Serum sialic acid levels and selected mineral status in patients with type 2 diabetes mellitus 总被引:2,自引:0,他引:2
The aim of the present study was to investigate whether altered serum total sialic acid (TSA), lipid-associated sialic acid
(LSA), copper (Cu), manganese (Mn), zinc (Zn), chromium (Cr), iron (Fe), and magnesium (Mg) levels had an interactive connection
with diabetes and also whether they were correlated with each other in diabetic patients. Two study groups (control and type
2 diabetic subjects) were included. Two hundred patients (108 female and 92 male), diagnosed and treated for type 2 diabetes
in the Yuzuncu Yil University Hospital (Van, Turkey), were selected consecutively to represent type 2 diabetic patients. Fifty
healthy individuals (29 female and 21 male) served as the control group matched for age, sex, body mass index, and smoking
status were selected from hospital staff and other outpatient clinics. All participants had not taken vitamin or mineral supplements
for at least 2 wk before sampling. Blood samples were drawn after an overnight fasting in both groups for the determination
of serum glucose, TSA, LSA, Cu, Zn, Mn, Cr, Fe, and Mg. It was found that diabetics had higher TSA, LSA, Fe, Mn, Fe/Zn, and
Cu/Zn levels, and lower Zn and Mg levels than those of controls. Although, Cu levels were higher, and Cr levels were lower
in total and male diabetic patients, they were not different in female diabetic patients than in controls. The Cu/Fe ratio
was lower in total and female diabetic patients, but not different in male diabetic patients than controls. The Zn/Cr ratio,
on the other hand, was not different in diabetics than in controls. There was only a positive correlation between Fe-Mn levels
in male diabetic patients. There was a negative correlation in LSA-Mn, Fe-Cu, Cu-Fe/Zn, and Mn-Cu/Zn levels in total diabetic
patients. There was a positive correlation in TSA-Cr, TSA-Mg, LSA-Cu/Fe, LSA-Zn/Cr levels, and a negative correlation in TSA-Cu/Zn,
LSA-Mn, Fe-Cu, Mn-Cu, Cu-Fe/Zn, Fe-cholesterol, and Cr-cholesterol in female diabetic patients. Our results showed that TSA,
LSA, and selected minerals have interactive connections with diabetes mellitus (DM). There are also many sex-related positive
or negative correlations between the altered parameters in diabetic patients. These parameters might be used as diagnostic

This study was designed to investigate the effects of Cd exposure on the glycogen localization in the placenta and in fetal
and maternal livers in streptozotocin (STZ)-induced-diabetic pregnant rats. Ninety-nine virgin female Wistar rats (200–220
g) were mated with 33 males for at least 12 h. From the onset of pregnancy, the rats were divided into four experimental groups
(control, Cd treated, STZ treated, and Cd+STZ treated). The Cd-treated group was injected subcutaneously daily with CdCl2 dissolved in isotonic NaCl, starting at the onset of pregnancy throughout the experiment. Diabetes was induced on d 13 of
pregnancy by a single intraperitoneal injection of STZ in the STZ-treated group. In addition to the daily injection of Cd,
a single intraperitoneal injection of STZ was also given on d 13 of pregnancy in the Cd+STZ-treated group. The rats received
the last injection 24 h before being sacrificed and 10 randomly selected rats in each group were sacrificed on d 15 and d
20 of pregnancy. Blood samples were taken for determination of the serum glucose and insulin levels. Fetal and maternal livers
of sacrificed rats in all groups were harvested on d 15 and d 20 of pregnancy, whereas placentas were harvested only on d
20 of pregnancy for histochemical examination. Although both Cd and STZ caused hyperglycemia and decreased insulin secretion,
Cd-alone treatment increased the glycogen content only in the placental labyrinth, whereas STZ-alone treatment increased the
glycogen content only in the maternal part of the placenta. Increased glycogen localization was observed in both the placental
labyrinth and the maternal part of placenta when Cd and STZ were given together. Fetal and meternal livers of control and
other treatment groups were not different regarding the glycogen content on d 15 or d 20 of pregnancy. It was concluded that
Cd exposure during pregnancy might produce a glycogen localization in the placenta of diabetic rats. However, the function
and the mechanisms of increased glycogen contents in the placenta of Cd-exposed pregnant diabetic rats remain unclear and

The vitamin D receptor (VDR) is a member of the steroid/retinoid receptor superfamily of nuclear receptors that controls mineral ion homeostatis and has potential tumor-suppressive functions for various cancer types, specifically prostate cancer. A VDR ablated transgenic animal model (VDDRII, vitamin D-dependent rickets type II) has been developed and the animals typically have various diseases including, hypocalcemia, hyperparathyroidism, rickets, osteomalacia, and alopecia. This transgenic mouse system provides us with a model to decipher the influences of the VDR on prostatic growth and function. VDRs are abundant both in prostatic epithelial and stromal cells, and vitamin D signaling can be studied in this model. Although, there were no gross differences between the prostate tissue of the experimental and control groups, VDR null mice showed fat necrosis and individual cell apoptosis in the periprostatic adipose tissue. This indicates a possible role of VDR in the signaling pathways resulting the prostate. This may be particularly attractive for VDR targets for the inhibition of cancer progression using VD(3) and its analogs as potential chemo-preventive agents. 相似文献

We investigated the effects of Nigella sativa on apoptosis and gamma-aminobutyric acid (GABAA) receptor density in cerebral cortical and hippocampal neurons in a pentylenetetrazol (PTZ)-induced kindling model in rats. The PTZ kindling model was produced by injecting PTZ in subconvulsive doses to rats on days 1, 3, 5, 8, 10, 12, 15, 17, 19, 22 and 24 of the study into animals of PTZ treated (PTZ) and PTZ + N. sativa treated (PTZ + NS) groups. Clonic and tonic seizures were induced by injecting a convulsive dose of PTZ on day 26 of the study. Rats in the PTZ + NS group were treated also with a 10 mg/kg methanolic extract of N. sativa 2 h before each PTZ injection. Rats in the control group were treated with 4 ml/kg saline. The number of neurons that expressed GABAA receptors in the hippocampus and cerebral cortex of rats in the PTZ and PTZ + NS groups increased significantly. There was no significant difference in the number of GABAA receptors between the PTZ and PTZ + NS groups. GABAA receptor density of the neurons in the cerebral cortex, but not hippocampus, was increased in PTZ group compared to controls. We observed a significant increase in the number of apoptotic neurons in the cerebral cortex of rats of both the PTZ and PTZ + NS groups compared to controls. We observed a significant decrease in the number of the apoptotic neurons in the cerebral cortex of rats in the PTZ + NS group compared to the PTZ group. N. sativa treatment ameliorated the PTZ induced neurodegeneration in the cerebral cortex as reflected by neuronal apoptosis and neuronal GABAA receptor frequency. 相似文献

The recombinant circumsporozoite protein (CS) based vaccine, RTS,S, confers protection against Plasmodium falciparum infection in controlled challenge trials and in field studies. The RTS,S recombinant antigen has been formulated with two adjuvant systems, AS01 and AS02, which have both been shown to induce strong specific antibody responses and CD4 T cell responses in adults. As infants and young children are particularly susceptible to malaria infection and constitute the main target population for a malaria vaccine, we have evaluated the induction of adaptive immune responses in young children living in malaria endemic regions following vaccination with RTS,S/AS01(E) and RTS,S/AS02(D). Our data show that a CS-specific memory B cell response is induced one month after the second and third vaccine dose and that CS-specific antibodies and memory B cells persist up to 12 months after the last vaccine injection. Both formulations also induced low but significant amounts of CS-specific IL-2(+) CD4(+) T cells one month after the second and third vaccine dose, upon short-term in vitro stimulation of whole blood cells with peptides covering the entire CS derived sequence in RTS,S. These results provide evidence that both RTS,S/AS01(E) and RTS,S/AS02(D) induced adaptive immune responses including antibodies, circulating memory B cells and CD4(+) T cells directed against P. falciparum CS protein. Diabetes Mellitus (DM) is a multisystemic disorder with serious complications and these patients may also have serious problems with their oral cavity probably because of the microangiopathic and neuropathic complications. In diabetic patients, there may be several problems of the oral cavity such as gingivitis, periodontitis, candidiasis, glossitis, oral ulcerations, loss of taste sensations, opportunistic infections and several other conditions dependent on these. One of the recent theories about complications in DM is the contribution of reactive oxygen radicals. Paraoxonase (PON1) is an enzyme that is synthesized in liver and having the capability of hydrolasing the active metabolite of an insectisid, parathion. Previously it was shown that there are two polymorphic areas on the PON1 gene: one causing a Leu → Met substitution at 55th position, the other causing Gln → Arg at the 192nd position. We investigated the differences in PON activities related to the oral lesions in Type 2 diabetics and control subjects to see their relationships with PON1 activity levels and the two main gene polymorphisms of PON1 genes, PON1 192 and PON1 55. We had 51 patients and 53 healthy subjects used in the study. PON activity was significantly decreased in Type 2 DM group compared to the control group. Neither PON1 192 nor PON1 55 genotypes had any differential effect on PON1 enzyme activity levels in either group. Acute ultraviolet-B (UV-B) irradiation is known to act as an initiator in the formation of reactive oxygen species. These
oxygen products are highly reactive and they are able to cause irreversible damage to cellular components. Oxygen free radicals
are normally neutralized by very efficient systems in the body. These include antioxidant enzymes like superoxide dismutase
(SOD). In a healthy subject, there is a balance between free radicals and the levels of antioxidants. In some pathological
conditions such as oxidative stress, the level of antioxidants is significantly reduced. The skin contains relatively high
levels of zinc (Zn), an essential element known to be a cofactor in some metabolic pathways. Zinc has also been reported to
have antioxidant properties.
In the present study, we investigated the effect of ginkgo biloba extract (Gbe), a potent free-radical scavenger, on UV-B-irradiated
skin by measuring SOD activity and Zn levels in the skin, before and after treatment. The SOD activity was decreased after
UV-B exposure, in comparison with the control group (p<0.05). After Gbe treatment, the SOD activity increased (p<0.05) as compared with the untreated UV-B irradiated group. The Zn levels changed in the same pattern as the SOD activity
