For a long time, targeted and discovery proteomics covered different corners of the detection spectrum, with targeted proteomics focused on small target sets. This changed with the recent advances in highly multiplexed analysis. While discovery proteomics still pushes higher numbers of identified and quantified proteins, the advances in targeted proteomics rose to cover large parts of less complex proteomes or proteomes with low protein detection counts due to dynamic range restrictions, like the blood proteome. These new developments will impact, especially on the field of biomarker discovery and the possibility of using targeted proteomics for diagnostic purposes. 相似文献

Mansonic schistosomiasis is a neglected disease transmitted by Biomphalaria spp. snails. Understanding what happens inside the intermediate host is important to develop more efficient ways of reducing schistosomiasis prevalence. Our purpose was to characterize metabolic and immunological changes in Biomphalaria glabrata 24 h after exposure to Schistosoma mansoni. For this purpose, proteins were extracted from snails’ whole tissue with Tris-Urea buffer and digested with tripsin. Mass spectrometry was performed and analyzed with MaxQuant and Perseus software. Also, the hemolymph of five snails 24 h post exposure was collected, and the numbers of hemocytes, levels of urea, uric acid, nitric oxide, calcium, glycogen and alanine and aspartate aminotransferases activities were assessed. Snails were also dissected for measurement of glycogen content in the cephalopodal region and gonoda-digestive gland complex. Globin domain proteins were found to be up-regulated; also the number of circulating hemocytes was significantly higher after 24 h of exposure to the parasite. NO levels were higher 24 h post exposure. Several proteins associated with energy metabolism were found to be up-regulated. Glycogen analysis showed a significant decrease in the gonad-digestive gland complex glycogen content. We found several proteins which seem to be associated with the host immune response, most of which were up-regulated, however some were down-regulated, which may represent an important clue in understanding B. glabrata – S. mansoni compatibility. 相似文献

We collected 729 Hypanus guttatus from the northern coast of the state of Rio Grande do Norte (RN), of which 196 were used to estimate age and growth. Ninety-five were male (12.7 to 57.0 cm disc width; WD) and 101 were female (13.0 to 88.5 cm WD); females were significantly larger than males. Cross sections of vertebrae showed band-pairs ranging from 0 to > 14 in females and from 0 to 9 in males. New-borns presented an opaque edge at birth in vertebrae without a birthmark. The average percentage of error (APE; %E) for the entire sample provided evidence that ages were repeatable. The mean monthly marginal increment (IM) indicates annual band-pair formation from August to November. The annual cycle model for one band-pair deposition provided the best fit to data based on the AIC, with peaks between August and October, similar to that found in the IM analysis, suggesting an annual formation pattern. A multi-model approach that included four models based on the observed mean WD at age indicated a modified von Bertalanffy growth model as the best for describing the species growth: W0 (WD at birth) = 14.6 cm for both sexes; females W∞ = 98.61 cm (95% CI = 87.34–114.61 cm); k = 0.112 year−1 (CI = 0.086–0.148 year−1); males W∞ = 60.22 cm (CI = 55.66–65.35 cm); k = 0.219 year−1 (CI = 0.185–0.276 year−1). The age-at-maturity in males and females is 5 years and 7 years, respectively. The age composition shows that most (84%) specimens were aged 0 to 2 years. The information provided here is essential for analytical assessments of H. guttatus, which is subject to significant fishing pressure mainly on new-borns and juveniles. 相似文献

This study evaluated the functional role of the highly generalist omnivore Melichthys niger in the remote St. Peter and St Paul's Archipelago (SPSPA), Brazil, where grazing herbivorous fishes are very scarce. We analysed patterns of distribution from zero to 30 m deep during three time intervals during the day and sampled different aspects of their feeding behaviour, including diel feeding rate, feeding substrate and diet. The density of M. niger increased with depth (26–30 m) and decreased by the end of the day. Although M. niger did not present a typical herbivore diel feeding pattern, they targeted the epilithic algal matrix as their primary feeding substrate, ingesting predominantly algae and detritus. The characteristic Caulerpa racemosa var. peltata from SPSPA was ingested only as detached fronds. We suggest that in the isolated SPSPA, the single species M. niger may perform a unique role as a link between benthic primary production and higher levels. Further studies on the trophic ecology of omnivorous species are necessary to better understand their roles in a reef system, especially in impoverished areas where they are likely to play a crucial role. 相似文献

Mycodrosophila is a cosmopolitan genus of Drosophilidae that comprises approximately 130 species with mycophagous habitats. In this study, we described a new species of Mycodrosophila based on morphological traits and included details of the male terminalia. The holotype is from Eugênio Lefévre, locality in Campos do Jordão municipality, SP, Brazil, located in the Atlantic rainforest biome and was sampled in the 1930s. 相似文献

New records and distributional notes of Chironomidae (Insecta, Diptera) are provided for four protected areas in the state of Pernambuco, northeastern Brazil. Additionally, we also present new records and update of distributional ranges from Brazil and the Neotropical Region. In total, 810 specimens belonging to 35 genera within the subfamilies Chironominae (22 taxa), Tanypodinae (11 taxa) and Orthocladiinae (2 taxa) were found. The subfamilies Chironominae and Tanypodinae predominated. Axarus Roback, 1980 and the Tanytarsus ortoni-group were recorded for the first time in the state of Pernambuco, while Nanocladius Kieffer, 1913a was recorded for the first time in the Northeast Region of Brazil.
