Out of 36 strains of fungi of the genus Penicillium, some of which were isolated from ancient permafrost, 14 strains synthesized mycophenolic acid (MPA). Maximal (over 500 mg/l) accumulation of MPA in culture liquid was observed in P. brevicompactum strains (VKM F-457, VKM F-477, and VKM F-1150). This was the first study to detect MPA in representatives of the species P. rugulosum; in three strains of this species (VKM FW-665, VKM FW-717, and VKM FW-733), the level of MPA accumulation exceeded 300 mg/l. The time course of the synthesis of MPA by P. rugulosum strain VKM FW-733 was studied. Physiological studies have shown that the epidermal head and arm lines in cephalopods are a mechanoreceptive system that is similar to the fish and amphibian lateral lines (Budelmann BU, Bleckmann H. 1988. A lateral line analogue in cephalopods: Water waves generate microphonic potentials in the epidermal head lines of Sepia officinalis and Lolliguncula brevis. J. Comp. Physiol. A 164:1-5.); however, the biological significance of the epidermal lines remains unclear. To test whether cuttlefish show behavioural responses to local water movements, juvenile Sepia officinalis were exposed to local sinusoidal water movements of different frequencies (0.01-1000 Hz) produced by a vibrating sphere. Five behavioural responses were recorded: body pattern changing, moving, burrowing, orienting, and swimming. Cuttlefish responded to a wide range of frequencies (20-600 Hz), but not to all of the frequencies tested within that range. No habituation to repeated stimuli was seen. Results indicate that cuttlefish can detect local water movements (most likely with the epidermal head and arm lines) and are able to integrate that information into behavioural responses.  相似文献

Over a period of five years (2000–2004), wheat and rye samples of integrated and ecological cultivation in the land Brandenburg were analyzed by HPLC for theFusarium toxins deoxynivalenol and zearalenone. In the years 2000, 2001, 2003 and 2004 the contamination generally was lower than in the year 2002. In the five years of testing, the contamination byFusarium toxins was significantly less frequent and lower in cereals of ecological cultivation than in cereals of integrated cultivation. The effect of denervation on the contractile activity of the saphenous artery in normotensive rats and rats with regional hypotension was studied. Hypotension was caused by partial occlusion of the abdominal aorta distally from the renal arteries, and then, in four weeks, to denervate the saphenous artery, a portion of the femoral nerve in one of the limbs was resected. In two more weeks, the contractile responses of ring preparations of the saphenous artery (after removal of the endothelium and block of neuronal uptake and β-adrenoreceptors) were investigated under isometric conditions. In normotensive rats, the denervation led to an increase in the vascular sensitivity to norepinephrine, phenylephrine, serotonin, and KCl. Similar changes in contraction were caused by chronic hypotension; however, rats with hypotension exhibited no additional denervation-induced increase in the vascular sensitivity. After treatment with glyoxylic acid, the fluorescence intensity of the vascular adrenergic fibers adapted to a reduced pressure was lower than that in the norm. It was assumed that the vascular hypersensitivity in hypotension is caused by impairment of sympathetic innervation.  相似文献

Drosophila melanogaster Heterochromatin Protein 1a (HP1a) is an essential protein critical for heterochromatin assembly and regulation. Its chromo shadow domain (CSD) homodimerizes, a requirement for binding protein partners that contain a PXVXL motif. How does HP1a select among its many different PXVXL-containing partners? HP1a binds tightly to Heterochromatin Protein 2 (HP2), but weakly to PIWI. We investigated differences in homodimerization and the impact of the C-terminal extension (CTE) by contrasting HP1a to its paralogue, HP1b. HP1a and HP1b differ in the dimerization interface, with HP1a having an Arg at position 188 rather than Glu. We find that while this substitution reduces the dimerization constant, it does not impact the binding surface as demonstrated by unchanged partner binding affinities. However, the CTE (only 4 residues in HP1a as compared with 87 residues in HP1b) is critical; the charged residues in HP1a are necessary for tight peptide binding. Examining a panel of amino acid substitutions in the HP1a CSD, we find that Leu-165 in HP1a interacts with HP2 but not PIWI, supporting the conclusion that different sites in the binding surface provide discrimination for partner selection. Partner sequence is also critical for affinity, as the remaining difference in binding between HP2 and PIWI polypeptides is eliminated by swapping the PXVXL motifs between the two. Taken together, these studies indicate that the binding surface of the HP1a CSD plus its short CTE provide the needed discrimination among HP1a''s partners, and that the CTE is important for differentiating the interactions of the Drosophila HP1 paralogs.  相似文献
