We validated a stochastic model of the evolution of resistance to adulticidal sprays of methyl-parathion in western corn rootworm, Diabrotica virgifera virgifera LeConte, populations in Nebraska. The population dynamics predicted by the model resembled that reported for field populations, and time until control failures occurred closely matched reports by commercial crop consultants. We incorporated uncertainty about the values used for 18 model parameters by replacing default values with random draws taken from a normal distribution. One parameter, the initial resistance allele frequency, was no longer measurable because of the evolution of resistance. We therefore proposed five candidate initial allele frequencies and developed probability distributions for the time to resistance for each by running 1000 simulations with parameters randomly varied. These distributions included variation because of stochastic effects as well as parameter uncertainty. We used Bayesian inference to estimate the candidate frequency most likely, given reported times to field control failures. The initial allele frequency of 10(-4) was most likely (29%), 10(-3) was less likely (28%), whereas 10(-6) was relatively unlikely (5%). Results from sensitivity analysis depended upon how evolution of resistance was measured. When resistance was examined as a genetic phenomenon, the rate of increase of the resistance allele depended almost entirely on genetic factors (LC50 values), the characteristics of the pesticide (residual activity), and the variance associated with emergence of adults. When resistance was measured as failure of methyl-parathion to reduce populations below threshold levels (0.5 gravid females per plant), parameters that contributed to population growth rate (mortality and fecundity) were also important. These data suggest two important phases in resistance evolution in corn rootworms: a genetic phase associated with negative growth rates and rapid changes in resistance allele frequencies and a rebound phase associated with positive growth rates and near fixation of the resistance allele.  相似文献

Here we report the annotated genome sequence of Moraxella catarrhalis strain RH4, a seroresistant-lineage strain isolated from the blood of an infected patient. This genome sequence will allow us to gain further insight into the genetic diversity of clinical M. catarrhalis isolates and will facilitate study of M. catarrhalis pathogenesis.  相似文献

For computational studies of protein folding, proteins with both helical and β‐sheet secondary structure elements are very challenging, as they expose subtle biases of the physical models. Here, we present reproducible folding of a 92 residue α/β protein (residues 3–94 of Top7, PDB ID: 1QYS) in computer simulations starting from random initial conformations using a transferable physical model which has been previously shown to describe the folding and thermodynamic properties of about 20 other smaller proteins of different folds. Top7 is a de novo designed protein with two α‐helices and a five stranded β‐sheet. Experimentally, it is known to be unusually stable for its size, and its folding transition distinctly deviates from the two‐state behavior commonly seen in natural single domain proteins. In our all‐atom implicit solvent parallel tempering Monte Carlo simulations, Top7 shows a rapid transition to a group of states with high native‐like secondary structure, and a much slower subsequent transition to the native state with a root mean square deviation of about 3.5 Å from the experimentally determined structure. Consistent with experiments, we find Top7 to be thermally extremely stable, although the simulations also find a large number of very stable non‐native states with high native‐like secondary structure. The origin-binding domain (OBD) of simian virus 40 (SV40) large T-antigen (T-Ag) is essential for many of T-Ag's interactions with DNA. Nevertheless, many important issues related to DNA binding, for example, how single-stranded DNA (ssDNA) transits along the T-Ag OBD, have yet to be established. Therefore, X-ray crystallography was used to determine the costructure of the T-Ag OBD bound to DNA substrates such as the single-stranded region of a forked oligonucleotide. A second structure of the T-Ag OBD crystallized in the presence of poly(dT)(12) is also reported. To test the conclusions derived from these structures, residues identified as being involved in binding to ssDNA by crystallography or by an earlier nuclear magnetic resonance study were mutated, and their binding to DNA was characterized via fluorescence anisotropy. In addition, these mutations were introduced into full-length T-Ag, and these mutants were tested for their ability to support replication. When considered in terms of additional homology-based sequence alignments, our studies refine our understanding of how the T-Ag OBDs encoded by the polyomavirus family interact with ssDNA, a critical step during the initiation of DNA replication.  相似文献

The surface profile of human skin influences characteristically the distribution pattern of topically applied formulations and consequently the efficacy of sunscreens. The volumes of furrows and the spectroscopically determined factors of inhomogeneity are investigated for three sunscreens. A clear correlation between both measurands exists. The average values for younger (<32 years) and older (≥51 years) volunteers do not show statistically significant differences. Systematic variations found for the individual values are due to a reduced elasticity of the skin with age improving the homogeneity of the sunscreen distribution. (© 2012 WILEY‐VCH Verlag GmbH & Co. The ecological integrity of Sagebrush ( Artemisia spp.) ecosystems in the Intermountain West (U.S.A.) has been diminished by synergistic relationships among human activities, spread of invasive plants, and altered disturbance regimes. An aggressive effort to restore Sagebrush habitats is necessary if we are to stabilize or improve current habitat trajectories and reverse declining population trends of dependent wildlife. Existing economic resources, technical impediments, and logistic difficulties limit our efforts to a fraction of the extensive area undergoing fragmentation, degradation, and loss. We prioritized landscapes for restoring Sagebrush habitats within the intermountain western region of the United States using geographic information system (GIS) modeling techniques to identify areas meeting a set of conditions based on (1) optimum abiotic and biotic conditions favorable for revegetation of Sagebrush; (2) potential to increase connectivity of Sagebrush habitats in the landscape to benefit wildlife; (3) location of population strongholds for Greater Sage-Grouse ( Centrocercus urophasianus , a species of conservation concern); and (4) potential impediments to successful restoration created by Cheatgrass ( Bromus tectorum , an invasive exotic annual grass). Approximately 5.8 million ha in southwestern Idaho, northern Nevada, and eastern Oregon met our criteria for restoring Wyoming big sagebrush ( Artemisia tridentata ssp. wyomingensis ) and 5.1 million ha had high priority for restoring Mountain big sagebrush ( A. tridentata ssp. vaseyana ). Our results represent an integral component in a hierarchical framework after which site-specific locations for treatments can be focused within high-priority areas. Using this approach, long-term restoration strategies can be implemented that combine local-scale treatments and objectives with large-scale ecological processes and priorities.  相似文献

Background  

Evolutionary theory suggests that the selection pressure on parasites to maximize their transmission determines their optimal host exploitation strategies and thus their virulence. Establishing the adaptive basis to parasite life history traits has important consequences for predicting parasite responses to public health interventions. In this study we examine the extent to which malaria parasites conform to the predicted adaptive trade-off between transmission and virulence, as defined by mortality. The majority of natural infections, however, result in sub-lethal virulent effects (e.g. anaemia) and are often composed of many strains. Both sub-lethal effects and pathogen population structure have been theoretically shown to have important consequences for virulence evolution. Thus, we additionally examine the relationship between anaemia and transmission in single and mixed clone infections.  相似文献

Two field experiments were conducted in 1995-1996 to determine if there are common yield responses among maize hybrids to larval western corn rootworm, Diabrotica virgifera virgifera LeConte injury. Three yellow dent hybrids, five white food grade dent hybrids, and a popcorn hybrid were included in the study. The minimum level of rootworm injury as measured by root damage ratings (3.2-4.2) that significantly reduced yield was similar across the hybrids included in the study. However, the pattern of yield response to different rootworm injury levels varied among hybrids. This suggests that maize hybrids may inherently differ in their ability to tolerate rootworm injury and partition biomass in response to injury and other stresses. The complex interaction among hybrid, level of injury, and other stresses suggests that a common western corn rootworm injury-yield relationship may not exist within maize.  相似文献
