MDCK cells are grown on various substrates (Thermanox pure, extracellular matrix (ECM), dried or wet collagen type I or type III), on floating collagen and enclosed in collagen gels, and their differentiation behaviour is investigated electron microscopically. The cells grown on ECM or dried collagen (type I and type III) do not show any changes as compared with the controls (Thermanox). Differentiation processes can only be observed when the cells are grown on wet collagen (type I and type III), especially on floating collagen and enclosed in collagen gels. These differentiation processes comprise changes in the cell shape, an increase in the number of microvilli, an increase in the length of the lateral contact zone with the formation of gap junctions and desmosomes, and an increase in the number and size of the cell organelles. A basement membrane only develops in the form of short segments. Moreover, on floating collagen and in collagen gels three-dimensional, organoid structures develop: cell aggregates with central lumina and tubuli. They are formed by cuboid cells that also exhibit indications of differentiation. Basement membrane fragments occur more often and are longer. It can be concluded from these findings that the chemical structure of the substrate does not play the primary role in the described process. It is rather the physical properties, probably the plasticity, that are of significance. Due to this property the cells change their shape and the contact areas increase in size. The establishment of contacts might be the triggering factor for differentiation. Organoid structures with lumina develop when the apical surface comes into contact with other cells or collagen gels. The pronounced tendency towards polarization necessitates a re-arrangement of three-dimensionally growing cells to structures with lumina. The formation of the basement membrane is the result and not the cause of differentiation.  相似文献

A novel cytochrome ba complex was isolated from aerobically grown cells of the thermoacidophilic archaeon Acidianus ambivalens. The complex was purified with two subunits, which are encoded by the cbsA and soxN genes. These genes are part of the pentacistronic cbsAB-soxLN-odsN locus. The spectroscopic characterization revealed the presence of three low-spin hemes, two of the b and one of the a(s)-type with reduction potentials of +200, +400 and +160 mV, respectively. The SoxN protein is proposed to harbor the heme b of lower reduction potential and the heme a(s), and CbsA the other heme b. The soxL gene encodes a Rieske protein, which was expressed in E. coli; its reduction potential was determined to be +320 mV. Topology predictions showed that SoxN, CbsB and CbsA should contain 12, 9 and one transmembrane alpha-helices, respectively, with SoxN having a predicted fold very similar to those of the cytochromes b in bc(1) complexes. The presence of two quinol binding motifs was also predicted in SoxN. Based on these findings, we propose that the A. ambivalens cytochrome ba complex is analogous to the bc(1) complexes of bacteria and mitochondria, however with distinct subunits and heme types.  相似文献

The oxidation and reduction of elemental sulfur and reduced inorganic sulfur species are some of the most important energy-yielding reactions for microorganisms living in volcanic hot springs, solfataras, and submarine hydrothermal vents, including both heterotrophic, mixotrophic, and chemolithoautotrophic, carbon dioxide-fixing species. Elemental sulfur is the electron donor in aerobic archaea like Acidianus and Sulfolobus. It is oxidized via sulfite and thiosulfate in a pathway involving both soluble and membrane-bound enzymes. This pathway was recently found to be coupled to the aerobic respiratory chain, eliciting a link between sulfur oxidation and oxygen reduction at the level of the respiratory heme copper oxidase. In contrast, elemental sulfur is the electron acceptor in a short electron transport chain consisting of a membrane-bound hydrogenase and a sulfur reductase in (facultatively) anaerobic chemolithotrophic archaea Acidianus and Pyrodictium species. It is also the electron acceptor in organoheterotrophic anaerobic species like Pyrococcus and Thermococcus, however, an electron transport chain has not been described as yet. The current knowledge on the composition and properties of the aerobic and anaerobic pathways of dissimilatory elemental sulfur metabolism in thermophilic archaea is summarized in this contribution.  相似文献

Background

Drowsiness compromises driving ability by reducing alertness and attentiveness, and delayed reaction times. Sleep-related car crashes account for a considerable proportion of accident at the wheel. Narcolepsy type 1 (NT1), narcolepsy type 2 (NT2) and idiopathic hypersomnia (IH) are rare central disorders of hypersomnolence, the most severe causes of sleepiness thus being potential dangerous conditions for both personal and public safety with increasing scientific, social, and political attention. Our main objective was to assess the frequency of recent car crashes in a large cohort of patients affected with well-defined central disorders of hypersomnolence versus subjects from the general population.

Methods

We performed a cross-sectional study in French reference centres for rare hypersomnia diseases and included 527 patients and 781 healthy subjects. All participants included needed to have a driving license, information available on potential accident events during the last 5 years, and on potential confounders; thus analyses were performed on 282 cases (71 IH, 82 NT2, 129 NT1) and 470 healthy subjects.

Results

Patients reported more frequently than healthy subjects the occurrence of recent car crashes (in the previous five years), a risk that was confirmed in both treated and untreated subjects at study inclusion (Untreated, OR = 2.21 95%CI = [1.30–3.76], Treated OR = 2.04 95%CI = [1.26–3.30]), as well as in all disease categories, and was modulated by subjective sleepiness level (Epworth scale and naps). Conversely, the risk of car accidents of patients treated for at least 5 years was not different to healthy subjects (OR = 1.23 95%CI = [0.56–2.69]). Main risk factors were analogous in patients and healthy subjects.

Conclusion

Patients affected with central disorders of hypersomnolence had increased risk of recent car crashes compared to subjects from the general population, a finding potentially reversed by long-term treatment.  相似文献

Zusammenfassung 1. Die Behandlung der nativen und formolfixierten Sehnenfibrillen mit einer ammoniakalischen Silberlösung führt immer zu einer Einlagerung von Silberpartikeln in den D-Teilen der Fibrillen.2. Bei den nativen Fibrillen liegen die Silberkörner in einem, zwei oder drei Streifen im D-Teil.3. In den formolfixierten Fibrillen ist das Silber nur in einem Streifen vorhanden.4. Die Behandlung der nativen und formolfixierten Sehnenfibrillen mit anderen Silbersalzen führt zu keiner Versilberung der Fibrillen.5. Die Behandlung der nativen Sehnenfibrillen mit neutraler Kochsalzlösung oder Trypsin und anschließender Versilberung führt zu keiner wesentlichen Änderung des Silberbildes.6. Hyaluronidase-, Citratpuffer- und Perjodateinwirkung auf native Sehnenfibrillen mit anschließender Versilberung führt zu keiner Innenversilberung der D-Teile.7. Acetylierung und Behandlung mit Bisulfit der nativen Fibrillen und anschließender Versilberung mit ammoniakalischer Silberlösung verhindert eine Innenversilberung der D-Teile.8. Die formolfixierten Fibrillen zeigen eine Innenversilberung der D-Teile nach einer Vorbehandlung mit einer neutralen Kochsalzlösung, Citratpuffer, Hyaluronidase, Trypsin und Perjodat. Nur die Acetylierung und die Behandlung mit Bisulfit verhindert eine Innenversilberung.9. Die Innenversilberung der Sehnenfibrillen durch eine ammoniakalische Silberlösung wird weder durch Licht noch durch Chloride oder lichtempfindliche Silbereiweißverbindungen hervorgerufen.10. Die Versilberung in den D-Teilen wird durch Stoffe in den Fibrillen bewirkt, die Silber aus einer ammoniakalischen Silberlösung ausfällen können.11. Die reduzierenden Stoffe haben enge Beziehungen zur citratlöslichen Fraktion und sind perjodat- und hyaluronidaseempfindlich. Formalinfixierung beeinflußt diesen Versilberungsmodus durch ein vermehrtes Auftreten von Querbindungen.12. Die Sonderstellung der ammoniakalischen Silberlösung für die Innenversilberung wird diskutiert. Sie kann stereochemische Gründe haben oder durch die große Beständigkeitskonstante erklärt werden.13. Das Ausfallen von metallischem Silber in den D-Teilen der Sehnenfibrillen kann nicht mit dem photographischen Prozeß in Verbindung gebracht werden. Das gilt auch für die Bindegewebsversilberung nachGömöri.14. Die Silberorte in den D-Teilen lassen sich nur teilweise mit den bekannten Querstreifungsbildern nach Osmium- oder Phosphorwolframsäurefixierung in Beziehung setzen.
Summary 1. After treatment of native or formalin-fixed tendon fibrils with an ammoniacal silver solution, silver particles are deposited in the D-bands of the fibrils. In the native fibrils these are arranged in one, two or three striae per band, but after formalin fixation they lie in one stria only.2. No external reducing agent is necessary for the production of the particles.3. Pretreatment of native fibrils with neutral salt solution or with trypsin has no effect on subsequent silvering. On the other hand, silvering is abolished by treatment with hyaluronidase, citrate buffer or periodate and also by acetylation and bisulphite.4. Formalin-fixed fibrils show the silvering effect after all these procedures except acetylation or bisulphite treatment.5. It is postulated that silvering of the D-bands is due to reducing substances which can precipitate silver from ammonical solutions and that formalin influences the process by the production of cross linkages.

Zusammenfassung An einer Reihe von normalen und pathologisch veränderten Organen wird die Porenstruktur, Porenbildung und Porenrückbildung elektronenmikroskopisch untersucht. Die Endothelpore ist eine Diskontinuität in der Endothelwand mit einem sehr konstanten Durchmesser von 500 Å. Das Diaphragma ist nur mit der äußeren Membranlamelle am Porenrand verbunden. Diaphragmalose Poren sind etwas größer (Ø650 Å), zeigen einen glatten Porenrand und kommen besonders in verdichteten Endothelteilen vor. Frustrane Poren münden blind in Vakuolen oder liegen in porösen Endothelfalten im Gefäßlumen. Die eigentliche Bildung der Poren geschieht immer in abgeflachten ( 800 Å) Endothelteilen. Die vorbereitende Abflachung geht jedoch in den verschiedenen Endothelzonen (Periksryon, dicker Endothelwand, Cytoplasmainseln) unterschiedliche Wege. Alle diese Vorgänge stellen Vesikulation in besonderer Lage und mit besonderer Fusionsrichtung der Vesikel dar. Wegen dieser Unterschiede wird die Endothelwand in 4 Zonen eingeteilt: Perikaryon; dicke, porenlose Wandteile mit cytoplasmatischen Vesikeln; dünne, porenhaltige Teile ohne Zellorganellen; dicke Cytoplasmainseln, die die porösen Wandteile voneinander trennen. Der Vorgang der Porenrückbildung bleibt unklar. Vielleicht besteht er in der Faltung des Endothels, die zu porösen Vakuolen führt. Die Porenbildung verändert die Endotheloberfläche nicht, kann aber das Cytoplasmavolumen vermindern. Der Aufbau des Diaphragmas sowie der Mechanismus und die auslösenden Faktoren der Porenbildung werden diskutiert.
Summary Ultrastructure, formation and disappearance of endothelial pores was studied electron microscopically in several normal or experimentally changed organs. Pores being discontinuies in the endothelial wall have a very constant diameter of 500 Å. The diaphragm at the margin of the pore is in contact with only the outer lamella of the unit membrane. Pores without a diaphragm being somewhat larger (Ø 650 Å) show a smoother margin and are to be found in endothelia with dense cytoplasm. Frustrated pores form blind openings in vacuoles or are situated in porous endothelial folds within the vessel lumen. Pores alway are formed in flattened parts of endothelium ( 800 Å). In thick capillary walls the endothelium previously is flattened by vesiculation, which differs in different zones of the wall (Perikaryon, thick continuous endothelium, and cytoplasmic islands in porous capillaries) in location and direction of vesicle fusion. Because of these differences the endothelial wall is divided in 4 zones: Perikaryon; thick parts without pores containing cytoplasmic vesicles etc.; flattened, porous parts containing no cytoplasmic organelles; thick cytoplasmic islands which separate porous parts. The process removing pores is not clear. Perhaps they are removed by folding of the endothelial surface and formation porous vacuoles. Formation of pores dosn't enlarge or reduce the surface, but may reduce the cytoplasmic volume of endothelium. The nature of diaphragm as well as the mechanism and releasing factors of the formation of pores are discussed.

The anti-angiogenic properties of thalidomide have led to the use of the agent as a remedy for multiple myeloma. Nevertheless, the anti-angiogenic moiety of thalidomide remains unidentified. In this study we examined the anti-angiogenic effects of thalidomide in an in vitro model using a three-dimensional collagen gel culture. Angiogenesis was significantly inhibited when the culture was treated with thalidomide plus cytochrome P-450 (CYP2B4), and the migrating cells and tubules were positive for active-caspase-3 in an accompanying immunohistochemical investigation. Transmission electron microscopic observation also confirmed that active-caspase-3-positive cells demonstrated apoptotic characteristics. This study is the first to morphologically demonstrate the effect of thalidomide in directly inducing the apoptosis of new tubules and migrating cells on a three-dimensional collagen gel culture of aorta. Taken together with earlier findings, our new results indicate that the thalidomide-induced inhibition of angiogenesis involves apoptosis in addition to the suppression of TNF- and inhibition of cell migration from aorta explants, i.e., the factors important for capillarogenesis.  相似文献

Abstract: Labeled palmitic acid ([16-14C]palmitate) (0).5 μCi) was injected into rat sciatic nerves in vivo to characterize thc incorporation of this fatty acid into complex peripheral nerve lipids after time lapses of 1 min to 2 weeks. For the first 30 min after intraneural injection, the label was concentrated in nerve diglycerides. Thereafter, the relative diglyccride label declined rapidly, and phospholipid radioactivity rose steadily. After 120 min, phospholipids contained over 70% of the total lipid radioactivity. Among the phospholipids, phosphatidylcholine had the largest percentage of total phospholipid label, and acylation of lysophosphatidylcholine accounted for approximately 75% of this label. With time, there was conversion of [16-14C]palmitate to other long-chain fatty acids by elongation and desaturation. Phosphatidic acid was labeled also, suggesting the operation of the de novo biosynthetic mechanism. However, the specific radioactivity of 1,2-diacylglycerol was much higher than that of phosphatidic acid, suggesting phosphorylation of diglycerides by diglyceride kinase. After nerve section and survival of 2 h to 50 days, the injection of [16-14C]palmitate into the degenerating distal segment revealed an overall decline of phospholipid labeling and a commensurate increase of triglyceride radioactivity. Phosphatidylcholine in degenerating nerve contained a larger percentage of the fatty acid label than that in normal nerve. Almost all of the labeling was due to acylation of lysophosphatidylcholine, implying a much smaller contribution of the de novo pathway. Phosphatidylethanolamine and phosphatidylserine showed a relative loss of radioactivity. The changes were apparent at 1 day, but not at 2 h, suggesting loss of homeostatic control, presumably by interruption of axonal flow. An incidental observation was the stimulation of phosphatidylcholine biosynthesis by acylation of lysophosphatidylcholine in the contralateral unoperated sciatic nerve.  相似文献
