Summary Polymorphism and inheritance of wheat storage protein, gliadin, of durum (macaroni) and carthlicum wheats have been studied. Analysis of gliadin in 78 cultivars and in F2 seeds of intercultivar crosses of durum wheat revealed three different chromosome 1A-encoded blocks of components similar to those found in common wheat (GLD1A2, GLD1A18, GLD1A19). Most of the durum cultivars studied had these three blocks; GLD1A2 was also frequent in common wheat. In contrast, all chromosome 1B-encoded blocks of durum clearly differed in component composition from those found in common wheat. Therefore, durum could not be an ancestor or a derivate of recent bread wheat. Analysis of gliadin in the collection of carthlicum wheat (14 accessions) revealed several suspected chromosome 1A, 1B, and 6A-controlled blocks, some of which were similar to those in common wheat, while others were different. Therefore, carthlicum is likely to be an ancestor or a derivate of some forms of bread wheat. There were also chromosome 1A and 6A-, but not 1B-encoded blocks which were identical in durum and carthlicum wheats. The results confirm that all three wheats share the same genome A, but emphasize the heterogeneity of genotypes among donors of this genome. Discovery of identical blocks in tetraploids and hexaploids indicates polyphyletic [from different genotypes of donor (s)] origin of these wheats.  相似文献

The effects of quisqualic acid (QA), N-methyl-D-aspartate (NMDA), and a number of NMDA and non-NMDA receptor antagonists on background and induced activity in afferent nerve fibers were investigated in skates by means of bath application to the basal membrane of electroreceptors (ampullae of Lorenzini). Perfusion with physiological saline containing QA or NMDA (minimum concentrations required: 10–8 and 10–5 M respectively) was found to exert an excitatory effect on afferent activity. Aminoadipate and aminophosphonobutyrate had no effect on synaptic transmission, which was blocked by aminophosphonovalerate, however. Raising magnesium ion concentration (of 30 mM) led to blockade of NMDA-induced response without changing that produced by QA. Aminophosphonovalerate blocked NMDA response and partially reduced the effects of L-aspartic acid. Glutamyl glycine produced blockade of synaptic transmission. The findings obtained would point to synaptic sensitivity to the action of amino acid agonists (QA and NMDA) in the ampullae of Lorenzini.Neurocybernetics Research Institute, Rostov-on-Don. The effects of cadmium ions on synaptic transmission in the frog tectum were investigated in acute experiments using quantal EEG recording techniques (readings of extracellular monosynaptic potential induced by activating the synapses of a single axon) [1]. Superfusion of the tectum by 10–200 µM CdCl2 reversibly inhibits EEG quanta, reduces their duration (measured at 50% amplitude level) and increases synaptic delay. The results of this study confirm the concept formed from in vitro experiments of votage-dependent calcium channels as one of the likely Cd2+ action sites at central synapses. The effects of stimulating the periaqueductal gray matter (PAG) on two types of startle reflex (spino-bulbo-spinal reflex produced by intensive stimulation of the peripheral nerves and low-threshold tactile spino-reticulo-spinal reflex) as well as high-threshold jaw-opening reflex arising in response to tooth pulp stimulation were investigated in cats anesthetized with chloralose. Simulating most PAG test sites led to pronounced inhibition of jaw-opening reflex, profound depression of spino-bulbo-spinal reflex, and moderate inhibition of tactile reflexes. The facilitatory effect of stimulating a number of PAG sites on the latter reflexes was demonstrated. Effects of PAG stimulation fell into two classes: brief, measurable in hundreds of msec and more prolonged, measured in minutes and seconds. Findings would indicate certain differences between the effects of PAG stimulation low-threshold (non-nociceptive) and high-threshold (nociceptive) startle reflexes, of which the possible mechanisms are discussed.A. Some of the factors that affect the intensities and the phases of the first five equatorial x-ray reflections from skeletal muscle are studied by simplified models describing axially projected mass distributions in unit cells. Examples of mass distributions that produce various phase combinations and intensities are presented. Effects due to radial movement of crossbridges and those due to mass transfer between the thick filament and the thin filament regions are compared. In addition, the study suggests that some features in the reconstructed filament structures could be due to the consequences of limited resolution.  相似文献

Seven out of twenty 30–50 kb genome fragments with an MDG1 copia-like element cloned in cosmids were found to carry homologous sequences which belong to a new family of non-mobile heterochromatic moderate repeats (the HMR family). These repeats along with the MDG1 copies inserted in them are under-replicated in polytene chromosomes. Such repeats may also be located in the intercalary heterochromatin site 12E of the X chromosome. Chromosomal heterochromatic regions are enriched with one of the two main genomic variants of MDG1, MDG1het, identifiable by EcoRI restriction. From Southern DNA blot analysis the number of MDG1het copies and their sites within the heterochromatin are invariant in all the stocks examined, while there is not a single MDG1 site along the polytene chromosomes shared by all the stocks in question.  相似文献

The sequence of U1 RNA has been determined in the eggs and embryos of two sea urchins, Lytechinus variegatus and Strongylocentrotus purpuratus. In both species the sequence of the U1 RNA changes as the embryos progress through development. The sequence of the major U1 RNA in the eggs of the two species differs in two nucleotides, while the sequence of the U1 RNA present in the late embryos and somatic tissue is identical in the two species. The U1 RNA in eggs and early embryos is primarily derived from the tandemly repeated gene set, which is not expressed in somatic tissues.  相似文献

Midshaft radial bone mass was first measured from 1970 through 1972 by photon absorptiometry in 42 pairs of monozygotic (MZ) and 38 pairs of dizygotic (DZ) male Caucasian twins (age 44-55 years). The MZ intraclass correlation (rMZ) of .70 was significantly larger (P less than .05) than the DZ correlation (rDZ) of .45, providing evidence for genetic influences (Smith et al. 1973). Radial bone mass measurements repeated 16 years later (1986-87) on 25 of the MZ pairs and on 21 of the DZ pairs revealed an rMZ of .61 and an rDZ of .44, but the difference was not significant (P greater than .05). The twins had an average radial mass loss of 0.49%/year between the two examinations. The rMZ (.52) and rDZ (.49) values for the 16-year loss in radial mass were both significantly different from zero, but their similar size indicated that the correlations were due to nongenetic factors. In a search for the source of genetic influences on adult radial mass, heritability was estimated by the formula 2(rMZ - rDZ) for radial width and was found to be .66 and .76 (P less than .05) for examinations 1 and 2, respectively. An index of radial density (mass/width) was calculated, and the differences between rMZ and rDZ were not significant at either examination. The intraclass correlations (rMZ = .35; rDZ = .43) were both significant for the loss of bone density between examinations but provided no evidence for genetic influences, results similar to the findings for the loss of mass.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The DMD gene, which spans more than 2,000 kbp, has been assigned to band Xp21 of the X chromosome. Two subclones (PERT 87-1 and PERT 87-15) of the intragenic locus DXS164 physically are separated by approximately 60 kbp. Linkage studies were done in 49 informative DMD families by using the LINKAGE program. Crossing-over between the loci studied occurred in four families. A recombination rate of 4% (support interval [Zmax-1] 1%-10%), which was 54 (support interval 14-135-fold) times higher than expected, was found with a maximum lod score of 13.50. These data suggest a hot spot for recombination within DXS164.  相似文献

The properties of natural bovine surfactant and its lipid extract have been examined with a pulsating bubble surfactometer which assesses the ability of surfactant lipids to adsorb to the air/liquid interface and reduce the surface tension to near 0 dynes/cm during dynamic compression. Studies conducted at 1 mg/ml phospholipid revealed that the surface activity (i.e., the ability to produce low surface tensions) of lipid extracts could be enhanced by incubating the sample at 37 degrees C for 120 min or by addition of CaCl2. In contrast, incubation at 37 degrees C only slightly improved the biophysical activity of natural surfactant and the addition of CaCl2 had a more modest effect than with lipid extracts. With 20 mM CaCl2, the surfactant activity of lipid extract surfactant was similar to that of natural surfactant. Incubation with EDTA reduced the biophysical activity of natural surfactant. Experiments in which increasing amounts of lipid extract were replaced by natural surfactant revealed that small amounts of natural surfactant enhanced the surfactant activity of lipid extract. The biophysical activity of lipid extract surfactant was also increased by the addition of soluble surfactant-associated protein-A (SP-A) (28-36 kDa) purified from natural bovine surfactant. These results indicate that SP-A (28-36 kDa) improves the surfactant activity of lipid extracts by enhancing the rate of adsorption and/or spreading of phospholipid at the air/liquid interface resulting in the formation of a stable lipid monolayer at lower bulk concentrations of either phospholipid or calcium.  相似文献
