文献

Developmental processes and stress-induction activate many key proteins in plants such as metacaspase which regulate programmed cell death (PCD). In this study, identification of barley metacaspases and their possible roles upon boron (B)-induction was investigated by using in silico and wet-lab methods. Genome-wide analysis revealed that barley genome harbor ten metacaspases which divided into three groups: Type-I, -I* and -II. Segmental and tandem duplication contributed their expansion. Metacaspase-specific catalytic residues (His and Cys) were found to be altered in HvMC1, 2, and 4, in which His exchanged to Meth or Ala, critical for their activity and substrate selectivity. Cis-acting elements were found to be associated with three main processes: stress response, growth/development, and light response. Digital expression analysis from eight tissues revealed tissue specific metacaspase expressions. In addition, RT-qPCR analysis conducted in appropriate (50 µM) and excess-B (1 and-3 mM) conditions in different time points (3 and 10 days). Toxic level of B caused growth inhibition and chlorosis which appeared at the leaf tips. Also, PCD initiation was detected after 3 days of excess-B exposure. Digital expression and qPCR analysis agreed with each other that HvMC4 expression was significantly increased upon excess-B supplementation. In opposite, HvMC5 was down-regulated in the leaf zones which was another critical B-responsive gene in barley. Hence, HvMC4 and HvMC5 seem to have antagonistic effect during PCD regulation. These results can provide insights for metacaspase functionality in barley, not only limited for B-induction but also various kinds of PCD-causing conditions.  相似文献

This research provides a summary of the general information contained in studies on the synthesis, characterization, photophysical characteristics, and possible applications of phenanthroimidazole derivative compounds. The focus of this study revolves around the topic of utilization in technological fields such as sensors, solar cells, optical brighteners, and organic light-emitting diodes, and covers significant studies on mentioned topics. We anticipate that this study will provide an outline for researchers aiming to further examine fluorescent organic compounds for technological innovations.
